Project description:As a response to insect attack, maize (Zea mays) has inducible defenses that involve large changes in gene expression and metabolism. Piercing/sucking insects such as corn leaf aphids (Rhopalosiphum maidis) cause direct damage by acquiring phloem nutrients as well as indirect damage through the transmission of plant viruses. To elucidate the metabolic processes and gene expression changes involved in maize responses to aphid attack, leaves of inbred line B73 were infested with R. maidis for two to 96 hours.

Proteomic analysis of Laodelphax striatellus in response to rice stripe virus infection

Project description:Persistent plant viruses multiply and circulate inside insect vectors following the route of midgut-hemolymph-salivary gland. Currently, how viruses interact with insect vectors after they are released into hemolymph is not entirely clear. In this study, we found that the hemolymph and fat body (HF) contained the highest RSV levels. Proteomic analysis on RSV-free and RSV-infected HF identified 156 differentially expressed proteins (DEPs), with the majority of them participating in metabolism, transportation, and detoxification.

Cuticular structure proteomics analyses of the pea aphid Acyrthosiphon pisum

Project description:Aphids are sap-feeding insects with piercing-sucking mouthparts. They are serious pests in agriculture and provoke significant losses, with a formidable capacity to transmit numerous plant viruses causing diseases in almost all important crops. The majority of viruses are acquired after feeding on an infected plant, retained on the aphid’s stylet and inoculated to healthy ones with a single probing puncture, promoting viral outbreaks. We have recently discovered the acrostyle, an organ located at the tip of aphid maxillary stylets, that harbors the receptors for Cauliflower Mosaic Virus (CaMV), and likely those for numerous other viruses. To our opinion, establishing an exhaustive inventory of the cuticular proteins composing the acrostyle, defining the peptide motifs accessible at the surface, identifying virus receptors among these proteins, will contribute to better understand virus/vector and plant/insect interactions at different scales including the molecular level. In this manuscript, we identify proteins or peptides at the surface of aphid stylets that might play a key role in plant virus binding. This may help to pave the way for future alternative “environmentally safe” strategies to control viral spread and aphid infestation in important crops worldwide.

Millet can be infected with several viruses if grown as a weed on crop fields

Project description:Millet is a dangerous weed in Hungary. Lack of seed dormancy helps it to spread easily and be present at maize, wheat and other crop fields. Our previous report revealed the possibility that millet can also play a role as a virus reservoir. In that study we detected the presence of several viruses in millet using DAS ELISA. Because serological methods can only detect the presence of the investigated particular pathogens, we suspected that other, previously unknown viruses can also be present in this weed. To investigate this theory, we randomly sampled two locations and collected millets showing stunting, chlorosis, and striped leaves and investigated the presence of viruses using small RNA HTS as a diagnostic method. Our result confirmed the widespread presence of wheat streak mosaic virus at both locations. Moreover, barley yellow striate mosaic virus and barley virus G were also identified, which have not been described from Hungary before. As these viruses can cause severe diseases on wheat, their presence on a weed mean a potential infection risk. Our study indicates that the presence of millets on the fields needs a special control in order to prevent emergence of new diseases at crop fields.

Plant and aphid vector manipulation by a persistent plant virus

Project description:Plant viruses rely on both host plant and vectors for a successful infection. This study investigated the global transcriptomic changes in Arabidopsis thaliana that were simultaneously exposed to both a plant virus (turnip yellows virus, polerovirus genus and Solemoviridae family) and its aphid vector (Myzus persicae). Some of these modifications in gene expression may promote in a timely manner viral transmission and dispersion.
