Project description:We employed a single-cell RNA sequencing approach using the 10X Genomics platform to study the diversity of telocyte populations in the mouse jejunum. Telocytes are a unique mesenchymal cell type expressing the transcription factor Foxl1 and forming a subepithelial network along the entire crypt-villus axis. This study characterizes the gene expression profiles and spatial distribution of telocytes along the crypt-villus axis of the mouse jejunum.

Terminal differentiation of villus-tip enterocytes is governed by distinct members of Tgfb superfamily

Project description:The protective and absorptive functions of the intestinal epithelium rely on differentiated enterocytes in the villi. The differentiation of enterocytes is orchestrated by sub-epithelial mesenchymal cells producing distinct ligands along the villus axis, in particular Bmps and Tgf. Here we show that individual Bmp ligands and Tgf drive distinct enterocytic programs specific to villus zonation. Bmp4 is expressed mainly from the centre to the upper part of the villus, and it activates preferentially genes connected to lipid uptake and metabolism. In contrast, Bmp2 is produced by villus-tip mesenchymal cells, and it influences the adhesive properties of villus-tip epithelial cells and the expression of antimicrobial peptides. Hence, Bmp2 promotes the terminal enterocytic differentiation at the villus-tip. Additionally, Tgf induces an epithelial gene expression programs similar to that triggered by Bmp2. The inhibition of Bmp receptor type I in vivo and using intestinal organoids lacking Smad4 revealed that Bmp2-driven villus-tip program is activated by a canonical Smad-dependent mechanism. Finally, we established an organoid cultivation system that enriches for villus-tip enterocytes and thereby better mimics the cellular composition of the intestinal epithelium. Altogether our data suggest that not only Bmp gradient, but also the activity of individual Bmp drives specific enterocytic programs.

Project description:We analyzed gene expression profiles in isolated mouse LGR5+ intestinal stem cells, lineage-specific enterocyte and secretory progenitors, and terminally differentiated villus enterocytes. Gene Ontology analyses of cell type-specific transcripts indicated their expected biological functions. We hybridized Affmetrix 430A 2.0 mouse microarrays with processed RNA isolated from purified Lgr5+ intestinal stem cells, secretory (Sec-Pro) and enterocyte (Ent-Pro) crypt progenitors, and mature villus enterocytes (ENT).

The developmental emergence and stem cell-based renewal of the intestinal telocyte niche

Project description:The intestinal telocytes that reside beneath the intestinal epithelium exert niche-supporting roles for intestinal stem cells (ISCs) and their progenies. They are heterogenous cells compartmentalized along the crypt-villus axis, but the lineage relationships and mechanisms governing the maintenance of this telocyte population remain poorly understood. Through single cell analysis of mouse small intestine development, here we identify a distinct subset of Foxl1+ mesenchymal cells marked by LIM Domain Only 3 (Lmo3), whose emergence at embryonic day E13.5-14.5 precedes the initiation of villus morphogenesis and the appearance of Lgr5+ ISCs. Later on, these villus-associated telocytes undergo proliferation and expansion, extending their presence to both villus and intervillus regions. After birth, the distribution of the Lmo3+ telocytes gradually becomes confined to the isthmus region, where they persist as long-lived cells and slowly give rise to the progenies that migrate either up- or down-wards and differentiate into villus- or crypt-associated telocytes, respectively. The Lmo3+ cells are responsive to intestinal damage and ablation of these cells compromises the epithelial repair process. Thus, the Lmo3+ cells residing in the isthmus region of the adult small intestine represent a population of telocyte stem cells (TeloSCs) that regulate telocyte niche homeostasis and regeneration. The single cell delineation of the ISC niche emergence and the discovery of TeloSCs open up new avenues for understanding the roles of the mesenchymal environment in intestinal development and diseases.

Single intestinal stem cells build crypt-villus structures in vitro without a cellular niche

Project description:The intestinal epithelium is the most rapidly self-renewing tissue in adult mammals. We have recently demonstrated the presence of approximately six cycling Lgr5+ stem cells at the bottoms of small intestinal crypts1. We have now established long-term culture conditions under which single crypts undergo multiple crypt fission events, whilst simultanously generating villus-like epithelial domains in which all differentiated cell types are present. Single sorted Lgr5+ stem cells can also initiate these crypt-villus organoids. Tracing experiments indicate that the Lgr5+ stem cell hierarchy is maintained in organoids. We conclude that intestinal crypt-villus units are self-organizing structures, which can be built from a single stem cell in the absence of a non-epithelial cellular niche.
