Project description:Immune dysregulation is increasingly recognized as a key contributor to Parkinson’s disease (PD) pathogenesis. While microglia are established central nervous system (CNS) immune regulators, the role of CNS-associated macrophages (CAMs) and their upstream modulation by immune checkpoints remains poorly defined. Here, we identify CD200R1, a myeloid-specific inhibitory receptor, as a checkpoint selectively expressed by embryonically derived CAMs at CNS borders. CD200R1 deletion reprograms CAMs toward a proinflammatory, antigen-presenting state enriched in interferon and pyroptosis-related gene signatures. In α-synuclein–based PD models, CD200R1 loss exacerbates dopaminergic neurodegeneration, enhances CD8⁺ T cell infiltration, and promotes formation of tertiary microglial–T cell nodules. Mechanistically, it activates the caspase-8/GSDMD pyroptosis pathway in CAMs, creating a permissive environment for pathogenic T cells. Pyroptosis blockade with disulfiram attenuates T cell infiltration and neurodegeneration. Notably, pharmacological activation of CD200R1 via CD200Fc suppresses CAM activation and confers neuroprotection. These findings define a border-specific immune checkpoint that governs CAM–T cell crosstalk and represents a tractable therapeutic target in PD.

Project description:Systemic inflammatory reactions mediated by chronic infections activate microglia in the central nervous system (CNS) and have been postulated to exacerbate neurodegenerative diseases. We now demonstrate in vivo that repeated systemic challenge of mice with bacterial lipopolysaccharides (LPS) maintains an elevated microglial inflammatory response and triggers neurodegeneration. Repeated chronic intraperitoneal application of LPS over four consecutive days induced loss of dopaminergic neurons in the substantia nigra, a process that was accompanied by decreased levels of dopamine in the striatum. In contrast, total cumulative LPS dose given intraperitoneally within a single acute application did not induce a decrease in dopamine levels nor neurodegeneration. Mice that received repeated systemic LPS application showed increased microglial activation, elevated production of proinflammatory cytokines and activation of the classical complement and its associated phagosome pathway in the brain. Loss of dopaminergic neurons induced by repeated systemic LPS application was rescued in complement C3 deficient mice, confirming an involvement of the complement system in neurodegeneration. Thus, our data demonstrate that repeated systemic exposure to bacterial LPS induces a microglial phagosomal inflammatory response, leading to complement-dependent damage of dopaminergic neurons.

Project description:Disruption of local iron homeostasis is a common feature of neurodegenerative diseases. We focused on dopaminergic neurons, asking how iron transport proteins modulate iron homeostasis in vivo. Inactivation of the transmembrane iron exporter ferroportin had no apparent consequences. However, loss of the transferrin receptor 1, involved in iron uptake, caused profound, age-progressive neurodegeneration with features similar to Parkinson’s disease. There was gradual loss of dopaminergic projections in the striatum with subsequent death of dopaminergic neurons in the substantia nigra. After depletion of 30% of the neurons the mice developed neurobehavioral parkinsonism, with evidence of mitochondrial dysfunction and impaired mitochondrial autophagy. Molecular analysis revealed strong signatures indicative of attempted axonal regeneration, a metabolic switch to glycolysis and the unfolded protein response. Project description:Apolipoprotein E4 (APOE4) is the strongest genetic risk factor for late-onset Alzheimer’s disease (LOAD), leading to earlier age of clinical onset and exacerbating pathologies. There is a critical need to identify protective targets. Recently, a rare APOE variant, APOE3-R136S (Christchurch), was found to protect against early-onset AD in a PSEN1-E280A carrier. We sought to determine if the R136S mutation also protects against APOE4-driven effects in LOAD. We generated tauopathy mouse and human iPSC-derived neuron models carrying human APOE4 with the homozygous or heterozygous R136S mutation. We found that the homozygous R136S mutation rescued APOE4-driven Tau pathology, neurodegeneration, and neuroinflammation. The heterozygous R136S mutation partially protected against APOE4-driven neurodegeneration and neuroinflammation, but not Tau pathology. Single-nucleus RNA-sequencing revealed that the APOE4-R136S mutation increased disease-protective and diminished disease-associated cell populations in a gene dose-dependent manner. Thus, the APOE-R136S mutation protects against APOE4-driven AD pathologies, providing a target for therapeutic development against AD.

Project description:Emerging evidence suggests that Parkinson’s disease (PD) may have its origin in the enteric nervous system (ENS), from where alpha-synuclein (αS) pathology spreads to the brain. Decades before the onset of motor symptoms, PD patients suffer from constipation and present with circulating T cells responsive to αS, suggesting that peripheral immune responses initiated in the ENS may be involved in the early stages of PD. However, cellular mechanisms that trigger αS pathology in the ENS and its spread along the gut-brain axis remain elusive. Here, we demonstrate that muscularis macrophages (ME-Macs), housekeepers of ENS integrity and intestinal homeostasis, modulate αS pathology and neurodegeneration in models of PD. ME-Macs contain misfolded αS, adopt a signature reflecting endolysosomal dysfunction, and modulate the expansion of T cells that travel from the ENS to the brain via the dura mater as αS pathology progresses. Directed ME-Mac depletion leads to reduced αS pathology in the ENS and CNS, prevents T cell expansion, and mitigates neurodegeneration and motor dysfunction, suggesting a role for ME-Macs as early cellular initiators of αS pathology along the gut-brain axis. Understanding these mechanisms could pave the way for early-stage biomarkers in PD.

Project description:Neuroinflammation plays a key role in modulating dopaminergic neuronal (DAN) cell loss in Parkinson’s disease (PD). However, it remains unresolved how to effectively normalize this immune response given the complex interplay between the innate and adaptive immune responses occurring within a scarcely accessible organ like the brain. In this study, we uncovered a consistent correlation between neuroinflammation, brain parenchymal lymphocytes and DAN cell loss among several commonly used mouse models of PD generated by a variety of pathological triggers. We validated a viral therapeutic approach for the microglia-specific expression of Interleukin 10 (IL-10) to selectively mitigate the excessive inflammatory response. We found that this approach induced a local nigral IL-10 release that sustained a robust neuroprotection against α-Synuclein (αSYN)-induced neurodegeneration. Single-cell transcriptomics revealed the emergence of a molecularly alternative microglial cell state specifically induced by IL-10 enriched in markers of cell activation with enhanced expression of prophagocytic pathways. Indeed, Il-10 strongly promoted phagocytotic and clearance activities that effectively reduced the αSYN aggregate burden in the nigral area. Furthermore, IL-10 stimulated the differentiation of CD4+ T lymphocytes into active T regulatory cells. Finally, CD8+ T cells exhibited enhanced inhibitory characteristics in the presence of IL-10. Thus, IL-10 local transduction elicited a strong immunomodulation in the nigral tissue with enhanced suppression of lymphocyte toxicity, which rescued DAN cell loss. These results offer insights into the therapeutic benefits induced by IL-10, showcasing a promising gene delivery approach that minimizes undesired side effects and has a strong translational relevance.
