Comparative Proteomic Analysis of the Telogen-to-Anagen Transition in Cashmere Goat Secondary Hair Follicles

Project description:This study investigates the proteomic changes associated with the telogen-to-anagen transition in secondary hair follicles (SHFs) of cashmere goats. Using tandem mass tag (TMT) labeling combined with LC-MS/MS, we identified 3,654 proteins, with 458 differentially expressed proteins (DEPs) between telogen and anagen phases. Key proteins such as ADAM17, SFRP1, and PPP1CA were found to play crucial roles in regulating SHF cycle transitions. These findings provide insights into the molecular mechanisms underlying cashmere fiber growth and offer potential targets for improving cashmere production.

The function of DP cells during the development of hair follicles in cashmere goats

Project description:OBJECTIVE: To study the function of DP cells during the development of hair follicles in cashmere goats, we used single-cell sequencing to perform transcriptome sequencing of skin samples from different periods. Method: Single cell barcoding and library preparation were performed based on 10x Genomics single-cell RNA sequencing platform, CellRanger (v2.2.0) software was used for analyzing raw sequencing data, the quality control (QC) and cell clustering were analyzed with single- Cell RNA seq Seurat software (v2.3.0) based on R environment, Monocle (v 2.4.0) was used to order single cells along pseudotime according, Metascape (http://metascape.org/gp/index.html#/main /step1) was used to perform gene ontology (GO) analysis Conclusion: we show the function of four different intermediate DP cells: Intermediate cells 10 show important functions in the growth of villi and maintenance of villus attachment to the skin, and intermediate cell 1 shows important functions in the process of apoptosis and villous shedding of hair Follicles, intermediate cell 0 initiates a new follicular cycle, completes the migration of hair follicles and the occurrence of villi, and intermediate cell 15 maybe DP progenitor cells, In addition, we speculated the apoptotic order of different bulge cells. More importantly, we provided a molecular landscape for the mechanism of DP cells in the process of hair follicle cycle initiation. Skin samples from telogen, anagen, and catagen were used to sequence single-cell transcriptomes, revealing the function and evolution of DP cells.

Generation of trichogenic adipose-derived stem cells by expression of three factors

Project description:Background: Previous studies demonstrated that adipose-derived stem cells (ASCs) can promote hair growth, but unmet needs exist for enhancing ASC hair inductivity. Objective: Therefore, we introduced three trichogenic factors platelet-derived growth factor-A, SOX2, and β-catenin to ASCs (tfASCs) and evaluated whether tfASCs have similar characteristics as dermal papilla (DP) cells. Method: Global gene expression was examined using NGS analysis. Telogen-to-anagen induction, vibrissae hair follicle organ culture and patch assay were used. Results: tfASC cell size is smaller than that of ASCs, and they exhibit short doubling time. tfASCs also resist aging and can be expanded until passage 12. Cell proportion in S and G2/M increases in tfASCs, and tfASCs express high mRNA levels of cell cycle related genes. The mRNA expression of DP markers was notably higher in tfASCs. Moreover, NGS analysis revealed that the global gene expression of tfASCs is similar to that of DP cells. The injection of tfASCs accelerated the telogen-to-anagen transition and conditioned medium of tfASCs increased the anagen phase of vibrissal hair follicles. Finally, we found that the injection of 3D-cultured tfASCs at p 9 generated new hair follicles in nude mice. Conclusion: Collectively, these results indicate that 1) tfASCs have similar characteristics as DP cells, 2) tfASCs have enhanced hair-regenerative potential compared with ASCs, and 3) tfASCs even at late passage can make new hair follicles in a hair reconstitution assay. Because DP cells are difficult to isolate/expand and ASCs have low hair inductivity, tfASCs and tfASC-CM are clinically good candidates for hair regeneration.

Bmal1 controls circadian cell proliferation and susceptibility to UVB-induced DNA damage in the epidermis [Anagen]

Project description:While several physiological skin parameters vary in a circadian manner, the identity of genes participating in chronobiology of skin remains unknown, leading us to define the circadian transcriptome of mouse skin at two different stages of the hair cycle, telogen and anagen. The circadian transcriptomes of telogen and anagen skin are largely distinct, with the former dominated by genes involved in cell proliferation and metabolism. The expression of many metabolic genes is antiphasic to cell cycle related genes, the former peaking during the day and the latter peaking at the night. Consistently, accumulation of reactive oxygen species, a byproduct of oxidative phosphorylation, and S-phase are antiphasic to each other in telogen skin. Furthermore, the circadian variation in S-phase is controlled by BMAL1 intrinsic to keratinocytes as keratinocyte-specific deletion of Bmal1 obliterates time of day dependent synchronicity of cell division in the epidermis leading to a constitutively elevated cell proliferation. Consistent with higher cellular susceptibility to UV-induced DNA damage during S phase, we found that mice are most sensitive to UVB-induced DNA damage in the epidermis at night. As maximum numbers of keratinocytes go through S phase in the late afternoon in the human epidermis, we speculate that in humans the circadian clock imposes regulation of epidermal cell proliferation such that skin is at a particularly vulnerable stage during times of maximum UV exposure, thus contributing to the high incidence of human skin cancers.

Paracrine TGFß Signaling Counterbalances BMP-Mediated Repression in Hair Follicle Stem Cell Activation

Project description:Hair follicle (HF) regeneration begins when communication between quiescent epithelial stem cells (SCs) and underlying mesenchymal dermal papillae (DP) generates sufficient activating cues to overcome repressive BMP signals from surrounding niche cells. We uncovered a hitherto unrecognized DP transmitter, TGFβ2, which activates Smad2/3 transiently in HFSCs concomitant with entry into tissue regeneration. We used microarrays to detect the genes specifically affected by TGFß receptor II-deficient mice upon HFSC activation. Hair follicle stem cells (HFSCs) of hair gem (HG) and bulge, and total skin keratinocytes were FACS-purified from the mouse back skin at 2nd telogen-to-anagen transition stages.

Transcriptome analysis of cells in mouse hairy melanocytic nevi

Project description:RNA-seq data was obtained from different skin cell types at two time points: postnatal day P30, when WT dorsal hair follicles are in actively growing state (aka anagen) and P56, when WT hair follicles are in resting state (aka telogen). Cell types include: Bulge stem cells (B), dermal papilla fibroblasts (DP), secondary hair germ cells (HG), melanocytes (Melan), myeloid cells (Myel) and CD45-positive immune cells (I). All cell were FACS sorted and only live single cells were collected for RNA extraction.
