Project description:Peripheral blood DNA methylation was profiled in 46 patients with treatment-resistant depression prior to treatment with transcranial magnetic stimulation (TMS). Genome-wide DNA methylation was measured using the Illumina Infinium MethylationEPIC BeadChip (850K array). Following quality control, data from 46 subjects were included in downstream analyses. Treatment outcomes were assessed using the PHQ-9 depression scale and included treatment response (≥50% reduction in PHQ-9), symptom trajectory (ΔPHQ-9), and remission (PHQ-9 score < 5). Methylation data were analyzed to identify differentially methylated regions (DMRs) associated with treatment outcomes. Both raw IDAT files and processed beta-value data are provided.

Project description:Transcranial Magnetic Stimulation (TMS) is a noninvasive technique that uses pulsed magnetic fields to affect the physiology of the brain and central nervous system. Repetitive TMS (rTMS) has been used in the study and treatment of neurological conditions including major depression, stroke, epilepsy, schizophrenia, multiple sclerosis, Parkinson’s, and Alzheimer’s disease, as well as other, non-neurological disorders. Among the effects of rTMS, low-frequency protocols (≤ 1Hz) are generally understood to result in cortical inhibition, whereas high-frequency protocols (≥ 3Hz) increase cortical excitability. However, the complex molecular basis of rTMS is largely unexplored. Using three complementary rat models, in vitro, ex vivo, and in vivo, we show complex patterns of hippocampal and neocortical transcriptional response to stimulation in glutamatergic and GABAergic signaling pathways, as well as multiple inflammatory pathways, among others. This broad-based molecular survey helps provide a foundation to tease out the complex molecular mechanisms of the effects of rTMS.

Project description:Repetitive transcranial magnetic stimulation (rTMS) is a widely adopted non-invasive tool used to study the brain and as an alternative treatment for neurological disorders. However, the molecular mechanisms affected in the brain regions at and beyond the stimulation site, and how this varies with different rTMS protocols in an aged brain remains unclear. Here we use a combination of bulk RNA-sequencing and spatial transcriptomics to map the molecular changes across a mouse brain in response to two commonly used rTMS protocols: intermittent (iTBS) and continuous theta burst stimulation (cTBS).
