Project description:Epigenetic plasticity is critical for tumor transformation, progression, and response to immunotherapy. Although pseudouridine is the first discovered and most abundant epitranscriptomic modification, its cellular functions remain poorly understood. In this study, we identified pseudouridine synthase (PUS) as a key driver of tumor immune evasion. Specifically, we found that PUS was aberrantly overexpressed in tumors and correlated with tumor malignant progression as well as poor patient prognosis, particularly in patients with TP53 mutations. Notably, genetic ablation of PUS effectively suppressed tumor progression, increased T cell infiltration, and boosted T cell function in a de novo MYC/Trp53-/- mouse liver cancer, as well as in chemically induced liver cancer models. Mechanistically, PUS loss induced the expression of retrotransposon sequences, resulting in elevated levels of double-stranded RNA and subsequent activation of innate antiviral immune signaling. We further demonstrated the role of PUS1 in tumor immune evasion was dependent on its enzymatic activity. Importantly, inhibition of PUS1 could sensitize tumors to anti-PD-1 therapy in a MYC/Trp53-/- mouse liver cancer model. Similarly, we revealed that 5-fluorouracil could inhibit pseudouridine synthesis and significantly enhance the efficacy of PD-1 inhibition. Clinically, low PUS level showed a higher response rate and better clinical outcome to immune checkpoint blockade therapies. Overall, our findings demonstrate PUS1 as a critical regulator of immune evasion and targeting pseudouridine synthesis may a novel strategy to enhance immunotherapy efficacy by activating dsRNA-sensing pathways.

Project description:E. fischeriana has long been used as a traditional Chinese medicine.Recent studies reported that some compounds of E. fischeriana exhibited antimicrobial and immune enhance activity. Innate immune system is essential for the immune surveillance of inner and outer threats, initial host defense responses and immune modulation. The role of natural drug compounds, including E. fischeriana, in innate immune regulation is largely unknown. Here we demonstrate that E. fischeriana compound Dpo is involved in antiviral signaling. The genome wide RNA-seq analysis revealed that Dpo synergized the induction of ISGs by viral infection. Consistently, Dpo enhanced the antiviral immune responses and protected the mice from death during viral infection. Dpo however was not able to rescue STING deficient mice lethality caused by HSV-1 infection. The enhancement of ISG15 by Dpo was also impaired in STING, IRF3, IRF7 or ELF4 deficient cells, demonstrating that Dpo activates innate immune responses in a STING/IRFs/ELF4 dependent way. The STING/IRFs/ELF4 axis is therefore important for Dpo induced ISGs expression, and can be used by host to counteract infection.

Project description:In an effort to produce a mouse model of Mitochondrial Myopathy with Lactic acidosis and Sideroblastic Anemia (MLASA), we knocked out the gene for Pseudouridine synthase 1 (PUS1), an enzyme that modifies uridine to pseudouridine in many cytoplasmic and mitochondrial tRNAs, as well as other cellular RNAs. The Pus1-/- mice are viable, are born at the expected Mendelian frequency, and are non-dysmorphic. The PUS1 mRNA and certain pseudouridine modifications are absent in cytoplasmic and mitochondrial tRNAs in the Pus1-/- mice. The Pus1-/- mice display reduce exercise capacity at 14 weeks, with alterations in muscle morphology, histology, and physiology. Red gastrocnemius muscle from Pus1-/- mice shows reduced number and size of mitochondria and reduced Cytochrome C oxidase activity. Two-condition, two-color experiment: Mouse wild type PUS1 and homozygous mutant PUS1 kidney tissue samples: 4 biological replicates each.
