Project description:Abstract: Our intake of ultra-processed foods has dramatically increased over the past few decades in line with the prevalence of obesity and diabetes, key risk factors for microvascular diseases such as chronic kidney disease (CKD). The extent to which long-term intake of highly processed food influences CKD outcome is unclear. Here, we show in rodent models that a highly processed diet drives intestinal barrier permeability and an increased risk of CKD. Inhibition of the advanced glycation pathway, which generates Maillard reaction products within foods upon thermal processing, reversed kidney injury. Consequently, a highly processed diet leads to innate immune complement activation and local kidney inflammation via the potent proinflammatory effector molecule complement 5a (C5a). C5a receptor inhibition ameliorated albuminuria. In a mouse model of diabetes, a high resistant starch fiber diet led to a redistribution of the gut commensal consortium, prevented impaired gut barrier function and decreased the severity of kidney injury via suppression of complement. These results provide mechanistic insight into the role of highly processed foods on inflammation and chronic disease risk. One Sentence Summary: Ultra-processed diets promote chronic kidney disease

Project description:Excessive ultra-processed foods (UPFs) consumption has been reported to increase the risk of inflammatory bowel disease (IBD). However, the specific mechanisms remain unclarified. As an important ingredient of UPFs, 7-ketositosterol (KS) is mainly synthesized from high-temperature heating oils. We find that KS intake is higher in IBD patients and related to disease activity. KS exacerbats colitis in a gut microbiota-dependent manner in mice, altering the gut microbiota composition, increasing the abundance of potential pathogenic bacteria, especially Staphylococcus_lentus (SL). Morevoer, SL aggravates DSS-induced colitis. Mechanically, KS up-regulates the expression of PDZ and LIM Domain 3 (PDLIM3). SL-derived lysin motif peptidoglycan-binding domain-containing protein (LPDP) interacts with PDLIM3, and activates p38MAPK/NF-κB signaling pathway. Furthermore, tubuloside B, selected by high-throughput screening, blocks the interaction of PDLIM3 and LPDP, and ameliorates SL-aggravated colitis. Our study reveals that KS exposure promotes colitis via gut microbiota and PDLIM3 interaction, providing evidence of IBD pathogenesis and potential therapeutic strategy for IBD treatment.

Project description:Acute respiratory distress syndrome (ARDS) is a leading cause of intensive care unit (ICU) admissions and is associated with high short- and long-term mortality, particularly among elderly patients. Aging is a major risk factor for ARDS development and worsened outcomes, yet the mechanisms underlying age-related susceptibility remain poorly defined. Pulmonary microvascular endothelial cell (PMVEC) barrier dysfunction plays a central role in ARDS pathophysiology, where disruption of cell-cell junctions leads to increased vascular permeability and alveolar edema. While aging has been associated with increased microvascular permeability in several vascular beds, the specific effects of age on pulmonary endothelial cell junctions remain largely unexplored. Here, we investigated the impact of aging on PMVEC barrier function and junctional integrity under basal conditions. Using the XperT permeability assay, we demonstrate that PMVECs from aged mice exhibit significantly increased paracellular leak and disrupted VE-cadherin localization compared to those from young mice. Western blot analyses further revealed age-associated reductions in the tight junction protein claudin-5 and the adherens junction protein γ-catenin, despite an increase in VE-cadherin levels. Proteomic profiling of PMVECs uncovered broad age-related differences in pathways related to mRNA processing, protein folding, cytoskeletal regulation, and inflammation. These findings suggest that aging impairs pulmonary endothelial barrier integrity through cell junction disruption and proteome remodeling, offering mechanistic insight into the heightened vulnerability of elderly individuals to ARDS. Targeting age-associated endothelial dysfunction may represent a promising therapeutic strategy to mitigate ARDS morbidity and mortality in aging populations

Project description:Food classification systems have been proposed to improve food quality criteria. Among these systems, "processing level" has been used as a criterion. NOVA classification, as the denotation "ultra-processed" food (UPF), has been widely used in different countries. However, even though some studies have pointed out some controversial aspects, no study has evaluated its comprehension by the population where it is used as reference. Therefore, this study explored the understanding of the term UPF for Brazilian consumers, where this denotation has been used in the last 8 years. A questionnaire was used, with questions referring to different aspects of self-assessment of knowledge about UPF. Altogether, 939 valid participants completed the questionnaire, and 81.9% of them declared to know the term UPF. For 78.2%, a better definition for UPF should be "foods that have gone through many processes in industry". Finally, it was concluded that the term UPF is still confusing for most Brazilians, indicating the risk of use and the urgent necessity to improve the classifications systems and consequently consumer understanding. Only when all parties interested in healthy food work together could this problem be solved.

Project description:BackgroundUltra-processed foods (UPF) are becoming extensively available in the food environments. UPF are industrial formulations that are designed to maximize palatability and consumption through a combination of calorie-dense ingredients and chemical additives. UPFs are also aggressively marketed, which may make them more attractive than unprocessed/minimally processed foods (UMPF). Since consumers' purchase decisions are guided by food-evoked emotions, we aimed to provide evidence that UPF visual cues trigger higher emotional responses and approach motivation than UMPF visual cues, with potential impacts on individuals' intention to consume the UPF over the UMPF.MethodsParticipants (n = 174; 144 women; mean age = 20.7 years; standard deviation = 4.35) performed two tasks. In the first task, 16 pictures of foods (8 UPF and 8 UMPF), and 74 pictures from other affective categories, were presented. After viewing each picture, the participants rated it along two basic dimensions of emotion through the Self-Assessment Manikin scale: pleasantness and arousal. In the second task, the participants viewed the same food pictures, and they rated their intention to consume the foods depicted in the pictures. Each picture was plotted in terms of its mean pleasantness and arousal ratings in a Cartesian plane, which resulted in an affective space.ResultsPictures of UPF and UMPF were positioned in the upper arm of the boomerang-shaped affective space that represents approach motivation. Pictures containing UPF triggered higher approach motivation and intention to consume than pictures containing UMPF. We also found a stronger association between emotional responses and intention to consume UPF relative to UMPF.ConclusionThese results shed new light on the role of ultra-processed foods evoked emotions that contribute to less healthy and sustainable food environments.

Project description:<p>The rising consumption of ultra-processed foods (UPFs), linked to multiple health risks, underscores the need to characterise the chemical profile of formulated and processed goods to improve their quality and safety. This study aims to develop untargeted approaches based on LC-HRMS and LC-HRMS/MS, coupled with feature-based molecular networking (FBMN), to explore, for the first time, the chemical reactivity within a well-characterised UPF-like food matrix (sponge cake). Three controlled baking conditions were applied to the formulated cake to induce thermal reactivity and generate diverse chemical profiles. Principal component analysis and heatmap clustering of untargeted LC-HRMS data from cake extracts were able to effectively discriminate samples based on the thermal process intensity. Approximately 75% of the detected features were indeed involved in the reactivity. FBMN revealed different groups of compounds (including precursors and advanced products) associated with Maillard and caramelisation reactions, and helped annotation of several reaction markers. This is the first application of FBMN on widely consumed processed foods such as baked products, opening new perspectives for generating high-throughput untargeted data to annotate reaction markers from complex food matrices.</p>
