Project description:In the present study, we compared transcriptional response to salinity between male and female individuals of Populus yunnanensis. We found that several functional groups of genes involved in important pathways were differentially expressed, including photosynthesis-related genes which were mainly up-regulated in males but down-regulated in females. This gene expression pattern is consistent with physiological observation that salinity inhibited photosynthetic capacity more in females than in males. In conclusion, our study provided molecular evidence of sexual differences in poplar salinity tolerance. Identified sex-related genes in salinity tolerance and their functional groups will enhance our understanding of sexual differences to salinity stress at the transcription level.

Project description:Illumina technology was used to generate mRNA profiles of stem apex of Populus yunnanensis with cutting and inverted cutting. Total RNA was extracted separately from each plant and pooled to three biological replicates per condition. RNA concentration and purity was measured using NanoDrop 2000(Thermo Fisher Scientific, Wilmington, DE). RNA integrity was assessed using the RNA Nano 6000 Assay Kit of the Agilent Bioanalyzer 2100 system (Agilent Technologies, CA, USA).

Project description:In low rainfall regions soils are naturally conditioned with frequent co-occurrence of salinity and alkalinity. Plant salinity responses both at physiological and molecular level have been extensively researched. However, effects of the combined treatment of alkaline salinity that could greatly reduce plant growth and the mechanisms responsible for tolerance remain indeterminate. In Brassica juncea, large reductions in biomass and increased leaf Na+ concentration under alkaline salinity indicates that the combined treatment had greater negative effect than salinity on both growth and the physiological responses of the plant. To determine molecular mechanisms potentially controlling adaptive tolerance responses to salinity and alkaline salinity, the moderately tolerant genotype NDR 8501 was further investigated using microarray analysis. The transcripts of treated leaf tissues verses those of the untreated control sample were analysed after prolonged stress of four weeks. In total, 528 salinity responsive and 1245 alkaline salinity responsive genes were indentified and only 101 genes were expressed jointly in either of the two treatments. Transcription of 37% more genes involved in response to alkaline salinity than salinity alone, which suggests the increased impact and severity of the combined stress on the plant, indicating the transcription of a far greater number of genes likely involved in mitigation and damage control. Transcription of KUP2 and KUP7 genes involved in potassium homeostasis under salinity alone and NHX1 and ENH1 genes for ion (K+ and Na+) homeostasis under alkaline salinity, clearly demonstrated that different genes and genetic pathways are involved in response to each stress. They further provide supporting evidence for the physiological responses that occur in the plant, with massive reprogramming of the transcriptome leading to partial ion exclusion, shuttling and compartmentation.

Salinity and alkaline salinity induced gene expression in Brassica juncea leaf was measured at 4 weeks of prolonged treatment of 50 mM NaCl alone and combined with 2.5 mM HCO3- versus non-stressed control. A single experiment was conducted using Brassica juncea genotype NDR 8501 at a single time point (fours weeks after treatment) with three replications per treatment.

Project description:Alkali-salinity is a major abiotic stress that limits plant growth and productivity. Studying mechanisms of alkali-salinity tolerance in halophytic plants will provide valuable information for underlying plant alkali-salinity tolerance. Puccinellia tenuiflora is considered as an ideal model plant for studying the alkali-salinity tolerant mechanisms in plants. In this study, the NaHCO3-responsive molecular mechanisms in P. tenuiflora leaves were investigated using a combined physiological and proteomic approaches. Our results implied some specific NaHCO3-responsive mechanisms in leaves from P. tenuiflora. They are (1) reduction of photosynthesis attributed to the decrease of the abundance of Calvin cycle enzymes, (2) accumulation of Na+ and K+ caused ion-specific stress, (3) accumulation of proline, soluble sugar and betaine enhanced the ability of osmotic regulation, (4) diverse reactive oxygen species (ROS) scavenging mechanisms under different NaHCO3 concentrations, and (5) alternative protein synthesis and processing strategies in chloroplast and cytoplasm. All these provide important evidence for understanding NaHCO3-responsive mechanisms in P. tenuiflora.

Project description:Salinity stress poses a significant risk to agricultural yield and productivity. Therefore, elucidation of plant salt-response mechanisms has become essential to identify stress-tolerance genes. In the present study, two pearl millet genotypes with contrasting salt-tolerance showed differential morpho-physiological and proteomic responses under 150 mM NaCl, and the genotype IC 325825 could withstand salt-stress better than IP 17224. The salt-tolerance potential of IC 325825 was associated with its ability to maintain ionic, osmotic balance and membrane integrity under stress. The IC 325825 exhibited better growth under salinity as compared to IP 17224 due to higher expression of C4 photosynthesis enzymes, efficient antioxidant system, and lower Na+/K+ ratio. Comparative proteomics analysis revealed greater metabolic perturbation in IP 17224 under salinity, in contrast to IC 325825 that harbored pro-active stress-responsive machinery, allowing its survival and better adaptability under salt-stress. The differentially expressed proteins were in-silico characterized for their functions, subcellular-localization, pathway/ interaction analysis, and relative transcript levels. This study has provided novel insights into salinity stress adaptive mechanisms in pearl millet, demonstrating the power of proteomics-based approaches. The critical proteins identified in the present study could be further explored as potential objects for increasing salt-tolerance in sensitive crop plants.
