Project description:Murine pancreatic cancer cells (HY15549 cells) established from genetically engineered mouse models (GEMM) of PDAC (p48-Cre+, KrasLSL-G12D/+, Trp53lox/+; KPC mice) were transfected with the GFP-LC3-RFP autophagy flux reporter from Mizushima lab, wherein reduction in the GFP/RFP ratio indicates increase in autophagy flux. Organoids derived from these cells were dissociated into single cells and sorted into autophagy-high (AThi) and autophagy-low (ATlo) populations according to GFP/RFP signal ratio. To validate this reporter, RNA was extracted from these two populations and expression of autophagy/lysosome related genes was examined.

KNSTRN knockdown impairs autophagy flux and inhibits bladder cancer progression through ROS- mediated lysosomal dysfunction

Project description:Bladder cancer (BLCA) is a prevalent cancer affecting the urinary tract, the mitochondria-localized Astrin-binding protein (KNSTRN) has been shown to be strongly associated with the development of several cancers. In addition, abnormal levels of autophagy have been shown to have a dramatic impact on tumor development. However, the potential mechanisms of BLCA autophagy regulation by KNSTRN remain poorly understood. In this research, we found that knockdown of KNSTRN inhibited the autophagic flux of BLCA. Mechanistically, knockdown of KNSTRN leads to reactive oxygen species (ROS)-dependent disruption of the lysosomal acidic environment and reduction of protease activity, hindering autophagosome-lysosome fusion. Clioquinol reactivates lysosomal activity by normalizing lysosomal pH, which in turn reinstates autophagic flux. Intracellular ROS production is also critical for this process, as ROS scavengers (NAC) reverse lysosomal dysfunction and reactivate autophagic flux. Additionally, the autophagy activator rapamycin (Rapa) effectively protected against KNSTRN knockdown-induced cell death in vitro and in vivo. Thus, we demonstrate that knockdown of KNSTRN leads to intracellular ROS accumulation and lysosomal dysfunction, which impairs autophagic flux and inhibits BLCA progression.

Autophagy is required for the development and functionality of lacrimal gland-like organoids

Project description:Lacrimal glands (LGs) serve as pivotal exocrine glands crucial for protecting the ocular surface. Dysfunction in LG cell composition or secretion is implicated in dry eye disease (DED). While autophagy plays a vital role in tissue. homeostasis in many organs, how it affects LG development and secretory function is not known. Here we have undertaken a genetic study by utilizing autophagy-deficient human embryonic stem cells and differentiating them into LG-like organoids. Autophagy-deficient LG-like organoids exhibited improper development and secretion, along with increased protein aggregation, proliferation, and cell death. These phenotypes were associated with an accumulation of PAX6, a transcription factor crucial for brain and eye development, which we identified as an autophagy substrate. Pharmacological interventions with nicotinamide mononucleotide and melatonin were able to rescue the cellular dysfunction in autophagy-deficient LG-like organoids. Together, our study highlights the role of autophagy in LG along with potential therapeutic interventions for DED.

Miz1 is required to maintain autophagic flux

Project description:Miz1 is a zinc finger protein that regulates expression of cell cycle inhibitors as part of a complex with Myc. Cell cycle-independent functions of Miz1 are poorly understood. Here, we use a Nestin-Cre transgene to delete an essential domain of Miz1 in the central nervous system (Miz1M-NM-^TPOZNes). Miz1M-NM-^TPOZNes mice display cerebellar neurodegeneration characterized by the progressive loss of Purkinje cells. Chromatin immunoprecipitation sequencing and biochemical analyses show that Miz1 activates transcription upon binding to a non-palindromic sequence present in core promoters. Target genes of Miz1 encode regulators of autophagy and proteins involved in vesicular transport that are required for autophagy. Miz1M-NM-^TPOZ neuronal progenitors and fibroblasts show reduced autophagic flux. Consistently, polyubiquitinated proteins and p62/Sqtm1 accumulate in the cerebella of Miz1M-NM-^TPOZNes mice, characteristic features of defective autophagy. Our data suggest that Miz1 may link cell growth and ribosome biogenesis to the transcriptional regulation of vesicular transport and autophagy. Regulation of autophagy by Rab27B in colorectal cancer

Project description:Autophagy is a cellular recycling process that can promote tumor growth, anti-tumor immune response, and resistance to therapy in colorectal cancer (CRC). Here, we show that small GTPase Rab27B, a known regulator of vesicle trafficking and extracellular vesicle secretion, to control the autophagy process in CRC. Depletion of Rab27B in CRC cells showed an abnormal accumulation of autophagy vesicles and increased autophagy markers, indicating a defect in autophagy flux. Imaging analysis indicated that autophagy flux is blocked at the autophagosome/lysosome fusion step when Rab27B is lost. Loss of Rab27B significantly impacted CRC cell growth in both in vitro 3D growth and in vivo tumorigenesis studies. Together, these results demonstrate a new role of Rab27B in the autophagy trafficking process in CRC and identify Rab27B as a potential therapeutic target for CRC.
