Project description:We profiled a time-course single-cell RNA-seq on C57BL/6 mouse ovary from E11.5 to E14.5 to study the cell population heterogeneity and regulation of meiosis initiation. We collected single cell from XX gonads at E11.5, E12.5, E13.5 and E14.5, and were further captured with 10x Genomics based single-cell system. By using single-cell RNA seq, we recapitulated the progression of meiosis using pseudotime ordering of all germ cells and delineated key molecular network among all germ cell clusters. Besides, we provide candidate meiosis stage-specific master regulons along the progression of meiosis and provide detailed transcriptome profile information for controversial Xist+ PGCs. Our data here offers the opportunity for exploring mechanisms underlying germ cell meiosis progression at single cell resolution.

Project description:To explore the molecular signatures of tracheal cells during tracheal development, we performed single cell RNA-seq of developing whole tracheas. We identified 18 cell clusters that constitue trachea tissue. Using sub-clustering analysis of chondrocyte progenitors at E12.5, we found Wnt-responsive chondrocytes that higly express wnt signaling target genes such as Lef1, Notum, Axin2.

Comprehensive single cell RNA-seq datasets of mice trachea epithelial progenitors in 6 time points from E12.5 to E18.5 and mature epithelial cells in 2-month mice

Project description:We delineate the comprehensive developmental trajectories of trachea epithelial progenitors in mice using scRNA-seq datasets. After obtaining the scRNA-seq data in 6 time points from E12.5 to E18.5, we merged all the datasets to describe the establishment process of basal cell population during development, which works as airway facultative stem cells. We found that Krt17 is a novel intermediate marker for basal cell progenitors and Id genes are key transcriptional factors for balancing the proliferation and differentiation programs by promoting their cell cycle.

Project description:Embryonic day (E)12.5 whole murine embryos, E11.5 - E14.5 whole murine embryos, E11.5 - E14.5, post-natal day (P)3 and P35 murine forelimbs, E14.5 brains, and COL1A2-mutant and COL1A2-WT forelimbs were fractionated and specific fractions were analyzed via LC-MS/MS. Aha-enrichment experiments consisted of in vivo protein labeling with azidohomoalanine (Aha) followed by tissue fractionation of the forelimbs and enrichment of labeled ECM proteins from the final IN pellet ('enriched'). 'Unenriched samples', or the background from which newly synthesized proteins were enriched from, were also analyzed via LC-MS/MS.
