Reduced representation bisulfite sequencing (RRBS) methylation profiles of placenta and umbilical cord blood with or without gestational diabetes mellitus (GDM)

Project description:Objective: To explore the characteristics and underlying molecular mechanisms of genome-scale expression profiles of women with- or without- gestational diabetes mellitus and their offspring. Materials and Methods: We recruited a group of 21 pregnant women with gestational diabetes mellitus (GDM) and 20 healthy pregnant women as controls. For each pregnant women, RRBS were performed using the placenta and paired neonatal umbilical cord blood specimens. Differentially methylated regions (DMRs) were identified. Then, functional enrichment analysis was performed to differential methylated genes (DMGs) separately or interactively in placenta and umbilical cord blood. Results: Through the comparison of GDM and healthy samples, 2779 and 141 DMRs were identified from placenta and umbilical cord blood, respectively. Functional enrichment analysis showed that the placenta methylation and expression profiles of GDM women mirrored the molecular characteristics of “type II diabetes” and “insulin resistance”. Methylation-altered genes in umbilical cord blood were associated with pathways “type II diabetes” and “cholesterol metabolism”. DMGs illustrated significant overlaps among placenta and umbilical cord blood samples, and the overlapping DMGs were associated with cholesterol metabolism. Conclusions: Our research demonstrated the epigenomic alternations of GDM mothers and offspring. Our findings emphasized the importance of epigenetic modifications in the communication between pregnant women with GDM and offspring, and provided reference for the prevention, control, treatment, and intervention of perinatal deleterious events of GDM and neonatal complications.

Panoramic Elucidation of the Molecular Mechanisms Underpinning the Epigenetic Impact of Gestational Diabetes Mellitus on Offspring

Project description:Background: Gestational diabetes mellitus (GDM), a common pregnancy complication with profound effects on maternal and offspring health, involves epigenetic modifications, particularly DNA methylation, but the molecular mechanisms elevating metabolic disease risk in offspring remain elusive. Methods: Integrating public data with whole-genome methylation data from umbilical cord blood of GDM fetuses as the validation cohort, this study characterizes GDM-associated epigenetic features and develops a predictive model validated across external datasets to assess potential long-term health risks in offspring. Results:We identified consistent patterns of differential methylation in the offspring of GDM, with key DMPs associated with glucose homeostasis and insulin sensitivity genes. Pathway analysis revealed enrichment in insulin signaling, AMPK activation, and adipocytokine signaling pathways. Our predictive model demonstrated robust performance across different sample sizes (AUC=0.89 in public data, AUC=0.82 in validation cohort). CpG sites associated with PPARG and INS genes showed persistent predictive significance across datasets. Conclusions:This study provides a comprehensive characterization of the epigenetic landscape in the offspring of GDM, revealing consistent methylation patterns and pathway enrichments across diverse datasets. The developed predictive model holds potential for assessing health risks of GDM in offspring, advancing understanding of GDM pathophysiology and offering epigenetic biomarkers for future diagnostic, therapeutic, and preventive strategies.

Placental Remodeling in Gestational Diabetes Mellitus (GDM) Disrupts Lipid Metabolism

Project description:Gestational diabetes mellitus (GDM) is a pregnancy-specific metabolic disorder associated with adverse maternal and fetal outcomes, with epigenetic dysregulation increasingly implicated in fetal programming. As the placenta plays a central role in maternal–fetal nutrient and hormonal exchange, this study investigated structural, epigenetic, and metabolic alterations in placentas from GDM pregnancies. Pregnant women recruited from PRS Hospital Pvt. Ltd. were classified as normoglycemic and GDM based on oral glucose tolerance test results, and maternal blood, cord blood, and placental tissues were collected for histological evaluation, DNA methylation profiling, lipid analysis, and gene expression studies. GDM placentas exhibited pronounced morphological abnormalities, including increased syncytial knots, chorangiosis, villous immaturity, and fibrinoid necrosis. Epigenomic analysis identified 521 differentially methylated regions, with significant enrichment in lipid metabolism pathways; notably, the lipid-associated gene PLIN1 was hypermethylated and significantly downregulated in GDM placentas. Concordantly, lipid profiling of maternal and cord blood revealed marked dyslipidemia in GDM subjects, indicating disrupted lipid homeostasis at both molecular and systemic levels. Collectively, these findings demonstrate that GDM induces coordinated structural, epigenetic, and metabolic alterations in the placenta that may influence fetal lipid exposure and contribute to long-term metabolic risk, highlighting PLIN1 as a potential biomarker and therapeutic target.

Gestational diabetes adversely affects pancreatic islet development and function in the rat offspring

Project description:Gestational diabetes mellitus (GDM) exposure and obesity are strong risk factors for type 2 diabetes development; however, the connections between GDM exposure, postnatal diet and islet dysfunction in offspring metabolic health remain unclear. Reduced glucose-stimulated insulin secretion was observed in islets isolated from 15-week-old offspring prenatally exposed to GDM, which was exacerbated by postnatal HFS consumption. In the HFS-fed offspring of Lean dams, islet size and number increased, an adaptation that was not observed in the HFS-fed offspring of GDM dams. Islets from GDM exposed offspring revealed upregulation of 102 genes (e.g. Sod2, Il1b) and 13 downregulated genes. In the GDM offspring that consumed a postnatal HFS-diet, 126 genes were significantly upregulated (e.g. Ccl2, Rps14, Atp5f1) and 17 genes were downregulated (e.g. Hnf1a, Slc2a2). These results demonstrate that GDM exposure induced changes in gene expression in the young adult rat offspring that worsen islet function and whole-body glucose homeostasis.

Project description:Gestational diabetes (GDM) refers to diabetes diagnosed as diabetes in the second or third trimester of pregnancy, but not obvious before pregnancy. It is one of the most common complications of pregnancy, which can lead to perinatal asphyxia, neonatal hypoglycemia, macrosomia, cardiovascular abnormalities, obesity and other short-term and long-term adverse outcomes of the offspring. This study explores the differential expression of transcriptome between GDM and non GDM placenta, and explores the impact of subtle metabolic changes in the placenta on fetal growth and development during GDM in pregnant women.

Gestational diabetes mellitus alters placental structure, efficiency, and plasticity

Project description:The hemochorial placenta provides a critical barrier at the maternal-fetal interface to modulate maternal immune tolerance and enable gas and nutrient exchange between mother and conceptus. Pregnancy outcomes are adversely affected by gestational diabetes mellitus (GDM); however, the effects of GDM on placental formation, and subsequently fetal development, are not fully understood. In this report, streptozotocin was used to induce hyperglycemia in pregnant rats for the purpose of investigating the impact of GDM on placental formation and fetal development. GDM caused placentomegaly and placenta malformation, decreasing placental efficiency and fetal size. Elevated glucose disrupted rat trophoblast stem (TS) cell differentiation in vitro. Evidence of altered trophoblast differentiation was also observed in vivo, as hyperglycemia affected the junctional zone transcriptome and interfered with intrauterine trophoblast invasion and uterine spiral artery remodeling. When exposed to hypoxia, rats with GDM showed decreased proliferation and ectoplacental cone development on gestation day (gd) 9.5 and complete pregnancy loss by gd 13.5. Furthermore, elevated glucose concentrations inhibited TS cell responses to hypoxia in vitro. Overall, these results indicate that alterations in placental development, efficiency, and plasticity could contribute to the suboptimal fetal outcomes in offspring from pregnancies complicated by GDM.
