Project description:Hepatitis B virus (HBV) infection is one of the major causes of hepatocellular carcinoma (HCC). Nucleos(t)ide analogue (NA) therapy is effective to reduce the HCC incidence, while it doesn’t completely prevent HCC. In this study, we explored the involvement of microRNA (miRNA) in the post-NA treatment HCC development. Chronic hepatitis B (CHB) patients who received NA treatment (n = 18) were divided into 2 groups: those who didn’t develop HCC during the follow-up period (non-HCC group) and those who developed HCC after treatment (HCC group). Liver specimens were obtained before and after NA therapy, and HCC tissues were also obtained from the HCC group patients. Normal liver tissues were also obtained from 4 liver hemangioma patients who underwent surgical resection. miRNA expression was analyzed using Agilent miRNA microarray V3.

Single cell RNA sequence data from intrahepatic immune cells of CHB(chronic hepatitis B) mice

Project description:We successfully established a novel immunocompetent mouse model of CHB(chronic hepatitis B) at the immune-active phase by a simple combination of a Fumarylacetoacetate hydrolase (FAH) knockout mouse, hydrodynamic tail vein injection, and sleeping-beauty transposon system. The employment of single-cell RNA sequence in this model revealed the comprehensive intrahepatic immune landscape in the immune-active phase, which contributes to the persistence of HBV infection. Our model also made it possible to profile the intrahepatic immune response to the immunomodulators.

Nucleos(t)ide analogs for hepatitis B virus infection differentially regulate the growth factor signaling in hepatocytes

Project description:Recent clinical studies have suggested that the risk of developing hepatocellular carcinoma might be lower in chronic hepatitis B (CHB) patients receiving tenofovir disoproxil fumarate (TDF) than in patients receiving entecavir (ETV), although there is no difference in biochemical and virological remission between two drugs. The effects of nucleoside analogs (NsAs; lamivudine and ETV) or nucleotide analogs (NtAs; adefovir disoproxil [ADV], TDF, and tenofovir alafenamide [TAF]) on cell growth and the expression of growth signaling molecules were investigated. The tumor inhibitory effects of NsAs or NtAs were evaluated using a mouse xenograft model, and protein phosphorylation profiles were investigated.

The serum proteomics tracking of HBsAg low-level nucleoside analogues treatment of chronic hepatitis B combined with Pegylated Interferon therapy

Project description:Chronic hepatitis B (CHB) remains a major public health problem worldwide. Combined use of nucleoside analogues (NAs) and polyethylene glycolated interferon (PEG-IFNα) can rapidly reduce hepatitis B surface antigen (HBsAg) levels and lead to clinical cure in some advantaged populations. However, the specific molecular mechanisms by which NAs in combination with interferon therapy achieve rapid HBsAg reduction are unknown. To investigate the biomolecular differences associated with combination therapy, we analysed serum protein mass spectra from 25 patients with chronic hepatitis B who experienced a rapid reduction in HBsAg levels after combination therapy. Analyses were performed using data-independent acquisition (DIA)-based liquid chromatography-mass spectrometry before treatment, at week 12 of treatment, and at week 24 of treatment. A total of 3174 proteins were identified in our study. Among these proteins, 54 and 154 dysregulated proteins were detected at week 12 and 24 of treatment, respectively, compared with the pretreatment period.
