Project description:Genome wide DNA methylation profiling of blood samples collected from patients prior to diagnosis with hepatocellular carcinoma (HCC) vs. blood samples collected from healthy individuals without family history of cancer. The Illumina Infinium 450K Human DNA methylation Beadchip v1.2 was used to obtain DNA methylation profiles across approximately 450,000 CpGs in human samples corresponding to cases (pre-diagnostic HCC) and controls. Samples included 21 cases and 21 controls. Cases were matched with controls on gender, age, ethnicity, hepatitis C infection, and diabetes. The presence or absence of HCC in our study was determined based on the AASLD criteria.

Identification of differentially expressed microRNAs in liver tissue of Hepatitis B virus (HBV) infected hepatocellular carcinoma (HCC)

Project description:Global expression profiling of miRNAs in liver tissue of HBV infected HCC and chronic hepatitis B (CHB) with no fibrosis was evaluated. A total of 40 differentially expressed miRNAs were identified in HCC. Top 10 miRNAs are validated in more numbers of HCC tisuues by qRT PCR. Finally six miRNAs (miR-15a, miR-16, miR-21,miR-29b-3p, miR-126, miR-142-3p, miR-193a-5p) showed similar expression pattern in both microarray and qRT PCR Differential expression analysis of microRNAs in HBV infected HCC (n=4) compared to CHB patients with no fibrosis (n=8) as control.

HBV remodels PP2A complexes to rewire kinase signaling in hepatocellular carcinoma

Project description:Hepatitis B virus (HBV) infection promotes liver cancer initiation by inducing inflammation and cellular stress, but its impact in established tumors is not well understood. We used affinity purification mass spectrometry to comprehensively map a network of 145 physical interactions between HBV and human proteins in hepatocellular carcinoma (HCC). A subset of the host factors targeted by HBV proteins are preferentially mutated in non-HB¬V-associated HCC suggesting that their interaction with HBV influences HCC biology. These include proteins involved in mRNA splicing, mitogenic signaling and DNA repair, with the latter set interacting with the HBV oncoprotein X (HBx). We show that HBx remodels the PP2A phosphatase complex by excluding striatin regulatory subunits from the PP2A holoenzyme. We find that HBx effects on PP2A cause Hippo kinase activation, particularly in cells with high striatin levels. In parallel, HBx activates mTOR complex 2 (mTORC2) to prevent YAP degradation. mTORC2 effects on YAP can be observed in human HCC specimens and mouse HCC models and can be targeted with mTOR kinase inhibitors. Thus, HBV interaction with host proteins rewires HCC signaling rather than directly activating mitogenic pathways. These findings provide a new paradigm for the cellular effects of a tumor promoting virus and support a model where HBV may have therapeutic actionable effects on HCC biology.

Engineered Oncolytic VSV Enhances Tumor Killing in HBV-Positive HCC

Project description:Hepatocellular carcinoma (HCC) is a leading cause of cancer-related death worldwide, with chronic hepatitis B virus (HBV) infection being a major contributor to its development. Despite this, effective treatment for HBV-associated HCC remains a significant challenge. In this study, we combined the strengths of vesicular stomatitis virus (VSV) and therapeutic vaccines to propose a novel strategy for treating HBV-related liver cancer. We engineered an oncolytic virus that delivers hepatitis B surface antigen (HBsAg) and the IL-15 cytokine directly to tumor cells, thereby enhancing immune activation and promoting tumor cell destruction. As the tumor cells die, they release additional virus particles, spreading HBsAg and IL-15 expression to neighboring tumor cells. This approach demonstrated promising efficacy in multiple HBV-positive HCC mouse models.

Viral-Track integrated single-cell RNA-sequencing reveals HBV lymphotropism and immunosuppressive microenvironment in HBV-associated hepatocellular carcinoma

Project description:The tumor microenvironment (TME) is a crucial mediator of tumor progression and treatment response. Here, we compare the immune microenvironments of HBV and non-HBV-HCC and investigate the reason for the persistence of HBV infection in the liver. We combine the Viral-Track method with single-cell RNA sequencing and profile the transcriptomes of 71,466 cells from HBV and non-HBV-HCC patients. In addition to hepatocytes and macrophages, HBV transcripts were also detected in T and B cells using the Viral-Track method, confirming the HBV-lymphotropic nature for the first time in scRNA-sequencing data. HBV-HCC tumors have reduced levels of CD4+ Tregs, NK, macrophages, dendritic cells (DCs), and increased malignant hepatocytes compared with non-HBV tumors. Notably, we explicitly reported the enrichment of metallothioneins (MTs), particularly MT1G expression in HBV-related HCC TAMs, which was associated with a worse prognosis. HBV-tumor-infiltrated CD8+ T cells exhibited a cytotoxic dysfunctional T cell phenotype, characterized by upregulated MDK and CTLA4 expression and reduced IFN-γ production, unlike the non-HBV-HCC. Additionally, HBV-HCC exhibited immunosuppressive ligand-receptor interactions, whereas non-HBV-HCC exhibited antitumor ligand-receptor interactions. Our comprehensive understanding of the HBV HCC ecosystem by viral-track integrated scRNA sequencing analysis provides deeper insights into immune evasion mechanisms and HBV lymphotropism associated with viral persistence.
