Project description:Communication between individuals via molecules, termed chemosignaling, is widespread among animal and plant species. However, we lack knowledge on the specific functions of the substances involved for most of such systems. The femoral gland is an organ secreting a waxy substance, which is involved in chemical communication in lizards. While the lipids and volatile substances secreted by the femoral glands have been investigated in several biochemical studies, the protein composition and functions of secretions are completely unknown. Applying a proteomic approach, we provide the first attempt to comprehensively characterize the protein composition of femoral gland secretions from the Galápagos marine iguana. Using samples from several organs, the marine iguana proteome was assembled by next generation sequencing and mass spectrometry, resulting in 7,513 proteins; 4305 proteins were present in the femoral gland, including keratins, small serum proteins and fatty acid-binding proteins. Surprisingly, no proteins with obvious roles in partner recognition or inter-species communication could be identified. However, we found several proteins with direct associations to the innate immune system, such as lysozyme C, antileukoproteinase (ALP), pulmonary surfactant protein (SFTPD) and galectin (LGALS1), suggesting the femoral glands function as an important barrier to infection. The identification of previously unrecognized effective anti-microbial peptides from the femoral gland show similar activities against E.coli and B. subtilis compared to oncocin. This proteomics dataset is a valuable resource for future functional protein analysis and demonstrates that femoral glands secretions covers also functions of the innate immune system.

Project description:Proteins are ubiquitous macromolecules displaying a vast repertoire of chemical and enzymatic functions making them suitable candidates for chemosignals used in intraspecific communication. Proteins are present in skin gland secretions of vertebrates but their identity, and especially, their functions, remain largely unknown. Many species of lizards possess femoral glands, i.e. epidermal organs primarily involved in the production and secretion of chemosignals playing a pivotal role in mate choice and intrasexual communication. The lipophilic fraction of femoral glands has been well studied in lizards. In contrast, proteins have been the focus of only a handful of investigations. Here, we study the identity, inter-individual expression patterns and functionality of proteins present in femoral glands of sand lizards (Lacerta agilis) by applying mass-spectrometry proteomics. Our results showed that the total number of proteins varied substantially among individuals. None of the identified femoral gland proteins could be directly linked to chemical communication in lizards, although this result hinges on protein annotation in databases in which squamate semiochemicals are poorly represented. In contrast to our expectations, proteins consistently expressed across individuals were related to immune system, antioxidant activity and lipid metabolism as the main functions, adding support to the hypothesis that proteins in reptilian epidermal glands have other functions besides chemical communication. Interestingly, we found that major histocompatibility complex class I (MHC) expression is enriched in femoral gland secretions. Previously, MHC was hypothesized to have been coopted to serve a semiochemical function in sand lizards, specifically in partner recognition. We speculate with the possibility that MHC proteins could be linked to semiochemical function in sand lizards.

Project description:Picocyanobacteria from the genus Synechococcus are ubiquitous in ocean waters. Their phylogenetic and genomic diversity suggests ecological niche differentiation, but the selective forces influencing this are not well defined. Marine picocyanobacteria are sensitive to Cu toxicity, so adaptations to this stress could represent a selective force within, and between, “species” also known as clades. We compared Cu stress responses in cultures and natural populations of marine Synechococcus from two co-occurring major mesotrophic clades (I and IV). Using custom microarrays and proteomics to characterize expression responses to Cu in the lab and field, we found evidence for a general stress regulon in marine Synechococcus. However, the two clades also exhibited distinct responses to copper. The Clade I representative induced expression of genomic island genes in cultures and Southern California Bight populations, while the Clade IV representative downregulated Fe-limitation proteins. Copper incubation experiments suggest that Clade IV populations may harbor stress-tolerant subgroups, and thus fitness tradeoffs may govern Cu-tolerant strain distributions. This work demonstrates that Synechococcus has distinct adaptive strategies to deal with Cu toxicity at both the clade and subclade level, implying that metal toxicity and stress response adaptations represent an important selective force for influencing diversity within marine Synechococcus populations.

Project description:Still in its infancy, the functions of lactylation remain elusive. To address this, we established a comprehensive workflow for lactylation studies that integrates the discovery of lactylation sites with proteomics, the expression of site-specifically lactylated proteins in living cells via genetic code expansion (GCE), and the evaluation of the resulting biological consequences. Specifically, we developed a wet-and-dry-lab combined proteomics strategy, and identified highly conserved lactylation at ALDOA-K147. Driven by its potential biological significance, we site-specifically expressed this lactylated ALDOA in mammalian cells and interrogated the biological changes. We discovered that it not only inhibited enzyme activity but also elicited gain-of-function effects——it dramatically reshaped the functionality of ALDOA by improving stability, enhancing nuclear translocation and affecting gene expression. Further, we demonstrated broad applicability of this workflow to study distinct histone lactylation sites. Together, we anticipate its wide uses in elucidating causative links between site-specific lactylation and target-centric or cell-wide changes.
