Project description:Osteoarthritis (OA) is characterized by progressive, irreversible erosion of articular cartilage accompanied with severe pain and immobility. This study aimed to assess the effect and mechanism of action of HU308- a selective cannabinoid receptor type 2 (CB2) agonist, in potentially preventing OA-related joint damage. To test this assumption, Cn2r null mice and WT mice were aged to reach 20 months, and analyzed for joint structural features. OA was induced in Wild type (WT) mice via ageing or post-traumatic procedure, followed by HU308 systemic (intraperitoneal) or local (intra-articular) administration, respectively. Additional analysis of time and dose courses for HU308 were carried out in human primary chondrocytes, analyzed by RNA-sequencing, RT-PCR, ChIP and immunoblotting. Our results show that Cnr2r null mice exhibited enhanced age-related OA severity and synovitis compared to age-matched WT mice. Systemic administration of HU308 to 16-month mice improved pain sensitivity and maintained joint integrity, which was consistent with the intra-articular administration of HU308 in post-traumatic OA mice. When assessing human chondrocytes treated with HU308, we uncovered a dose- and time related increase in ACAN and COL2A1 expression, which was preceded by increased SOX9 expression due to pCREB transcriptional activation of the SOX9 gene. Finally, transcriptomic analysis of patient-derived human chondrocytes, identified HU308 populations exhibiting enhanced SOX9 expression, accompanied by enriched gene networks related to carbohydrate metabolism. Collectively, the results show that HU308 reduced trauma and age-induced OA, via CB2-pCREB dependent activation of SOX9, contributing to augmented genes networks related to metabolism.

Project description:E3 ligase BRUTUS (BTS), a putative iron sensor, is expressed in both root and shoot tissues in seedlings of Arabidopsis thaliana. The role of BTS in root tissues has been well established. However, its role in shoot tissues has been scarcely studied. Comparative transcriptome analysis with shoot and root tissues revealed that BTS is involved in regulating energy metabolism by modulating expression of mitochondrial and chloroplast genes in shoot tissues. Moreover, in shoot tissues of bts-1 plants, levels of ADP and ATP and the ratio of ADP/ATP were greatly increased with a concomitant decrease in levels of soluble sugar and starch. The decreased starch level inbts-1 shoot tissues was restored to the level of shoot tissues of wild-type plants upon vanadate treatment. Through this study, we expand the role of BTS to regulation of energy metabolism in the shoot in addition to its role of iron deficiency response in roots.

Project description:Regulation between the fed and fasted state in mammals is partially controlled by peroxisome proliferator activated receptor-alpha (PPAR-alpha). Expression of the receptor is high in liver, heart and skeletal muscle, but decreases with age. A combined 1H NMR spectroscopy and GC-MS metabolomic approach has been used to examine metabolism in liver, heart, skeletal muscle and adipose tissue in PPAR-alpha null mice and wild type controls during ageing between 3-13 months. For the PPAR-alpha-null mouse multivariate statistics highlighted hepatic steatosis, reductions in the concentrations of glucose and glycogen in both liver and muscle tissue, and profound changes in lipid metabolism in each tissue, reflecting known expression targets of the PPAR-alpha receptor. Hepatic glycogen and glucose also decreased with age for both genotypes. These findings indicate the development of age related hepatic steatosis in the PPAR-alpha-null mouse, with the normal metabolic changes associated with ageing exacerbating changes associated with genotype. Furthermore, the combined metabolomic and multivariate statistics approach provides a robust method for examining the interaction between age and genotype. </p> The GC-MS assay for this study can be found in the MetaboLights study MTBLS314.

Project description:Perfluorooctane sulfonate (PFOS) is a perfluoroalkyl acid (PFAA) and a persistent environmental contaminant found in the tissues of humans and wildlife. Although blood levels of PFOS have begun to decline, health concerns remain because of the long half-life of PFOS in humans. Like other PFAAs, such as perfluorooctanoic acid (PFOA), PFOS is an activator of peroxisome proliferator-activated receptor-alpha (PPARα) and exhibits hepatocarcinogenic potential in rodents. PFOS is also a developmental toxicant in rodents where, unlike PFOA, it’s mode of action is independent of PPARα. Wild-type (WT) and PPARα-null (Null) mice were dosed with 0, 3, or 10 mg/kg/day PFOS for 7 days. Animals were euthanized, livers weighed, and liver samples collected for histology and preparation of total RNA. Gene profiling was conducted using Affymetrix 430_2 microarrays. In WT mice, PFOS induced changes that were characteristic of PPARα transactivation including regulation of genes associated with lipid metabolism, peroxisome biogenesis, proteasome activation, and inflammation. PPARα-independent changes were indicated in both WT and Null mice by altered expression of genes related to lipid metabolism, inflammation, and xenobiotic metabolism. Such results are similar to prior studies done with PFOA and are consistent with modest activation of the constitutive androstane receptor (CAR) and possibly PPARγ and/or PPARβ/δ. Unique treatment-related effects were also found in Null mice including altered expression of genes associated with ribosome biogenesis, oxidative phosphorylation and cholesterol biosynthesis. Of interest was up-regulation of Cyp7a1, a gene which is under the control of various transcription regulators. Hence, in addition to its ability to modestly activate PPARα, PFOS induces a variety of “off-target” effects as well.

Our previous temporal microarray analysis of ESC commitment identified the E3 Ubiquitin Ligase Protein Makorin-1 (MKRN1) as a potential novel component of the ESC GRN. Here, using multilayered systems-level analyses we compiled a MKRN1-centered interactome in undifferentiated ESCs at the proteomic and ribonomic level. Proteomic analyses revealed that MKRN1 is a novel RNA-binding protein that exists within messenger ribonucleoprotein (mRNP) complexes in undifferentiated ESC populations. In accordance with its presence in mRNPs, MKRN1 is mobilized to stress granules (SG) upon arsenite-induced stress, yet MKRN1 is not required for SG formation. RIP-chip analysis revealed that MKRN1 associates with mRNAs encoding functionally related regulatory proteins involved in diverse processes such as cell differentiation, apoptosis, or secreted proteins. Thus, our unbiased systems level analyses supports a role for MKRN1 as a novel RNA-binding protein and a potential gene regulatory protein within the ESC GRN.

Project description:We used gene expression microarray to explore the correlation between gene expression profiles and find out the target genes related to lipid and glucose metabolism in NAFLD. A rat model of NAFLD was established by feeding male rats with high-fat diet and Agilent DNA microarray was used to detect gene expression profiles of liver tissues. We explored differentially expressed genes and intersection of them by using Gene ontology (GO) and Pathway analyses. Then, target genes were verified by Real-time polymerase chain reaction (RT-PCR). There were 932 genes, including 783 up-regulated and 149 down-regulated, differed in expression compared to those of control rats. The up-regulated genes were involved in biosynthesis, cell development, cell differentiation and down-regulated genes mainly contributed to biological metabolic process and stress reaction. We identified genes involved in insulin signaling pathway, Notch signaling pathway, adipocytokin and linoleic acid metabolism to be closely related to liver fat content and insulin resistance. Among them, genes which supposed to connect glucose and lipid metabolism were screened (IGFBP7, Notch1, HMGCR and ACACB) and then validated by real-time PCR. These data provide innovative information on the whole genome expression profile due to high-fat diet feeding and bring important insights into the mechanism of NAFLD.
