Project description:IFNg is a pro-inflammatory and pro-atherogenic cytokine that leads to macrophage activation. Adenosine has well-documented anti-inflammatory properties. We used microarrays to compare the global gene expression profile in mouse macrophages stimulated with IFNg alone and those cells treated with IFNg and adenosine. We determined that adenosine suppressed the expression of many IFNg-regulated pro-inflammatory cytokines, chemokines, and other pro-atherogenic genes. Keywords: treatment response RAW 264.7 cells were treated for 4 hours with either IFNg or IFNg plus adenosine. Following treatment, total RNA was extracted and treatment groups were pooled from 2 separate experiments for hybridization of Affymetrix microarrays.

Skewed macrophage polarization in aging skeletal muscle

Project description:Skeletal muscle aging is a major causative factor for disability and frailty in the elderly. Recent theories about the origins of the progressive impairment of skeletal muscle with aging emphasize a disequilibrium between damage and repair. Macrophages participate in muscle tissue repair first as pro-inflammatory M1 subtype and then as M2 anti-inflammatory subtype. However, information on macrophage presence in skeletal muscle is still sporadic and the effect of aging on different phenotypes remains unknown. In this study, we sought to characterize the polarization status of macrophages human skeletal muscle at different ages. We found that most macrophages in human skeletal muscle are M2, and that this number increased with advancing age. On the contrary, M1 macrophages decline with aging, making the total number of macrophages invariant with older age. Notably, M2 macrophages co-localized with increasing intermuscular adipose tissue (IMAT) in aging skeletal muscle. The mouse strain BALB/c, intrinsically M2-prone, showed increased IMAT and regenerating myofibers in skeletal muscle, accompanied by elevated expression of adipocyte markers and M2 cytokines. Collectively, we report that polarization of macrophages to the major M2 subtype is associated with IMAT, and propose that increased M2 in aged skeletal muscle may reflect active repair of aging-associated muscle damage.

Viral miRNA-mediated impact was assessed on the host cellular microRNA profiles by miRNA microarray analysis. Five different viral miRNA representing 4 different human herpesviruses were overexpressed in M1 and M2 macrophages and the changes in cellular miRNA compared to control mimic were examined. Viral miRNA exhibit unique impact on M1 and M2 miRNA profiles.

Effects of TRIM59 on Raw 264.7 macrophage gene expression

Project description:Macrophages have many functions, such as acting as phagocytic pathogens, antigen presentation and playing a key role in immune response. Tripartite motif containing 59 (TRIM59) protein is highly expressed in a variety of tumor cells and regulates the proliferation of tumor cells as well as their innate immune response. Recent studies have shown that the expression of TRIM59 was differentially expressed under different stimuli of activated macrophages, however, the effects of TRIM59 on macrophage gene expression profiles are still unknown. In our study, we constructed RAW 264.7 macrophages with high and low expression of TRIM59, and used transcriptome sequencing to identify the effects of TRIM59 on macrophage gene expression profiles. Results showed that TRIM59 affected an abundant number of genes, and may affect phagocytosis and cell cycles.

RNA-seq analysis identifies regulators of macrophage reprogramming from M2 to M1-like phenotype

Project description:We analyzed expression profiles of thioglyolate elicted peritoneal exudate cells (peritoneal macrophages). Peritoneal macrophages were polarized into M1 and M2 macrophages by activation with IFNgamma+ LPS and Il4, respectively. We predicted a gene-regulatory network, which consists of four transcription factors (E2f1,Myc, Stat6, Pparg) regulating metabolic genes, M1 and M2-associated genes. The predicted regulators were all active in M2 macrophages. We hypothesized that these transcription factors are essential regulators to maintain M2 phenotype. To further validate our findings, we treated M2-polarized macrophages with siRNA-pools targeting E2f1, Myc, Stat6 and Pparg. In this study, we observed a switch towards an M1-like phenotype after transfection of siRNA-pools. In addition, Inflammatory pathways were upreguated while fatty acid metabolism was down regulated.
