Carnosic acid in topical rosemary extract promotes TRPA1-mediated scarless wound healing

Project description:Mammalian skin wounds typically heal with a scar, characterized by fibrotic tissue that disrupts original tissue architecture and function. Therapies to reduce fibrosis and scar formation remain a large unmet clinical need. Rosemary extract, particularly in the form of topical oils and creams, has gained massive popularity within the general public as a potential treatment to reduce scar formation. However, its efficacy and potential mechanism of action remain unknown. We show in adult wound healing mouse models that an ethanol-based rosemary extract reduces fibrosis and promotes scarless wound healing. More specifically, carnosic acid within rosemary leaves acts as an active ingredient and stimulates the TRPA1 nociceptor on cutaneous sensory neurons to promote scarless wound healing. Mice lacking TRPA1 on sensory neurons do not exhibit rosemary-induced tissue regeneration. Taken together, topical rosemary extract may be an effective and cost-efficient therapeutic to reduce scar formation in skin wounds.

Modulation of gene expression induced by supercritical fluid rosemary extract in colon cancer cells

Project description:Supercritical rosemary extract (containing 16.90% carnosic acid, 1.90% carnosol and 13.59% volatile compounds) showed antitumor activity on colon cancer cells in vitro. We treated colon cancer cells with the extract and we employed whole genome microarray expression profiling to identify genes potentially involved in its antitumor mechanism of action. We analyzed gene expression of colon cancer SW620 cells after treating during 48h with supercritical rosemary extract at concentrations that cause 50% inhibition of cell viability (30 M-NM-<g/mL), citostatic effect (60 M-NM-<g/mL) and 50% cell death (100 M-NM-<g/mL), in comparison to control cells (0 M-NM-<g/mL). Two independent experiments were performed in triplicate. Each sample is the pool of the triplicates of one of the experiments (a or b) at the indicated concentration.

A novel micronutrient blend mimics calorie restriction transcriptomics in multiple tissues of mice and increases life and healthspan in C. Elegans

Project description:Calorie restriction is a major intervention consistently demonstrated to retard aging and delay age-associated diseases. A novel micronutrient blend, a putative calorie restriction mimetic, was developed based on a screening tool we previously described. Whole transcriptomic analysis was examined in brain cortex, skeletal muscle and heart in three groups of mice: old controls (30 months), old + calorie restriction and old + novel micronutrient blend. The micronutrient blend elicited transcriptomic changes in a manner similar to those in the calorie-restricted group and unique from those in the control group. Subgroup analysis revealed that nuclear hormone receptor, proteasome complex and angiotensinogen genes, all of which are known to be directly related to the aging process, were the most affected by the micronutrient blend and by calorie restriction. Thus, these three genes may be considered master regulators of the favorable effects of calorie restriction and of the micronutrient blend. Based on the calorie restriction mimetic effects on transcriptomics, it was hypothesized that the micronutrient blend would promotes longevity and vitality. To test this hypothesis, a functional analysis in C. Elegans was used to examine the effects of the micronutrient blend on longevity and biomarkers of vitality. Results indicate that feeding C. Elegans the micronutrient blend increased longevity as well as vitality. Further studies are required to confirm that the calorie restriction mimicking benefits described here are elicited by the micronutrient blend in humans.
