Project description:Brain aromatase, an enzyme responsible for the local synthesis of estrogens, plays a key role in regulating behavior and neuroplasticity in mammals. In teleost fish, brain aromatase is encoded by the cyp19a1b gene, which is strongly expressed in radial glial cells but the specific functions of this enzyme are currently unknown. To investigate its role, a cyp19a1b-mutant zebrafish line was generated using gene-editing techniques. Behavioral, neurogenic, and neurotransmission-related parameters were assessed in adult male and female zebrafish. Behavioral analysis highlighted significant alterations in mutant zebrafish including changes in swimming activity, boldness, sociability, and aggression with a stronger effect in males compared to females. Beyond these behavioral modifications, mutant zebrafish exhibited disrupted cell proliferation patterns, as assessed by PCNA immunofluorescence in key forebrain regions. Specifically, proliferation decreased in the telencephalon and in the caudal hypothalamus of mutant zebrafish while increasing in the olfactory bulbs. The number of dopaminergic and serotonergic neurons, visualized by immunofluorescence, remained unchanged. Similarly, HPLC-ED quantification of monoamines and their metabolites showed no significant differences between mutant and wild-type zebrafish. To further explore the impact of cyp19a1b mutation on gene expression, transcriptomic analysis was performed using BRBSeq technology. Gene expression analyses identified several processes affected by the mutation, including cell proliferation, apoptosis, estrogen signaling, neuroplasticity, and behavioral regulation, in a sex- and region-dependent manner. In conclusion, our results demonstrate that several behaviors, including locomotor activity, sociability, aggressiveness, and anxiety, exhibit marked sexual dimorphism. They show that cyp19a1b mutation affects locomotor activity in a context-dependent manner, increases boldness, and reduces aggressiveness. In addition, transcriptomic analyses revealed widespread dysregulation of gene expression, which likely contributes to the observed behavioral alterations. Taken together, these findings underscore the crucial role of brain aromatase in neurobiological regulation of diverse behaviors.

Project description:Cytochrome P450 aromatase is an essential enzyme for vertebrates. It is responsible for the conversion of androgens to estrogens in the brain and gonadal tissues. Aromatase converts testosterone into estradiol and therefore participates in the regulation of many processes which are controlled by estrogens such as development and fertility. Teleosts have been characterized as having exceptionally high levels of brain estrogen biosynthesis when compared to the brains of the other vertebrates. Little is known about the effects of estrogens on brain function in teleosts. The main objective of this study was to assess the effects of fadrozole, a powerful aromatase inhibitor, on gene expression in the zebrafish brain. To achieve this, male zebrafish were exposed to fadrozole for 10 days. This exposure causes a decrease in estrogens along with an increase in androgens. Plasma testosterone levels showed a 3-fold increase in response to the exposure. In the telencephalon, 235 genes were identified by Affymetrix GeneChip analysis as being differentially regulated. Real-time RT-PCR was used to validate the data obtained from the microarrays. This technique was also used to evaluate the response in the hypothalamus, which appears to follow similar transcriptional regulation. Gene ontology analysis of the microarray data revealed common biological processes and molecular functions such as sensory perception and detection of light stimulus, homeobox and transcription factor complexes. This study has identified genes which are directly and/or indirectly controlled by estrogens and androgens which helped to demonstrate the pronounced effects of endocrine disrupting chemicals on gene expression in the brain of teleosts. Collectively, the results provide a better understanding of the effects of aromatase inhibitors on gene expression and also shed light on the underlying effects of sex hormone variation and their importance in the brain of teleosts.

Project description:Cortical atrophy and dysfunction are hallmarks of many neuropsychiatric diseases including depression, post-traumatic stress disorder (PTSD), and addiction. Psychoplastogens are small molecules with the potential to rapidly rescue these structural and functional deficits, leading to sustained therapeutic effects after a single administration. Classic psychedelics are among the most potent and efficacious psychoplastogens known, with recent data indicating that their clinical and preclinical effects might be more robust and durable than those of ketamine. Moreover, the advent of non-hallucinogenic psychoplastogens structurally related to psychedelics, such as tabernanthalog (TBG), has opened new opportunities for developing scalable alternatives capable of promoting structural and functional neuroplasticity in the prefrontal cortex (PFC). Currently, it is unclear what roles 5-HT2A receptors (5-HT2ARs) and immediate early gene activation play in the neuroplasticity-promoting effects of non-hallucinogenic psychoplastogens like TBG—information necessary to engineer optimized neurotherapeutics related to psychedelics. Here, we use a combination of pharmacological and genetic tools to demonstrate that several non-hallucinogenic psychoplastogens promote cortical neuron growth through the same biochemical pathway as classic psychedelics. Furthermore, we demonstrate that there is a causal relationship between TBG-induced cortical spinogenesis and its sustained antidepressant-like effects. In contrast to psychedelics, TBG promotes cortical neuron growth in the absence of a glutamate burst or immediate early gene (IEG) activation, effects assumed to be necessary for psychedelic-induced neuroplasticity. Our results elucidate how the hallucinogenic effects of psychedelics can be decoupled from their beneficial effects on cortical neuron growth.
