Project description:Many genes involved in epithelial-to-mesenchymal transition (EMT) have been identified to date, but mechanisms contributing to the phenotypic diversity and those governing the coupling between the dynamics of epithelial genes and that of the mesenchymal genes are unclear. In this study, we employed single-cell transcriptome analysis to study the gene expression changes during dynamic E-to-M transition. Dose-dependent response to TGFbeta signal suggested that gene expression patterns between E and M states are rather continuous unlike binary switching between two discrete populations. E and M genes can be classified into several subclusters based on expression correlation and it turned out that some E and M genes are mutually exclusive but others are not. Our data suggests that complex and continuous changes in molecular phenotypes occur during EMT at a single-cell level.

Project description:Epithelial to Mesenchymal Transition (EMT) has been associated with cancer cell heterogeneity, plasticity and metastasis. It has been the subject of several modeling effort. This logical model of the EMT cellular network aims to assess microenvironmental signals controlling cancer-associated phenotypes amid the EMT continuum. Its outcomes relate to the qualitative degrees of cell adhesions by adherent junctions and focal adhesions, two features affected during EMT. Model attractors recover epithelial, mesenchymal and hybrid phenotypes, and simulations show that hybrid phenotypes may arise through independent molecular paths, involving stringent extrinsic signals. Of particular interest, model predictions and their experimental validations indicated that: 1) ECM stiffening is a prerequisite for cells overactivating FAK-SRC to upregulate SNAIL1 and acquire a mesenchymal phenotype, and 2) FAK-SRC inhibition of cell-cell contacts through the Receptor Protein Tyrosine Phosphates kappa leads to the acquisition of a full mesenchymal rather than a hybrid phenotype.

Thymidylate synthase drives the phenotypes of epithelial-to-mesenchymal transition in non-small cell lung cancer

Project description:Epithelial to mesenchymal transition (EMT) is a developmental process driving metastasis and chemoresistance. Thymidylate synthase (TS) is a proliferation enzyme and a chemotherapeutic drug target we previously correlated with EMT. Here we report a rather direct role of TS in determining EMT phenotypes in non-small cell lung cancer (NSCLC). TS knockdown and a promoter reporter system revealed the control of TS on EMT, and upstream regulators (HMGA2, HOXC6) and downstream effectors (AXL, SPARC, FOSL1) were identified. In vivo, TS knockdown suppressed lung colonization and metastasis formation in a proliferation-independent manner. Transcriptomic analyses showed a significant enrichment of EMT signature genes in NSCLC patients with high TS levels. These results establish the role of TS as a theranostic NSCLC marker mediating survival, chemo-resistance and EMT, and identifies a regulatory network that could be exploited to target EMT-driven NSCLC.

Membrane-bound TGF-beta on tumor-associated mesenchymal stem-like cells initiates epithelial-to-mesenchymal transition in human colorectal cancer

Project description:Growing evidence indicates that tumor-associated stroma plays a negative role in human colorectal cancer (CRC). Nature of specific stromal cell populations involved and mechanisms underlying their negative impact remain to be fully understood. In this study we describe the expansion from human primary CRCs of a mesenchymal cell population, referred to as tumor-associated stromal cells (TASCs), resembling bone marrow-derived mesenchymal stem cells (BM-MSCs) in morphology, phenotypes and differentiation potential. We found that, upon co-culture with tumor cells, TASCs acquire membrane-bound TGF-mbTGF-expression, a phenomenon mediated by v6 integrin. MbTGF-expression proved to be critical for triggering epithelial-to-mesenchymal transition (EMT) in tumor cells, eventually leading to enhanced dissemination of circulating tumor cells and increased metastasis formation, in an orthotopic mouse model. Our data identify CRC-associated mesenchymal stem-like cells as critical EMT initiators and suggest mbTGF- as potential novel therapeutic target.

Project description:Using the highly sensitive cricRNA array, we screened 13617 circRNAs abundant in the human liver cancer and Adjacent normal liver tissues, and the function of differentially expressed circRANs were analyzed by bioinformatics. The results revealed that cFUT8 controls epithelialâmesenchymal transition in liver cancer by cFUT8/miR-548c-3p/FUT8 axis. In this study, three cases of liver cancer samples were used to acquire the circRNA expression profiling, and qRT-PCR was employed to confirm the results of circRNA microarrays. The functions of the differentially expressed circRNA were analyzed by bioinformatics methods. Translational reprogramming during epithelial-mesenchymal transition is associated with fine-tuning of ribosome composition

Project description:Epithelial-mesenchymal transition (EMT) is driven by transcriptional and epigenetic reprogramming. However, it is now well established that the final step of gene expression, translation, is also key to the establishment of physiopathological phenotypes, and one of its regulators is the ribosomes themselves. Using genome-wide analyses, we report on how the translational landscape evolves and how the translational machinery rearranges itself during EMT. Ribosome profiling revealed that translation was as profoundly modulated as transcription during EMT. In addition, riboproteomics revealed an increased proportion of RPL36A-containing ribosomal particles in mesenchymal cells, indicating that ribosome composition is finely tuned during EMT. Finally, RPL36A, but not RPL19, is sufficient to induce the acquisition of mesenchymal features. Taken together, these data demonstrate that EMT is associated with both a profound translational reprogramming and changes in ribosome composition, and that a single change in ribosomal protein is sufficient to drive EMT.
