Project description:ES cells grown in growth factor depleted matrigel could recapitulate the morphogenesis of the epiblast from E4.5 to E5.5. This process is characterized with the establishment of epithelial polarity. We used Next-generation sequencing (NGS) technique to analyze the transcriptional change of ES cells grown in 3D culture conditions in the presence of DMSO, 2i, CH or FGF2/Activin. The comparison of transcriptom between different conditions helps us to understand the molecular mechanism underlying morphogenesis of the epiblast. We found naïve pluripotency factors are kept in a high level in 2i and CH treatment in comparison with DMSO and FGF2/Activin treatment. We identified 5 naïve pluripotency factors which may have potential regulatory function in epithelial polarity. To further analyze these 5 pluripotency factors, we found Esrrb is the core transcriptional factor countering epithelial polarity. We also used Next-generation sequencing (NGS) technique to analyze the transcriptional change of Esrrb delta/delta (+4OHT) and control Esrrb fl/fl (-4OHT) ES cells in the presence of CH. Esrrb delta/delta (+4OHT) cell grown in 3D become polarized at 24 hours, whereas the control Esrrb fl/fl (-4OHT) ES cells are still non-polarized. By analyzing the RNA-Seq data, we found the enrichment of MAPK, Apoptosis, Focal adhesion and Tight junction KEGG pathways in Esrrb delta/delta compared to control Esrrb fl/fl cells. This result suggests the essential function of Esrrb in regulating epithelial polarity.

Project description:Studies in the mouse demonstrate the importance of fibroblast growth factor (FGF) and extra-cellular receptor tyrosine kinase (ERK) in specification of embryo-fated epiblast and yolk-sac-fated hypoblast cells from uncommitted inner cell mass (ICM) cells prior to implantation. Molecular mechanisms regulating specification of early lineages in human development are comparatively unclear. Here we show that exogenous FGF stimulation leads to expanded hypoblast molecular marker expression, at the expense of the epiblast. Conversely, we show that specifically inhibiting ERK activity leads to expansion of epiblast cells functionally capable of giving rise to naive human pluripotent stem cells. Single-cell transcriptomic analysis indicates that these epiblast cells downregulate FGF signalling and maintain molecular markers of the epiblast. Our functional study demonstrates the molecular mechanisms governing ICM specification in human development, whereby segregation of the epiblast and hypoblast lineages occurs during maturation of the mammalian embryo in an ERK signal-dependent manner.

Project description:The impact of WNT signalling activity on the acquisition and restriction of lineage propensity of germ layer progenitors and the gene network activity for cell fate decision during the development of the embryonic head was modelled in the epiblast stem cells derived and maintained under different signalling conditions. Our findings showed that the modulation of WNT activity is critical for the specification of the anterior (head) tissue progenitors in the multipotent early epiblast and the repression of WNT activity enhances the ectoderm lineage potency of the epiblast cells and poises the activation of endogenous WNT activity that drives neurogenesis during head morphogenesis.

Project description:Naive and primed human pluripotent stem cells (hPSC) provide valuable models to study cellular and molecular developmental processes. The lack of detailed information about cell-surface protein expression in these two pluripotent cell types prevents an understanding of how the cells communicate and interact with their microenvironments. Here, we used plasma membrane profiling to directly measure cell-surface protein expression in naive and primed hPSC. This unbiased approach quantified over 1700 plasma membrane proteins including those involved in cell adhesion, signalling and cell interactions. Notably, multiple cytokine receptors upstream of JAK-STAT signalling were more abundant in naive hPSC. In addition, functional experiments showed that FOLR1 and SUSD2 proteins are highly expressed at the cell surface in naive hPSC but are not required to establish human naive pluripotency. This study provides a comprehensive stem cell proteomic resource that uncovers differences in signalling pathway activity and has identified new markers to define human pluripotent states.
