Project description:This study explores the use of short settling times as a strategy to enhance microbial selection and prevent bulking induced by nutrient shortage in an aerobic dynamic feeding (ADF) process using mixed microbial cultures (MMCs) for polyhydroxyalkanoate (PHA) production from fermentation digestate. A 5.0-L aerobic reactor was operated under three conditions: Long Settling (LS, 30 minutes), Short Settling (SS, 10 minutes), and Short Settling under Nutrient Shortage (SS-NS, 10 minutes with reduced nutrient load). Short settling significantly improved biomass settleability, reducing the Sludge Volume Index (SVI) from 126 to 25 mL g⁻¹, and promoted the formation of dense flocs enriched in PHA-accumulating bacteria, as confirmed by transmission electron microscopy. Process kinetics revealed enhanced substrate uptake rates and improved storage yields under SS conditions, despite an elevated C/P ratio. Notably, nutrient shortage conditions were inadvertently reached by the proliferation of Polytoma mirum in the feeding tank. Although Polytoma mirum did not affect VFA composition, its presence significantly reduced nitrogen and phosphorus concentrations. These nutrient shortage conditions were maintained for at least 30 days until the system began to exhibit stress, as demonstrated by an increased SVI (178.6 mL g⁻¹). Microbial community analyses indicated marked shifts: the eukaryotic assemblage transitioned from sessile to motile ciliates under SS, while bacterial diversity within the PHA‐accumulating fraction remained high, with key taxa such as Sphaerotilus and Neomegalonema becoming more prevalent under phosphorus-limited conditions. Overall, short settling not only improved microbial selection but also prevented bulking by retaining well-aggregated biomass, thereby mitigating nutrient shortage conditions.

Project description:We used whole genome transcriptome as gene discovery to further understand the differential behaviour of the first (embryonic) and second waves of thymic settling progenitors in the murine embryo. Mouse embryos at days 13 and 18 of gestation were isolated and the thymic lobes were dissected. Single cell suspension was prepared and treated with biotinilated antibodies to CD3, CD4, CD8, TER119, Gr1, CD25, CD19, NK1.1 and CD11c. Cell suspensions were then incubated with streptavidin Miltenyi Macs beads. Lineage negative thymocytes were recovered in Macs columns. Cell suspensions were then incubated with antibodies against CD135, CD117, CD44, CD24 and Streptavidin bound to PECy7. Lin-, CD117+, CD44+, CD24low, CD135+ cells were then isolated by cell sorting. 3 biological replicates of each sample were isolated with 104-3x104 cells

Improved drag coefficient and settling velocity for carbonate sands.

Project description:Sediment transport calculations are used globally in the numerical models that coastal managers, scientists and engineers use to assess and forecast coastal change. Most of the existing sediment transport equations were defined based on experimental results using siliciclastic sands. Yet these equations are applied to all types of sand, including carbonate sands that have different characteristics and therefore, settling behaviour. A rigorous management of the transport of carbonate sand is essential for the present and future management of sedimentary features in coral reefs such as sandy beaches or reef islands. Here we present a new approach to estimating the drag coefficient of carbonate sands that considers both friction and pressure. Based on our new method, the calculated drag coefficients explain the great variability in settling velocities of carbonate sand observed in nature (from 0.025 m/s to 0.364 m/s in our database). Using our formula, we demonstrate that even small differences in the settling velocity obtained with the new drag coefficient can lead to substantial changes in sediment transport and call for an update of numerical models.
