Project description:Abstract: Controlled release of immunomodulatory cytokines like interleukin-4 (IL-4) from biomaterials can modulate host immune cell behavior to improve tissue integration, however options for modifying complex biomaterials remain limited. Bioconjugation strategies using biotin-avidin interactions offer a promising approach since biotin can be conjugated to proteins, biomaterials, and even cells, without compromising their function. Although it is known that conjugation of biotin to large biomolecules reduces its binding affinity for avidin and avidin variants, the potential to control release of biotinylated molecules via leveraging affinity interactions has not been thoroughly investigated. Moreover, the effects of biotin and avidin variants on innate immunity are poorly understood. Therefore, the goal of this study was to determine if biotin-avidin interactions could be manipulated to control the release of IL-4 from biomaterials and to investigate the subsequent effects on primary human macrophage phenotype. First, we characterized the effects of soluble biotin, avidin, and avidin variants, streptavidin and CaptAvidin, on the phenotype of primary human macrophages using RNA sequencing and found that CaptAvidin significantly influenced macrophage gene expression while other variants did not. Then, after evaluating how bioconjugation parameters influenced biotin density and avidin variant binding to porous gelatin scaffolds, we found that biotin-avidin affinity interactions influenced the spontaneous release of biotinylated IL-4 (bIL-4) from biotinylated and desthiobiotinylated scaffolds bound with either avidin or streptavidin for up to ten days in vitro. Finally, we measured the response of primary human macrophages (8 donors) cultured on modified scaffolds, finding an increase in IL-4 driven genes when bIL-4 was released via biotin-streptavidin affinity interactions compared to desorption-based release. These results highlight how biotin-avidin interactions can be leveraged for spontaneous release to achieve an immunomodulatory drug delivery system.

Project description:We engineering a G4-targeted biotin ligase probe (G4PID), which consists of three parts, RHAU23 at the C-terminus, miniTurbo at the N-terminus, and a flexible linker in the middle to prevent the disturbance of the two functional moieties. We used eCLIP to verified the G4s affinity of G4PID. We also used eCLIP method to verify the direct G4 interaction of Splicing factor 3B subunit 4 (SF3B4) in cellular transcriptome.

Project description:The purpose of the study is to assess a new treatment for patients with liver tumor metastases from colorectal cancer. The treatment has never been used in humans before. The treatment foresees the use of two compounds: AvdinOX and [177Lu]DOTA-biotin.
AvidinOX is a new compound, essentially a natural protein obtained from hen eggs, while [177Lu]DOTA-biotin is a new chemical compound resulting from the combination of the DOTA-biotin (also deriving from a natural vitamin which is biotin) with the 177Lutetium, an atom which emits radiation.
AvidinOX will be injected directly into the metastases in the liver and [177Lu]DOTA-biotin will be injected into the arm vein.
One specific property of AvidinOX is that it chemically links to the tumor tissues when it is injected while maintaining the capacity to take up [177Lu]DOTA-biotin. Once locally bound in tumor tissue, AvidinOX becomes an "artificial receptor" for intravenously injected [177Lu]DOTA-biotin, which allows an internal radiation therapy of the tumor tissue.
The treatment of liver metastases with local injection of AvidinOX and the following intra-venous injection of [177Lu]DOTA-biotin could be simpler and more tolerable than the current available treatments.

Project description:In addition to its essential metabolic functions biotin is suggested a critical role in regulating gene expression. The first committed enzyme in biotin biosynthesis in Arabidopsis, 7-keto-8-aminopelargonic acid synthase is encoded by At5g04620 (BIO4). We isolated a novel T-DNA insertion mutant of BIO4 (bio4-1) showing a spontaneous cell death phenotype that could be rescued both by exogenous biotin and genetic complementation. The bio4-1 plants exhibited massive accumulation of hydrogen peroxide. The T-DNA insertion in the bio4-1 mutant contains a Methionine Sulfoxide Reductase B9 (AT4G21850) under a constitutive 35S CaMV promoter. To dissect the specific effects of biotin defiency on gene expression on a genome wide scale, we compared expression of wt Col-0 leaves with bio4-1 mutants and bio4-1 mutants complemented with D-biotin.

Project description:Rotator cuff tears are the most common conditions in sports medicine and attract increasing attention. Scar tissue healing at the tendon-bone interface results in a high rate of retears, making it a major challenge to enhance the healing of the rotator cuff tendon-bone interface. Biomaterials currently employed for tendon-bone healing in rotator cuff tears still exhibit limited efficacy. 3D printing, a promising technology, enables the customization of scaffold shapes and properties. Bone marrow mesenchymal stem cells (BMSCs) have multi-differentiation potential and valuable immunomodulatory effects. Basic fibroblast growth factor (bFGF), known for its role in proliferation, has been reported to promote osteogenesis. These properties make them applicable in tissue engineering. In this study, we developed a 3D-printed PCL scaffold loaded with bFGF and BMSCs (PCLMF) to restore the tendon-bone interface and regulate the local inflammatory microenvironment. The PCLMF scaffolds significantly improved the biomechanical strength, histological score, and local bone mineral density at regenerated entheses at 2 weeks post-surgery and achieved optimal performance at 8 weeks. Furthermore, PCLMF scaffolds facilitated BMSC osteogenic differentiation and suppressed adipogenic differentiation both in vivo and in vitro. In addition, RNA-seq showed that PCLMF scaffolds could regulate macrophage polarization and inflammation through the MAPK pathway. The implanted scaffold demonstrated excellent biocompatibility and biosafety. Therefore, this study proposes a promising and practical strategy for enhancing tendon-bone healing in rotator cuff tears.

Project description:Microarray analysis was used to evaluate expression differences from a single donor human bone marrow stromal cells (hBMSCs) as a function of varied polymer-based tissue engineering scaffolds. These scaffolds include polycaprolactone (PCL) nanofibers (PCL_NF), films (PCL_SC), poly D,L-lactic acid (PDLLA) nanofibers (PDLLA_NF), films (PDLLA_SC), tissue culture polystyrene (TCPS) and TCPS with osteogenic supplements (TCPS_OS). The results revealed that scaffold structure was able to significantly affect gene expression, with nanofiber scaffolds inducing similar gene expression patterns to hBMCSs cultured with osteogenic media. A library of scaffolds prepared from polycaprolactone or poly D,L-lactic acid was sythesized and cultured with hBMSCs for 14 days with RNA extracted from cells on Day 1 and Day 14.
