Project description:affy_infection_medicago - affy_infection_medicago - MtNF-YA1 is a transcription factor involved in the regulation of nitrogen fixing nodule organogenesis. Mtnf-ya1-1 mutant has been shown to be more resistant to Aphanomyces euteiches (Ae), a root pathogen, than the A17 wild type (WT) line. In this study we want to compare both WT and mutant plant responses, 1 and 6 days post inoculation (dpi) between WT and the Mtnf-ya1-1 mutant, in order to identify MtNF-YA1-dependent gene networks during infection. Data about non inoculated A17 control plants and infected A17 at 1 dpi have been already obtained in another experiment in which the conditions of in vitro culture of M. truncatula plants and ofA.euteiches(Ae) inoculation were exactly the same than the ones used in these experiments.-Fifteen-day-old A17 or Mtnf-ya1-1 plants were grown in vitro on M medium. The entire roots were then inoculated (or not = controls) with a A. euteiches zoospores (105/ml) and harvested one or six days after inoculation. Three independent repeats were performed.

Project description:We performed a split-root experiment with non-nodulated roots of Medicago truncatula (A17 accession) to identify systemic molecular responses induced by the inoculation of the oomycete Aphanomyces euteiches. The root systems of nodulated symbiotic plants were divided into two compartments called the \"local root compartment\" (treated) and the \"systemic root compartment\", from which samples were obtained for RNAseq analysis. Roots, localised in the \"systemic compartment\", were harvested in inoculated or control (mock-inoculated) plants at the same time-points, after 6 hours, 1-, 3- and 6-days after A. euteiches inoculation.

To gain insight into the mechanism underlying partial resistance to this pathogen, a pathosystem has been developed using the model legume Medicago truncatula (Mt). Two Mt accessions A17 and F83005.5 have been shown to display differential responses upon infection with Ae (partially resistance and susceptibility, respectively). Genetic analyses using a RIL population derived from a cross between those two lines lead to the identification of, prAe1, a major QTL involved in the observed partial resistance. Near isogenic lines (NIL) NR2 and NS2 whose genomes are identical except at the prAe1 locus (resistant alleles for NR2, susceptible alleles for NS2) have been generated, and the same differential response toward Ae has been found. The aim of this experiment is to analyse and compare NR2 and NS2 transcriptomes at 1 and 6 days after Ae inoculation to identify gene networks that are regulated in a prAe1 dependent manner.--Fifteen-day-old NR2 or NS2 plants were grown in vitro on M medium. The entire roots were then inoculated (or not = 0dpi) with a A. euteiches zoospores (75000zsp/ml) and harvested one or six days after inoculation. Three independent repeats were performed.

A reverse genetic approach was undertaken to study the role of this gene in the pathosystem Medicago truncatula – Aphanomyces euteiches (Ae). Results suggest that lyr3-1 and lyr3-2 mutants are more susceptible to the root pathogen Aphanomyces euteiches than the A17 wild type (WT) line.
