Project description:Purpose: The goal of this study is to compare NGS-derived transcriptome profiling (RNA-seq) in AD patients’ hippocampus. Methods: mRNA profiles of human hippocampus from AD patients and healthy controls were generated by deep sequencing, using Illumina GAIIx. Results: Using an optimized data analysis workflow, more than 20000 transcripts in hippocampus were detected. Top 6 genes were selected and related to “ion transport”. Correlation analysis demonstrated negative relationship of gene expression level with the neuropathologic changes.
Project description:To understand the genetic regulation of gene expression and patterns of gene co-expression, we sequenced the transcriptome of the hippocampus of founder mice of both sexes. Mice were sacrificed between 6-8 weeks of age and hippocampus dissected. Total hippocampal RNA was isolated using a TRIzol Plus RNA purification kit (Life Technologies) and mRNA sequencing library was prepared using a TruSeq kit (Illumina), both according to manufacturer's protocols. Paired-end 100bp reads were obtained using the Illumina HiSeq 2000.

DO mice (fourth and fifth generations of outcrossing) were sacrificed between 6-8 weeks of age and hippocampus dissected.
