Project description:Olivocochlear (OC) efferent neurons are the final component of the descending auditory system. The lateral olivocochlear (LOC) neurons innervate type I spiral ganglion neurons (SGN). Their role in hearing is unknown, but they may have roles in protection from noise trauma or changing the threshold of SGN activation. The medial olivocochlear (MOC) efferent neurons innervate outer hair cells (OHC), inhibiting OHC function and thus dampening cochlear vibrations. LOC and MOC function is poorly understood at the single cell level, but recent use of transgenic mice (ChAT-IRES-Cre; tdTomato) expressing fluorescent reporters in cholinergic neurons, including LOC and MOC neurons, has allowed patch-clamp recordings from the neurons in mouse brainstem slices to begin to characterize their intrinsic electrical properties and synaptic activity. Recent single nucleus sequencing experiments from LOC and MOC neurons has given insight into the genetic programs that generate their function, including changes to expression profiles following noise trauma. Here we utilize the PatchSeq technique to combine patch-clamp electrophysiological recordings with deep single-cell transcriptome analysis from identified LOC and MOC neurons in transgenic ChAT-IRES-Cre; tdTomato mice. In this preliminary dataset, we generated patch-clamp electrophysiology data and single cell transcriptome data from 18 identified LOC and 13 identified MOC neurons.

Intermediolateral column, medial, and lateral motoneurons

Project description:Three cell types, intermediolateral column motoneurons, medial motoneurons, and lateral motoneurons were isolated from a single adult spinal cord using laser capture microscopy. Four hundred captures were collected for each cell type. For a given cell type, RNA was extracted from the 400 captures using an Arcturus picopure kit. RNA was split in half and two targets were produced using a double amplification protocol. Each target was hybridized to Affymetrix chips and signals were normalized with R-pack. Inverse logs are provided. Five animals were used in these experiments, and all three cell types were collected from each animal. Thus, for each cell type, there are five biological replicates, and for each biological replicate there are two technical replicates. In all thirty chips were analyzed. Techinical replicates are indicated as Set 1 and Set 2. Animal numbers are indicated by Pair1 through Pair 5. Keywords: other

Transcription profiling of human Parkinsons medial and substania nigra

Project description:Affymetrix HG_U133 array sets (A and B chips) were used to determine the whole genome transcription profile of clinically documented and neuropathologically confirmed cases of sporadic Parkinson's disease as well as controls. Experiment Overall Design: Post mortem brain tissue samples from Parkinson's disease and control cases (substantia nigra, split into medial and lateral portions, and frontal cortex) were prepared for this study. In total, 47 individual tissue samples were analyzed using one A and one B GeneChip per sample, i.e. 15 samples of medial parkinsonian SN, 9 samples of lateral parkinsonian SN, 8 medial nigra control samples and 7 lateral nigra control samples. Lateral and medial nigra samples were from the same cases.
