Project description:Diverse dataset of 1247 dogs from many breeds and wolves used to investigate the origins of dog domestication DNA for 1228 dogs from 35 breeds and 19 wolves was extracted from whole blood samples and genotyped on the Affymetrix Canine v2 Arrays. Genotypes were called using Affymetrix's snp5-probeset-genotype software and the BRLMM-P calling algorithm. The included breed designations are owner reported.

Project description:Transcriptome analysis of hemangiosarcoma samples were performed to assess EGFR and PLAUR/uPAR expression profiles and compare to human tumors from TCGA. Abstract Sarcomas differ from carcinomas in their mesenchymal origin. Therapeutic advancements have come slowly so alternative drugs and models are urgently needed. These studies report a new drug for sarcomas that simultaneously targets both tumor and tumor neovasculature. eBAT is a bispecific angiotoxin consisting of truncated, deimmunized Pseudomonas exotoxin fused to epidermal growth factor (EGF) and the amino terminal fragment (ATF) of urokinase. Here, we study the drug in an in vivo “ontarget” companion dog trial since eBAT effectively kills canine hemangiosarcoma (HSA) and human sarcoma cells in vitro. We reasoned the model has value due to the common occurrence of spontaneous sarcomas in dogs and a limited lifespan allowing for rapid accrual and data collection. Splenectomized dogs with minimal residual disease were given one cycle of eBAT followed by adjuvant doxorubicin in an adaptive dose-finding, phase I- II study of 23 dogs with spontaneous, stage I-II, splenic HSA. eBAT improved 6-month survival from <40% in a comparison population to ~70% in dogs treated at a biologically active dose (50 μg/kg). Six dogs were long-term survivors, living >450 days. eBAT abated expected toxicity associated with EGFR-targeting, a finding supported by mouse studies. Urokinase plasminogen activator receptor (uPAR) and EGFR are targets for human sarcomas, so thorough evaluation is crucial for validation of the dog model. Thus, we validated these markers for human sarcoma targeting in the study of 212 human and 97 canine sarcoma samples. Our results support further translation of eBAT for human patients with sarcomas and perhaps other EGFR-expressing malignancies.

Project description:DNA structural variation (SV) comprises a major portion of genetic diversity, but its biological impact is unclear. We propose that the genetic history and extraordinary phenotypic variation of dogs make them an ideal mammal in which to study the effects of SV on biology and disease. The hundreds of existing dog breeds were created by selection of extreme morphological and behavioral traits. And along with those traits, each breed carries increased risk for different diseases. We used array CGH to create the first map of DNA copy number variation (CNV) or SV in dogs. The extent of this variation, and some of the gene classes affected, are similar to those of mice and humans. Most canine CNVs affect genes, including disease and candidate disease genes, and are thus likely to be functional. We identified many CNVs that may be breed or breed class specific. Cluster analysis of CNV regions showed that dog breeds tend to group according to breed classes. Our combined findings suggest many CNVs are (1) in linkage disequilibrium with flanking sequence, and (2) associated with breed specific traits. We discuss how a catalog of structural variation in dogs will accelerate the identification of the genetic basis of canine traits and diseases, beginning with the use of whole genome association and candidate CNV/gene approaches. Project description:Post-traumatic osteoarthritis (PTOA) is a common sequela to anterior cruciate ligament (ACL) injury in humans and dogs. Identification of conserved targets between species may hasten biomarker identification and therapeutic development. To identify differentially regulated synovial fluid (SF) proteins in ACL injury, SF samples were collected from knees with ACL injury or control knees (n=8 per species) in human and dog patients. The SF proteome was evaluated using nano-scale reverse phase chromatography and tandem mass spectroscopy. Associations between proteins and ACL injury were analyzed in both species. Immunoassays were used to validate key proteins identified via mass spectrometry, including periostin, α-2-macroglublin, and lubricin, in additional SF samples (dog: n=57 ACL, n=31 controls; human: n=20 ACL, n=15 advanced OA) and plasma samples (human: n=18 ACL, n=15 advanced OA, and n=12 controls). Periostin was the most upregulated SF protein (log2 fold-change) in both dog (2.6) and human (3.5) ACL injury, followed by complement C1q (2.3) and α-2-macroglobulin (1.8) in dogs and fibrinogen (1.1) and α-2-macroglobulin (1.1) in humans. Top downregulated proteins included serum amyloid A1 (-1.9) and aggrecan (-1.8) in dogs and carbonic anhydrase 2 (-3.2) and hemoglobin subunit β (-2.8) in humans. Approximately 60% of proteins detected were shared between species, and immunoregulatory and macromolecular transport proteins were the most common families. Clinical Relevance: Findings in both species support further investigation into periostin as a potential biomarker or therapeutic target and the use of spontaneous ACL injury in dogs as a potential translational large animal model for human ACL injury.

Project description:Companion dog care market is annually increasing. A relationship with a dog who lives with humans gives a lot of psychological stability. Therefore, companion dog's health is important to the owner. It would be of great help if a non-invasive method could monitor health through bodily fluids (urine, serum). In addition to this, it is principal to discover and verify protein biomarkers that can detect the presence or absence of disease. Despite the growing application of proteomics in biomarker research using canine serum or urine, there is no spectral assay library publicly available. Here, we set up spectral assay libraries for canine serum and urine samples using 24 midpH-fractionated samples to enable deep proteome coverage. The canine serum assay library includes 4,749 protein groups and 25,228 peptides and urine assay library contains 1,132 protein groups and 5,483 peptides.
