Project description:The goal of this study was to compare the transcriptome of neurons from the locus coeruleus (LC) from wild type and mice that are conditional KO to argininosuccinatelyase (ASL) in the LC. Using an optimized data analysis, we were able to detect difference in the transcription of key enzymes in the synthesis of catecholamine, along with changes in the catecholamine synthesis pathway. Approximately 7% of the transcripts showed differential expression between the WT and ASL-LC cKO mice. The altered expression of the releant genes was confirmed with qRT–PCR. Our study represents the detailed analysis of the LC that revealed novel metabolic regulation on this region.

Project description:Preclinical work has long focused only on male animals, even though sexual divergence in both baseline behaviors and drug responses clearly impact treatment outcomes in patients. Psychiatric disorders are notably divergent, with males showing higher prevalence of ADHD and ASD, and females GAD and MDD. This divergence is reflected in quantitative differences in subclincal behaviors. The Noradrenergic neurotransmitter system is targeted by many psychiatric drugs, but is relatively uncharacterized at a molecular level. We developed a mouse to profile these neurons, defining their both a baseline transcriptome, including druggable receptors, and their molecular response to stimulation. We also discovered a remarkable sexual divergence in their gene expression, including functionally increased expression of the EP3 receptor in females – a difference that can be used to modulate stress-induced anxiety in a sex specific manner. These findings underscore the need to conduct preclinical studies in a manner balanced for sex, and suggest that baseline differences in noradrenergic neurons could underlay sexually divergent behaviors.

Single nuclei analyses of supraspinal neurons in the mouse brain

Project description:The mammalian brain contains numerous neurons distributed across forebrain, midbrain, and hindbrain that project axons to the lower spinal cord and work in concert to control movement and achieve homeostasis. Extensive work has mapped the anatomical location of supraspinal cell types and continues to establish specific physiological functions. The patterns of gene expression that typify and distinguish these disparate populations, however, are mostly unknown. Here we combined retrograde labeling of supraspinal cell nuclei with fluorescence activated nuclei sorting and single nuclei RNA sequencing analyses to transcriptionally profile neurons that project axons from the mouse brain to lumbar spinal cord. We identified fourteen transcriptionally distinct cell types and used a combination of established and newly identified marker genes to assign an anatomical location to each. To validate the putative marker genes, we visualized selected transcripts and confirmed selective expression within lumbar-projecting neurons in discrete supraspinal regions. Finally, we illustrate the potential utility of these data by examining the expression of transcription factors that distinguish different supraspinal cell types and by surveying the expression of receptors for growth and guidance cues that may be present in the spinal cord. Collectively these data establish transcriptional differences between anatomically defined supraspinal populations, identify a new set of marker genes of use in future experiments, and provide insight into potential differences in cellular and physiological activity across the supraspinal connectome.

Sex differences in transcriptome profiles in hypothalamic and extrahypothalamic nuclei of rats impacted by adolescent stress

Project description:In order to generate data worthy of translatability to known sex differences in human stress-induced maladies, adolescent male and female rats underwent chronic restraint stress (stress) or not (nonstress, control) from postnatal day (PD)32-PD44. Specific brain nuclei (PVN, VMH, ARC, BLA, CEA, CA1, CA3, DG, and OFC) were analyzed for genome-wide transcriptome using RNA sequencing (RNA-Seq). Global transcriptome profiles were generated, and genes differentially expressed (DE) due to stress were identified for each nuclear subregion listed. Canonical pathway analyses identified DE genes that are likely crucial for causing well-known stress-induced sex differences.

RNA-seq analysis of differentiated noradrenergic neurons (1C11NE) and microglia (BV2) following exposure to acrylamide (ACR)

Project description:Neurotoxicity of acrylamide (ACR), which is an environmental electrophilic chemical, has been reported in occupational human cases and experimental animal models. Our previous study using murine model demonstrated that exposure to ACR induced dose-dependent degeneration of noradrenergic axons, which has been reported to be extensively related with various cognitive disorders and neurodegenerative diseases. To investigate the mechanism of ACR-induced degeneration of noradrenergic neurons and the role of microglia, in this study, differentiated noradrenergic neuron model (1C11NE), as well as microglia cell line (BV2) were used. BV2 microglia were exposed to PBS or ACR (1mM) for 24 hrs. 1C11NE neurons were exposed to ACR or to conditioned medium (CM) from ACR-treated BV2 microglial cells (ACR-CM). Transcriptomic RNA-seq were carried out for 1C11NE neurons and BV2 microglia.

Proteoform Detection in 17000 Single Nuclei from Differentiating Skin Keratinocytes

Project description:We analyzed over 17000 single nuclei derived from the differentiating human epidermal keratinocytes using single-nucleus Proteoform imaging Mass Spectrometry (snPiMS). This study enabled highly sensitive detection of intact proteoforms of core histones and variants along with other nuclear proteins from the single nucleus. Integration of snPiMS data enabled distinct proteoform-defined clusters, with a continuum from proliferative progenitors through active cell-cycle phases to the terminal differentiation. This study establishes snPiMS as a powerful platform for cell-type and state characterization at the intact proteoform level, complementing single-nucleus transcriptomic frameworks and advancing the frontier of single-cell proteomics (SCP).
