Project description:Relapse of cancer is associated with multi-directional differentiation, and unrestricted proliferative replication potential of cancer stem cells. Herein, we propose the plastic differentiation strategy for irreversible differentiation of cancer stem cells. Whole gene expression analysis revealed that salinomycin induced the cancer stem cells into normal cells, dormant cells, and mature cancer cells. Besides, the results indicated that the gatekeeper was related to the inhibition of PKC α signaling pathway. The differentiated normal or dormant cells were incorporated into normal tissue, while the rest were killed by chemotherapy. Our findings would offer the evidence for plastic differentiation of cancer stem cells, and propose an invert strategy for cancer therapy.

Project description:Relapse of cancer is associated with multidirectional differentiation and unrestricted proliferative replication potential of cancer stem cells. Herein, we propose the plastic differentiation strategy for irreversible differentiation of cancer stem cells; further, salinomycin and its newly constructed functional liposomes are used to implement this strategy. Whole gene, cancer stem cell-related RNA, and protein expression analyses reveal that salinomycin induces the cancer stem cells into normal cells, dormant cells, and mature cancer cells. Besides, the results indicate that the gatekeeper is related to the inhibition of the protein kinase C (PKC) α signaling pathway. The differentiated normal or dormant cells are incorporated into normal tissue, whereas the rest are killed by chemotherapy. The findings would offer the evidence for plastic differentiation of cancer stem cells and propose a novel strategy for cancer therapy.

Project description:Cellular plasticity is a hallmark of rare Claudin-low (CL) and metaplastic (MBC) breast cancer subtypes, with a documented overlap whose exact extent is yet unknown, and which are associated to resistance and poor survival. We used spatial transcriptomics to further characterise these plastic subtypes, respectively defined molecularly and histopathologically. We identified 3 putative CL tumours (CL-like) and 4 genomically unstable TNBC samples via molecular analyses, combined with 4 MBCs identified by a breast pathologist.

¬¬¬Seabirds in crisis: plastic ingestion induces proteomic signatures of multi-organ failure and neurodegeneration

Project description:Understanding plastics’ harmful impacts has been hampered by the application of overly simplistic tools, mostly focusing on a single response variable which limits our ability to detect changes in wildlife health. Adopting a data-driven, proteomics approach, we assessed changes in 745 proteins in a non-model organism with differing levels of plastic exposure. In high plastic impacted seabirds, we observed a range of negative health consequences: intracellular components that should not be found in the blood were frequently detected, indicative of cell lysis. There was significant evidence of neurodegeneration, and secreted proteins were less abundant, indicating the kidney and particularly the liver are not functioning as normal. The proteomic signatures reflect the effects of plastic distal to the site of exposure (i.e., the stomach) demonstrating plastic shedding and leachate toxicity in the plasma. Notably, metrics commonly used to assess body condition do not provide an accurate description of health or plastic impaction.
