Project description:mRNA profiles of Candida albicans SC5314 treated with compound H55 and untreated Candida albicans SC5314 were generated by deep sequencing, in triplicate, using Illumina HiSeq X Ten. The sequence reads that passed quality filters were analyzed at the transcript isoform level. qRT–PCR validation was performed using SYBR Green assay. We find that some genes in ergosterol biosynthesis pathaway such as ERG3, ERG6 and ERG9 were upregulated, but HMG1, ERG2 and ERG12 were downregulated. Genes invovled in hyphal growth and adhesion were also accordingly regulated in H55 treated group.

Project description:mRNA profiles of Candida albicans wild-type (CAF2-1),cys4 mutant and CYS4 integrated strains were generated by deep sequencing, in triplicate, using Illumina HiSeq X Ten. The sequence reads that passed quality filters were analyzed at the transcript isoform level. qRT–PCR validation was performed using SYBR Green assay. We find that filament or biofilm-specific regulators such as UME6, TEC1 and BRG1 and adhesins such as hypha-specific genes ECE1 and ALS3 were downregulated and negative hyphae regulator NRG1 was upregulated in cys4 mutant. Genes invovled in cell wall synthesis and oxidative stress response were also accordingly regulated in cys4 mutant.

Project description:Candida albicans SC5314 cells were treated with 19ak for mRNA profiling analysis through deep sequencing, in triplicate, using Illumina HiSeq X Ten. The sequence reads that passed quality filters were analyzed at the transcript isoform level. qRT–PCR validation was performed using SYBR Green assay. We find that genes invovled in mitochondrial respiration such as NUO1, NUO2 and MCI4 and ribosomal biogenesis such as RPL5,RPS43,RPS12 were downregulated. Genes invovled in reduction-oxidation progress and iron and zinc ions uptake were also regulated in 19ak-treated C. albicans cells.

Project description:Illumina NovaSeq 6000 was used to deeply sequenced the mRNA expression of the AMB treated group, the AMB and Rimonabant combined treatment group and the vehicle-treated Candida albicans SC5314, and three repeated mRNA expressions were obtained. The analysis was performed at the isoform level by sequence reading through a quality filter. SYBR Green method was used for QRT-PCR validation. We found that after Amb treatment, the regulatory genes MDM12, PSD2, OPI3, INO2 and PMP3 related to lipid synthesis were up-regulated, and INO2 and MDM12 were significantly down-regulated after combined treatment.Some genes involved in lipid modification and oxidative phosphorylation also changed accordingly

Project description:Fungi have developed a variety of mechanisms to resist antifungals for survival. Uncovering their resistance mechanisms will aid in development of alternative therapeutics for fungal infections. In our study, a time-lapse microscopic imaging method was used to screen a molecule library and SP1 was identified as a short-time antifungal agent and exhibited biphasic killing pattern. Investigation of the underlying mechanism for the biphasic killing pattern and short-time activity of SP1 may expand the utility of this important class of spirobisnaphthalenes and uncover a new drug-resistant mechanism in fungi. mRNA profiles of SP1 treated Candida albicans SC5314, untreated Candida albicans SC5314 were generated by deep sequencing, in triplicate, using illumina NovaSeq 6000. The sequence reads that passed quality filters were analyzed at the transcript isoform level. qRT-PCR validation was performed using SYBR Green assay. The RNA-seq and qPCR results revealed an upregulation of heat shock response related genes including HSP90, SSA1 and osmotic and oxidative stress related genes including HOG1 and PBS2 and CAP1. Some common transporters encoding genes were also accordingly altered in SP1 treated group.
