Project description:We report transcriptional response to ribociclib treatment in resistant and sensitive CAMA-1 cells using RNA sequencing. Acquired ribociclib resistant cells were generated by treating CAMA-1 cells over a period of 5 month. Cells were then treated with ribociclib or DMSO control and processed for RNA sequencing.

Project description:MCF7, T47D and CAMA-1 cell lines were treated with long-term (continuous) capivasertib to induce resistant cell-lines (T47D P1, MCF7 P1 and CAMA-1 P1). The naïve (or parental) cell lines had RNAseq sequencing at treatment with DMSO and treatment with capivasertib. Resistant cell lines had RNAseq sequencing at treatment with continuous capivasertib, removal of continuous capivasertib and treatment with DMSO and then re-treatment with capivasertib monotherapy. This gave 5 time points across the different 3 different cell lines.

Project description:Do illuminate the vulnerabilities of CDK4/6i-resistant T47D breast cancer cells, we created CDK4/6i-resistant T47D cell clones through a gradual dose escalation protocol using the selective CDK4/6 inhibitors Ribociclib and Palbociclib. We then performed gene expression analysis targeting 2549 genes using the HTG Oncology Biomarker Panel (OBP) via HTG EdgeSeq.

Project description:We investigated the mechanisms of acquired resistance to the CDK4 selective inhibitor atirmociclib in HR+ HER2- breast cancer in vivo, using the ZR751 xenograft model. Atirmociclib resistant tumors appeared after long-term (>50 day) treatment with atirmociclib. Tumor material was isolated from resistant (85-day treatment) as well as sensitive/vehicle treated (3-day) tumor bearing mice and subjected to RNA-seq analysis.
