Project description:Mitochondria are the metabolic center and powerhouses of cells by coordinating the tricarboxylic acid (TCA) cycle, oxidative phosphorylation (OXPHOS) and electron transport to generate ATP, which provide energy for cell survival and functional maintenance. This is attributed to the key structure of mitochondria cristae, which are formed by the inward folding of inner mitochondrial membranes. Numerous studies have demonstrated that RNA-binding proteins play an important role in maintaining the normal structure and function of mitochondria. However, the relevant mechanism remains unclear. Here, we report that the RNA-binding protein LARP-1 affects the structure and function of mitochondria by disrupting crista-related proteins, including the MICOS complex, respiratory chain complexes, and the proton-transporting ATP synthase complex. Using APEX-based proximity labeling assays, we pulled down mRNAs associated with LARP-1::APEX2, and determined the identities of mRNAs by sequencing. Among the 653 LARP-1::APEX2-enriched mRNAs, 37.5% (245/653) encode mitochondrial proteins, of which 71.8% (176/245) were identified to be down-regulated in larp-1 mutants by LC-MS analysis of purified mitochondria. In addition, mRNAs encoding components of the MICOS complex, respiratory chain complexes, and ATP synthase complex were strongly enriched, suggesting that they were associated with LARP-1.

Project description:Mitochondria has its own mRNA translation system, which is mediated by mitochondria-specific protein synthesis machinery – ‘mitoribosomes’. Dysregulation of mitochondrial translation disrupts the metabolic homeostasis and has been implicated in pathologies including metabolic syndromes, neurological disorders and cardiac diseases. Here, we identify that EOLA1 (Endothelium and lymphocyte associated ASCH domain 1) as a novel mitochondrial protein and exclusively localizes to mitochondria matrix. We further show that knockout of EOLA1 dramatically affects mitochondrial translation and subsequently the oxidative phosphorylation in mammalian cells. Mechanistically, EOLA1 interacts with mt-rRNA (mitochondrial ribosomal RNA) and EFTU, which maintains the translation elongation of mitochondrial mRNAs. Importantly, we generated a Eola1 knocked-out mice model and found that heart functions are obviously abnormal upon depletion of EOLA1 homolog in mouse, implying that EOLA1 are important for proper functions of heart via regulating mitochondrial translation. Taken together, our work demonstrates that EOLA1 is a novel regulator for mt-RNA and mitochondrial translation.

Project description:CRISTA (SMIM26) is a mitochondrial microprotein important for assembly of complex I. Here we used differential complexome profiling to study the composition of ETC complexes in SMIM26 WT and KO HEK293T mitochondria. CRISTA potentiates mitochondrial serine import via direct interactions with the serine transporter SFXN1 in the inner mitochondrial membrane. Its C-terminus furthermore interacts with the mitochondrial ribosome to form a SFXN1-CRISTA-mitoribosome triad that is enriched for mt-ND5 mRNA encoding a key enzymatic subunit of Complex I. Deletion of CRISTA reduces mitochondrial serine uptake, leading to reduced levels of folate intermediates. This in turn impairs 5-taurinomethyl(thio)uridinylation of mitochondrial tRNAs (tm5U and tm5S2U), leading to a specific loss of ND5 translation and a stall in Complex I biogenesis.

Project description:Current evidence suggests that nuclear-encoded mitochondrial proteins can be locally translated at the mitochondrial surface and co-translationally or post-translationally imported into mitochondria. mRNA localization on the mitochondrial membrane, a prerequisite for localized translation, remains uncharacterized in higher eukaryotic organisms. We employed fractionation-sequencing to profile mitochondria-associated mRNAs in zebrafish larvae. Our transcriptome-wide analysis reveals the localization of mRNAs of only 12% of the nuclear-encoded mitochondrial proteins to the mitochondrial surface, which suggests that post-translational import is the dominant mode of protein import to mitochondria. Additionally, the mRNAs which were localized to the mitochondrial membrane consisted mostly of those encoding proteins involved in mitochondrial dynamics, suggesting their site-specific translation. Finally, we show that the loss of function of the MIA pathway responsible for the post-translational import of a subclass of mitochondrial proteins, triggers mitochondrial localization of mRNAs encoding proteins that are imported to mitochondria via other pathways. Thus, our study suggests that mRNA targeting and localized translation could be relevant in higher eukaryotes to combat stress conditions affecting mitochondrial biogenesis in general.

Project description:Recent studies have demonstrated that mitochondria can be transferred between different cell types to control metabolic homeostasis. However, whether the mitochondria transfer network occurred in the skeletal system or regulate skeletal metabolic homeostasis in vivo is not fully elucidated. Herein, we found that osteolineage cells transfer mitochondria to CD11b+ myeloid, B220+ lymphoid and hematopoietic stem and progenitor cells (HSPCs), of which monocytes/macrophages received the most transferred mitochondria. This process was inhibited by GC treatment contributing to the progression of glucocorticoid-induced osteoporosis (GIOP). Further analysis demonstrated that osteolineage cells transfer mitochondria to osteoclastic lineage cells via Miro1 mediated direct contact, and altered the glutathione metabolism, leading to the ferroptosis of osteoclastic lineage cells, thus inhibiting osteoclast activities. These findings revealed an unappreciated mechanism of mitochondrial regulation of skeletal metabolic homeostasis via mitochondria transfer and provide new insights of the mechanism of GIOP progression.

Project description:Nearly all mitochondrial proteins are nuclear-encoded and are targeted to their mitochondrial destination from the cytosol. Here, we used proximity-specific ribosome profiling to comprehensively measure translation at the mitochondrial surface in yeast. The majority of inner membrane proteins were co-translationally targeted to mitochondria, reminiscent of proteins entering the endoplasmic reticulum (ER). Comparison between mitochondrial and ER localization demonstrated that the vast majority of proteins were targeted to a specific organelle. A prominent exception was the fumarate reductase Osm1, known to reside in mitochondria. We identified a conserved ER isoform of Osm1, which contributes to the oxidative protein folding capacity of the organelle. This dual localization was enabled by alternative translation initiation sites encoding distinct targeting signals. These findings highlight the exquisite in vivo specificity of organellar targeting mechanisms.
