Project description:Gene expression profiling comparisons of HepG2.2.15 or PLC/PRF/5 cells either mock (M) transfected or transfected with 0.2 microM S2 RNA or Scrambled (SCR) siRNA were carried out in duplicate 48 hours after transfection. Project description:Analysis of LNCaP cell molecular differences and their response to R1881 in 2D and 3D cultures. Androgen regulated genes were differentially expressed between 2D and 3D cultures. These results provide insights into factors that influence the expression of androgen regulated genes In this study, LNCaP cells cultured in tissue culture plastic (2D) and in the hydrogel (3D) were maintained up to 3 days and 24 days respectively in serum containing media before they were androgen-starved for 48 hours. Cells were either treated with 1nM R1881 or continued to be androgen-deprived (without R1881 with 0.008% ethanol) for another 48 hours prior to cell harvest for gene expression analysis. Biological triplicates were prepared for each condition.

Effect of PARG or NUDIX5 inhibition on Progesterone Gene Expression Profiling

Project description:T47D cells were starved 0% FCS for 24 hours. Following starvation PARG was inhibited via the use of Tannic Acid (TA) and the gene expression in response to progesterone studied. Cells were stimiulated with R5020 for 1 or 6 hours. NUDIX5 was reduced in T47D cells 48 hours prior to hormone induction via the use of NUDIX5 specific siRNA. Progesterone response and gene expression changes were analyzed as described above following 6 hours of R5020.

Project description:We identified that knocking down Map4k4 in endothelial cells affected genes associated with the cell cycle, mitosis, and inflammatory genes. We used microarrays to identify genes of interest that are differentially expressed after the addition of scrambled or Map4k4 siRNA. HUVECs were treated with scrambled or siMap4k4. 24 hours later, cells were serum starved overnight. Cells were prepared with this protocol on three different occasions, and total RNA was isolated to provide three biological replicates per sample.
