Project description:We performed a microarray experiment to analyze the transcriptional profile of human fetal tissue derived neural stem/progenitor cells, human iPSCs, or human iPSC-derived neural stem/progenitor cells generated using xenogenic or xeno-free reagents. Gene expression patterns were compared among human fetal tissue derived neural stem/progenitor cells, human iPSCs, or human iPSC-derived neural stem/progenitor cells generated using xenogenic or xeno-free reagents. Samples with a title including "Xf" (201B7-lt-NES (Xf)-1, 201B7-lt-NES (Xf)-2, 1210B2-lt-NES (Xf)-1,1210B2-lt-NES (Xf)-2, 1231A3-lt-NES (Xf)-1, 1231A3-lt-NES (Xf)-2) correspond to human iPSC-derived neural stem/progenitor cells generated using xeno-free reagents, whereas samples with a title including "Con" (201B7-lt-NES (Con)-1, 201B7-lt-NES (Con)-2) correspond to those generated using xenogenic reagents.

Extracellular matrix-dependent generation of integration- and xeno-free iPS cells using a modified mRNA transfection method

Project description:In this study, we sought to examine whether an extracellular matrix (ECM)-based xeno-free culture system that we recently established could be used together with a microRNA-enhanced mRNA reprogramming method for the generation of clinically safe iPS cells. The notable features of this method are (1) the use of a xeno-free/feeder-free culture system for the generation and expansion of iPS cells rather than the conventional labor-intensive culture systems using human feeder cells or human feeder-conditioned medium and (2) the enhancement of mRNA-mediated reprogramming via the delivery of microRNAs. Strikingly, we observed the early appearance of iPS cell colonies (~11 days), substantial reprogramming efficiency (~0.2-0.3%), and a high percentage of ESC-like colonies among the total colonies (~87.5%), indicating enhanced kinetics and reprogramming efficiency. Therefore, the combined method established in this study provides a valuable platform for the generation and expansion of clinically safe (i.e., integration- and xeno-free) iPS cells, facilitating immune-matched cell therapy in the near future.

Chemically defined, serum-free and xeno-free generation of mature type 2 alveolar epithelial cells from hiPSCs

Project description:HiPSCs were differentiated into alveolar type 2 cells (AT2s) following a stepwise differentiation protocol, mimicking lung development, using a chemically defined, serum-free, and xeno-free differentiation protocol. Samples contain biological triplicates of each developmental stage (hiPSCs, definitive endoderm, lung progenitor stage, passage 0 AT2s, early and late passage AT2s) using the same cell line. Additionally, a biological triplicate for a cell line from an idiopathic pulmonary fibrosis patient with a homozygous surfactant protein C mutation was done for late-passage AT2s comparison.

Xeno-free and Chemically Defined Human System for Culturing Human Epidermal Keratinocytes

Project description:Developing an effective xeno-free and defined system for human keratinocyte culture has been one of the fundamental measures in current cellular therapy. For keratinocytes, in particular, such system will not only fulfill clinical safety and quality criteria, it will allow for the management of less severe burns and other skin injuries such as chronic wounds. To date, the expansion of autologous keratinocytes in the clinical settings still relies on 3T3 feeder co-culture system. Here we report a completely xeno-free and defined culture system by using human recombinant laminins as feeder replacement. We have thoroughly characterized the cells cultured in this new system both in vitro and in vivo. We found that laminin-511/-521, and the unanticipated laminin-411/-421, but not -332, -111, -121, -211, or LN-221 support adult human skin keratinocytes and could maintain their self-renewal properties comparable to the 3T3 system. We believe our new culture method should facilitate broader use of cultured epithelial cell products for today’s regenerative medicine.

Xeno-Free Biomimetic-Scaffold Supports hiPSC-Derived Dental Organoid Formation and Tooth Regeneration

Project description:Tooth regeneration remains an unmet clinical challenge due to the structural complexity of dental tissues and the requirement for precise epithelial–mesenchymal interactions during development. Here, we present a xeno-free, biomimetic strategy for dental organoid engineering by integrating human induced pluripotent stem cell (hiPSC)–derived oral epithelial cells (OECs) and neural crest cells (NCCs) within a biodegradable chitosan–alginate (CA) scaffold. The CA scaffold provides a mechanically compliant and highly porous 3D microenvironment that supports hiPSC maintenance, lineage-specific differentiation, and spatial organization. Recombined OECs and NCCs self-organized into dental organoids that recapitulated key features of early odontogenesis, including coordinated epithelial–mesenchymal interactions, progressive expression of odontogenic markers (DSPP and AMBN), and time-dependent mineral deposition. Transcriptomic profiling revealed activation of gene programs associated with odontogenesis, amelogenesis, and craniofacial morphogenesis. Upon orthotopic implantation into a rat maxillary molar extraction site, cell-laden scaffolds supported cell survival, early tooth-like tissue organization, and localized mineralized matrix formation in vivo. Together, these results establish a xeno-free, scaffold-based platform for dental organoid formation and early tooth regeneration, providing a translational framework for stem cell–based dental tissue engineering and regenerative therapies.
