Project description:Macrophages possess the hallmark feature of plasticity, allowing them to undergo a dynamic transition between M(LPS+IFN-γ) and M(IL-4) polarized phenotypes. The aim of the present study was to screen for differentially lncRNAs that were associated with macrophage polarization. The lncRNA profiles of three M(LPS+IFN-γ) and three M(IL-4) samples were obtained using microarray analysis. Based on the threshold of fold-change >2.0 and a p-value < 0.05, we screened a total of 1,251 lncRNAs, of which 627 were upregulated and 624 downregulated in M(LPS+IFN-γ) macrophages compared to that in M(IL-4) macrophages.This study provides new insights into the role of genes in macrophage differentiation and polarization.

Gene expression profiles from murine BALB/c macrophages at the extremes of the M1 and M2 polarized phenotypes

Project description:Macrophages possess the hallmark feature of plasticity, allowing them to undergo a dynamic transition between M1 and M2 polarized phenotypes. The aim of the present study was to screen for differentially expressed genes (DEGs) that were associated with macrophage polarization. The transcription profiles of three M1 and three M2 samples were obtained using microarray analysis. Based on the threshold of fold-change >2.0 and a p-value < 0.05, we screened a total of 1,253 DEGs, of which 696 were upregulated and 557 downregulated in M1 macrophages compared to that in M2 macrophages. Gene Ontology (GO) function and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were also performed. A gene-gene interaction network of the DEGs was constructed using the STRING database. GO annotation identified three categories, which included cellular component (CC), molecular function (MF), and biological process (BP), with 34 and 40 enrichment terms consisting of upregulated and downregulated DEGs, respectively. GO enrichment analysis of DEGs was mainly associated with protein binding, response to stimulus, cell differentiation, and regulation of biological process. KEGG enrichment identified 15 and four pathways involving upregulated and downregulated DEGs, respectively. Signaling pathway analysis showed that these DEGs were mainly involved in apoptosis, HIF-1 signaling pathway, innate immune system, TNF signaling pathway, cytokine-cytokine receptor interaction, and other signal transduction pathways. Interaction network analysis indicated that Tnf, Il6, Il1b, Socs3, Nos2, Hif1a, and other genes may play key roles in macrophage polarization. This study provides new insights into the role of genes in macrophage differentiation and polarization.

The multifaceted responses of human monocytes to IFN-gamma and IL-4 are not reducible to M1/M2 polarization of Macrophages

Project description:Macrophages are known to be polarized into inflammatory (M1) and immunoregulatory (M2) cells when they are stimulated by agonists such as IFN-gamma and IL-4, respectively. If circulating monocytes may be polarized in response to T cell signals is often misguidedly deduced from macrophage results. Here the transcriptional responses of human CD14+ monocytes to IFN-gamma and IL-4 were analyzed using whole genome microarrays. A principal component analysis and hierarchical clustering showed that monocyte and macrophage responses were distinct. Monocytes stimulated with IFN-gamma and IL-4 for 6 hours exhibited some features of macrophage polarization. Indeed, when 80 genes considered as M1 and M2 genes were analyzed, we found that M1 genes were modulated in response to IFN-gamma and that M2 genes were modulated in response to IL-4. The M1 polarization of monocytes was transient because only M2 genes were modulated when monocytes were stimulated with IFN-gamma and IL-4 for 18 hours. However, the activation of monocytes by IFN-gamma and IL-4 could not be reduced to M1/M2 polarization status. Indeed, monocytes exhibited early specific signatures composed of 46 and 39 up-regulated genes in response to IFN-gamma and IL-4, respectively, and a late signature common to both molecules that consisted of 57 up-regulated genes. Taken together, these results demonstrated the extreme plasticity of human monocytes and suggested the existence of a core transcriptional termination program. Using early and late signatures might be pertinent to investigate monocyte activation in inflammatory or infectious diseases. Monocytes were stimulated with IFN-gamma (20ng/mL) or IL-4 (20ng/mL) for 6 and 18 hours or culture for 6 and 18 hours without agonist (Unstimulated samples). Monocytes-derived-macrophages (MDM) stimulated with IFN-gamma and IL-4 for 18 hours were used as controls. Each microarray is derived from a single biological sample.
