Differing proteome responses to ocean acidification between two common pocilloporid corals

Project description:Ocean acidification increasingly threatens coral reef ecosystems by challenging calcifi-cation processes fundamental to reef accretion. Yet, many corals continue to calcify under elevated pCO₂, suggesting species-specific physiological plasticity and potential cellular compensations. Here, we investigate the proteomic responses of two common pocilloporid corals, Stylophora pistillata and Pocillopora damicornis, to moderate (~940 ppm) and high (~2,800 ppm) pCO₂ over a two-month period. Using label-free quantita-tive proteomics, we identify changes in protein abundances compared to control con-ditions (~480 ppm) and infer functional shifts that underlie their respective responses to acidification. S. pistillata exhibited extensive proteomic restructuring under high pCO₂, marked by downregulation of metabolic pathways and concurrent upregulation of ion transporters, cytoskeletal elements, and stress response proteins, suggesting a meta-bolically costly but flexible strategy to sustain calcification. In contrast, P. damicornis showed fewer and more moderate proteomic changes, with adjustments in structural proteins and redox regulation, indicating a more restrained acclimatization strategy. These divergent responses highlight contrasting modes of resistance (plasticity versus stability). In direct context to physiological and geochemical data, our findings reveal cellular mechanisms of calcification control and underscore the utility of proteomics in coral ecophysiology, offering new insights into species-specific vulnerability and po-tential adaptive capacities under future ocean conditions.

Longevity strategies in response to light in the reef coral Stylophora pistillata

Project description:Aging is a multifactorial process that results in progressive loss of regenerative capacity and tissue function while simultaneously favoring the development of a large array of age-related diseases. Evidence suggests that the accumulation of senescent cells in tissue promotes both normal and pathological aging. Oxic stress is a key driver of cellular senescence. Because symbiotic long-lived reef corals experience daily hyperoxic and hypoxic transitions, we hypothesized that these long-lived animals have developed specific longevity strategies in response to light. We analyzed transcriptome variation in the reef coral Stylophora pistillata during the day–night cycle and revealed a signature of the FoxO longevity pathway. We confirmed this pathway by immunofluorescence using antibodies against coral FoxO to demonstrate its nuclear translocation. Among genes that were specifically up- or downregulated on exposure to light, human orthologs of two “light-up” genes (HEY1 and LONF3) exhibited anti-senescence properties in primary human fibroblasts. Therefore, these genes are interesting candidates for counteracting skin aging. We propose a large screen for other light-up genes and an investigation of the biological response of reef corals to light (e.g., metabolic switching) to elucidate these processes and identify effective interventions for promoting healthy aging in humans.

RNA-Seq of S.pistillata samples exposed to a 6-months darkness

Project description:Telomere DNA length is a complex trait controlled both by multiple loci and environmental factors. Even though the use of telomere DNA length measurement, as a method of assessing stress accumulation and predicting how this will influence survival, is currently being studied in numerous human cohort studies, the importance of telomere length for stress response in ecological studies remains at its infancy. Here, we investigated the telomere changes occurring in the symbiotic coral Stylophora pistillata that has experienced a continuous darkness over 6 months. This stress condition led to the loss of its symbionts, as what is also observed when bleaching occurs in the field at a large-scale due to climate changes and anthropogenic activities, threatening the worldwide reef ecosystem. We found that the continuous darkness condition was associated to telomere DNA length shortening and a downregulation of the expression of the telomere-associated protein POT2. These results pave the way for future studies on the role of telomere in coral stress response and the importance of telomere dysregulation in endangered coral species

Identifying Genes Associated with the Molecular Scleractinian Coral Calcification Process (Stylophora pistillata study case)

Project description:Increasing seawater’s calcium concentration has shown to increase reef building (scleractinian) coral’s calcification rates. In this way the expression of the genes that are associated with the calcification process also altered and, thus can be identified. Needless to say that the overall gene repertoire that participate in the coral calcification process and its molecular mechanisms have not yet been revealed, although sporadic genes that are related to the process have been discovered and investigated. In this study, nubbins of the Red Sea scleractinian coral, Stylophora pistillata were treated with increased calcium concentrations seawater (addition of 100 gm/L) and the genes that have been up-regulated were compared to the genes expression profile of corals with natural seawater calcium concentration. Measurements of AT were taken at mid-day (11:00) and in nighttime (23:00), to record the calcification rates of coral individuals under normal and increased calcium seawater concentrations. In order to reveal the gene involved in the calcification process, S. pistillata fragments of normal and of increased calcium concentrations were sampled for microarray RNA transcriptional profiling at two time-points (mid-day and nighttime).Results of this study have revealed that Smad genes may play a role in the coral skeletal growth apparatus. This study show that the calcification molecular mechanism is conserved Among identified genes are large group of genes that are characterized in the TGF-b/BMP signal transduction pathways which have been revealed in other organisms to participate in bone and cartilage tissue development molecular processes.
