Project description:The study identifies cell type specific differentially expressed genes in peripheral blood mononuclear cells affected by THC infusion in two human subjects. Individual gene expression gene analysis and gene set enrichment analysis show that THC affects gene expression in the functional domains of immune response and cell proliferation. One of THC receptor genes, CNR2, is highly expressed in B cells and THC perbute CNR2 co-expressed genes in B cells. The findings provide new insight of acute THC immunotonary effects.

Single-cell Transcriptome Mapping Identifies Common and Cell-type Specific Genes Affected by Acute Delta9-tetrahydrocannabinol in Humans.

Project description:Delta-9-tetrahydrocannabinol (THC) is known to modulate immune response in peripheral blood cells. The mechanisms of THC's effects on gene expression in human immune cells remains poorly understood. Combining a within-subject design with single cell transcriptome mapping, we report that THC acutely alters gene expression in 15,973 blood cells. We identified 294 transcriptome-wide significant genes among eight cell types including 69 common genes and 225 cell-type-specific genes affected by THC administration, including those genes involving in immune response, cytokine production, cell proliferation and apoptosis. We revealed distinct transcriptomic sub-clusters affected by THC in major immune cell types where THC perturbed cell-type-specific intracellular gene expression correlations. Gene set enrichment analysis further supports the findings of THC's common and cell-type-specific effects on immune response and cell toxicity. This comprehensive single-cell transcriptomic profiling provides important insights into THC's acute effects on immune function that may have important medical implications.

Adverse effects of gestational Δ9-tetrahydrocannabinol exposure on the embryonic germline transcriptome and metabolome

Project description:An increasing number of women report cannabis use during their pregnancy, with little knowledge on the repercussions of such an exposure onto early embryonic processes. In particular, exposure to the most abundant phytocannabinoid, Δ9-tetrahydrocannabinol (Δ9-THC), could alter the development of embryonic germ cells, with potential consequences across generations. Here, we extensively characterized the impact of gestational exposure to Δ9-THC onto the developmental trajectory of an in vitro model for primordial germ cells. Our data reveals that Δ9-THC exposure increases glycolytic rates in embryonic stem cells, with consequences on their proliferation. We further show that, in the absence of continuous exposure, Δ9-THC has long-lasting adverse consequences on the metabolome and transcriptome of embryonic germ cells. These results constitute the first characterization of the impact of gestational Δ9-THC exposure onto the embryonic germline.

Proteomic Profile of Circulating Extracellular Vesicles in the Brain after Δ 9-Tetrahydrocannabinol Inhalation

Project description:Given the increasing use patterns of cannabis in the US, there is an urgent need to better understand the drug’s effects on central signaling mechanisms. Extracellular vesicles (EVs) have been identified as intercellular signaling mediators that contain a variety of cargo, including proteins. Here, we examined whether the main psychoactive component in cannabis, Δ9-tetrahydrocannabinol (THC), alters EV protein signaling dynamics in the brain. We first conducted in vitro studies which found that THC activates signaling in choroid plexus epithelial cells, resulting in transcriptional upregulation of the cannabinoid 1 receptor and immediate early gene c-fos, in addition to the release of EVs containing RNA cargo. Next, male and female rats were examined for the effects of either acute or chronic exposure to vaped THC on circulating brain EVs. Cerebrospinal fluid was extracted from the brain, and EVs were isolated and processed with label-free quantitative proteomic analyses via high-resolution tandem mass spectrometry. Interestingly, circulating EV-localized proteins were differentially expressed based on acute or chronic THC exposure in a sex-specific manner. Taken together, these findings reveal that THC acts in the brain to differentially modulate circulating EV signaling, thereby providing a novel understanding of how exogenous factors can regulate intercellular communication in the brain.

Project description:Cannabis has been used throughout history for medicinal and recreational purposes. The most notable cannabinoids derived from these plants are cannabidiol (CBD) and tetrahydrocannabinol (THC). Although well studied for their therapeutic effects, and highly debated concerning their recreational use, the underlying mechanisms of their biological effects are poorly defined. Here we used isobaric tag-based sample multiplexed proteome profiling to investigate protein abundance differences in the human neuroblastoma SH-SY5Y cell line treated with CBD and THC. We highlighted significantly regulated proteins by each treatment and performed pathway classification and associated protein-protein interaction analysis. Our data suggest that these treatments may result in mitochondrial dysfunction and induce endoplasmic reticulum stress. This dataset can be mined further to investigate the potential role of CBD and THC in various biological and disease contexts and thus provide a foundation for future studies.

Hippocampal differential expression underlying the neuroprotective effect of delta-9-tetrahydrocannabinol (THC) microdose on old mice

Project description:Delta-9-tetrahydrocannabinol (THC) is the primary psychoactive compound of the cannabis plant and an exogenous ligand of the endocannabinoid system. In previous studies, we demonstrated that a single microdose of THC (0.002mg/kg, 3–4 orders of magnitude lower than the standard dose for rodents) exerts distinct, long-term neuroprotection in model mice subjected to acute neurological insults. When administered to old, healthy mice, the THC microdose induced remarkable long-lasting (weeks) improvement in a wide range of cognitive functions, including significant morphological and biochemical brain alterations. To elucidate the mechanisms underlying these effects, we analyzed the gene expression of hippocampal samples from the model mice. Samples taken 5 days after THC treatment showed significant differential expression of genes associated with neurogenesis and brain development. In samples taken 5 weeks after treatment, the transcriptional signature was shifted to that of neuronal differentiation and survival. This study demonstrated the use of hippocampal transcriptome profiling in uncovering the molecular basis of the atypical, anti- aging effects of THC microdose treatment in old mice.
