Project description:Background: Responses to hypoxia have been investigated in many species; however, comparative study between conspecific geographical populations in different altitude regions is rare, especially for invertebrates . The migratory locust, Locusta migratoria, is widely distributed both on high-altitude Tibetan Plateau (TP) and on low-altitude North China Plain (NP). TP locusts have inhabited Tibetan Plateau since Quaternary glaciations events and thus probably have evolved superior capacity to deal with hypoxia. Results: Here we compared the hypoxic responses of TP and NP locusts from morphological, behavioral and physiological perspectives. We found that TP locusts were more tolerant of extreme hypoxia than NP locusts, with a lower proportion exhibiting stupor, a faster recovery time, and higher respiration rates. We compared the transcriptional profiles of field TP and NP locusts and found that their differences were possibly attributed to a combination of multiple factors, e.g. oxygen, UV radiation, temperature and nutrition. To evaluate why TP locusts respond to extreme hypoxia differently from NP locusts, we subjected them to extreme hypoxia and compared their transcriptional responses. We found that the aerobic metabolism was more active in TP locusts than in NP locusts. RNAi disruption of PDHE1b, an entry gene from glycolysis to TCA cycle, increased the ratio of stupor in Tibetan locusts and decreased the ATP content of Tibetan locusts in hypoxia, confirming the significant importance of this metabolic branch for TP locusts to conquer hypoxia. Conclusions: Here we show that TP locusts are better tolerant of hypoxia than NP locusts and the better capacity to modulate primary metabolism in TP locusts contributes to their superior tolerance of hypoxia compared to NP locusts.

Project description:Contemporary wisdom suggests that inclusion of legumes into crop rotations benefit subsequent cereal crop yields. To investigate whether this maxim was generically scalable, we contrast summer soybean-winter wheat (SW) with summer maize-winter wheat (MW) rotation systems in an extensive field campaign in the North China Plain (NCP). We identify heretofore unseen interactions between crop rotation, synthetic N fertilizer application, and stored soil water. In the year with typical rainfall, inclusion of soybean within rotation had no effect on wheat ear number and yield, while N fertilization penalized wheat yields by 6-8%, mainly due to lower dry matter accumulation after anthesis. In contrast, in dry years prior crops of soybean reduced the rate and number of effective ears in wheat by 5-27 and 14-17%, respectively, leading to 7-23% reduction in wheat yield. Although N fertilization increased the stem number before anthesis in dry years, there was no corresponding increase in ear number and yield of wheat in such years, indicating compensating reduction in yield components. We also showed that N fertilization increased wheat yield in MW rather than SW as the former better facilitated higher dry matter accumulation after flowering in dry years. Taken together, our results suggest that soybean inclusion reduced soil available water for subsequent wheat growth, causing yield penalty of subsequent wheat under drought conditions. We call for more research into factors influencing crop soil water, including initial state, crop water requirement, and seasonal climate forecasts, when considering legumes into rotation systems. Project description:The distribution frequency of pathogenic fungi associated with root and crown rot of winter wheat (Triticum aestivum) from 104 fields in the North China Plain was determined during the period from 2013 to 2016. The four most important species identified were Bipolaris sorokiniana (24.0% from roots; 33.7% from stems), Fusarium pseudograminearum (14.9% from roots; 27.8% from stems), Rhizoctonia cerealis (1.7% from roots; 4.4% from stems), and Gaeumannomyces graminis var. tritici (9.8% from roots; 4.4% from stems). We observed that the recovered species varied with the agronomic zone. Fusarium pseudograminearum was predominant in regions 1 and 3, whereas F. graminearum, F. acuminatum, and R. cerealis were predominant in regions 2 and 4. The incidence of F. pseudograminearum and R. cerealis was significantly different between regions 1 and 4, while no significant association was found in the distribution of the other species and the agronomic zones. A negative correlation between the frequency of occurrence of F. pseudograminearum and mean annual precipitation during 2013-2016 (r = -0.71; P < 0.01) in the North China Plain and a positive correlation between the mean annual precipitation during 2013-2016 and the frequency of occurrence of F. asiaticum (r = 0.74; P < 0.01) were observed. Several Fusarium species were also found with low frequencies of ~2.1%-3.4 % (F. graminearum, F. acuminatum, and F. sinensis) and ~0.1%-1.3% (F. equiseti, F. oxysporum, F. proliferatum, F. culmorum, F. avenaceum, and F. asiaticum). In more than 93% of the fields, from the root and crown tissues of wheat, two or more root and crown rot species were isolated. The coexistence of Fusarium spp. and B. sorokiniana in one field (65.4%) or in individual plants (11.6%) was more common than for the other species combinations. Moreover, this is the first report on the association between F. sinensis and root and crown rot of wheat. Our results would be useful in the framing guidelines for the management of root and crown rot fungi in wheat in different agronomic zones of the North China Plain.

Project description:Fungi associated with black point were isolated from three highly susceptible wheat genotypes in the North China Plain. The 21 isolates represented 11 fungal genera. The most prevalent genera were Alternaria (isolation frequency of 56.7%), Bipolaris (16.1%), and Fusarium (6.0%). The other eight genera were Curvularia, Aspergillus, Cladosporium, Exserohilum, Epicoccum, Nigrospora, Penicillium, and Ulocladium; their isolation frequencies ranged from 0.8 to 4.8%. The pathogenicity of the isolates was individually assessed in the greenhouse by inoculating wheat plants with spore suspensions. Ten of the 21 isolates caused significantly higher incidences of black point than that the controls. These isolates belonged to eight fungal species (A. alternata, B. sorokiniana, B. crotonis, B. cynodontis, C. spicifera, F. equiseti, E. rostratum, and E. sorghinum) based on morphological traits and phylogenetic analysis. The average incidences of black point in the eight fungal species were 32.4, 54.3, 43.0, 41.9, 37.2, 38.8, 50.1, and 34.1%, respectively. B. sorokiniana and A. alternata were determined to be the most important pathogens in the North China Plain based on fungal prevalence and symptom severity. This study is the first to identify E. rostratum as a major pathogen causing black point in wheat.
