RNA-seq study of human long-term and short-term hematopoietic stem cells from umblical cord blood with lentiviral overexpression of S1PR3

Project description:To investigate the molecular and biological pathways altered by S1PR3OE in human hematopoietic stem cells (HSC), we performed RNA-sequencing (RNA-seq) of LT- and ST-HSC 3 days after transduction with control or S1PR3 overexpression (OE) lentiviral vectors. LT-HSC and ST-HSC from 3 pool of CB lin- were FACS-purified, cells were prestimulated for 4 hours and transduced with lentiviral vectors. At day 3, 2000-5300 BFP+ cells were FACS-purified for RNA isolation with a PicoPure kit. We were able to isolate only 1600-1800 BFP+ cells from LT-HSC control samples as opposed to 4000-5400 BFP+ cells from S1PR3OE samples. Thus, we pooled all control BFP+ LT-HSC cells into one sample for RNA-seq analysis. BFP- LT-HSC from control vector transduction were purified from CB1 as an additional LT-HSC control. Nextera libraries generated from 10 ng RNA from 5 LT-HSC samples (2 controls, 3 S1PR3OE) and 6 ST-HSC samples (3 controls, 3 S1PR3OE) were subjected to 125 bp, paired-end RNA-sequencing on the Illumina HiSeq 2500 with an average of 50 million reads/sample at the Center for Applied Genomics, Sick Kids Hospital.

S1PR3 modulates the hyper-proliferation of keratinocytes in psoriasis by activating STAT3 in PKA/Src dependent manner

Project description:Background: Abnormal lipid metabolism influences the occurrence and development of psoriasis. Moreover, persistent activation of STAT3 signaling pathway in epidemies contributes to psoriasis. However, the molecular mechanism of lipid metabolism and STAT3 activation is unclear. Methods: Imiquimod (IMQ)-induced mouse psoriasis-like model was established to access the potential treatment of FTY720. The function of SIP receptors 3 (S1PR3) was determined by S1PR3 siRNA in HaCaTs and S1pr3 knock out in mice. RNA-seq in HaCaTs was undertaken to explore the downstream target of S1PR3. ChIP assay was applied to explore the transcription factor on S1PR3 promoter. Findings: The upregulation of S1PR3 in the lesion from psoriasis patients and IMQ-induced mice was observed. Moreover, the pan-S1PR modulator FTY720 ameliorated the lesion in IMQ-induced mice, which was mediated by internalizing S1PR3. S1PR3 knocking downing both in vivo and in vitro significantly suppressed the hyper-proliferation of keratinocytes in a P-STAT3-dependent manner. We found that upregulated-S1PR3 expression maintained STAT3 activation in psoriasis through both short and long term. Namely, S1PR3 enhanced early STAT3 activation via promoting the binding of P-Src to gp130, and long STAT3 activation by reducing Src. Ultimately, Upregulated-S1PR3 was due to STAT3 binding to S1PR3 promoter in psoriatic skin.

S1PR3–G12-biased agonist ALESIA targets cancer metabolism and promotes glucose starvation [HeLa RNA-seq]

Project description:Cancer tissues possess less glucose than surrounding normal tissue, because cancer cells predominantly produce energy by glycolysis than oxidative phosphorylation even in the presence of an adequate oxygen supply (Warburg effect). We found a novel compound ALESIA which causes apoptosis of malignant cells at a low glucose condition from an original phenotypic screening. Identified target molecule of ALESIA was Sphingosine-1-phosphate receptor 3 (S1PR3). Being different from the natural ligand, ALESIA selectively stimulated S1PR3-G12 coupling to S1PR3 without suppression by β-arrestin recruitment and continuously promoted NO synthesis. To reduce the enhanced oxidative stress, NAPDH production through pentose-phosphate pathway and resulting consumption of glucose were promoted in ALESIA-treated cells. ALESIA therefore accelerated glucose starvation of cancer cells and preferentially killed them both in vitro and in vivo.

Project description:RNA-seq of uncultured CD112high and CD112low long-term(LT) human hematopoietic stem cells (HSC) and cultured and lentiviral transduced (CTRL, INKA1-OE) LT- and short-term HSC from umbilical cord blood We performed RNA-sequencing to elucidate the biological pathways (1) altered by INKA1 overexpression in LT-HSC and ST-HSC that may contribute to the induced transient restraint in generating proliferative hematopoietic output in vivo and in vitro; and (2) that underlie the differential in vitro priming and regeneration phenotypes of CD112high and CD112low LT-HSC. Therefore, we prospectively isolated indicated subpopulations from 3 independent human umbilical cord blood pools and subjected them either to RNA-sequencing directly or following culture, lentiviral transduction and purification of the transduced cell fractions. We show that distinct subsets of human LT-HSC respond differently to regeneration-mediated stress and that INKA1 governs these distinct stemness states where CD112low LT-HSC are marked by an alternative state of quiescence with high INKA1 preserving self-renewal and regenerative capacity upon successive rounds of regeneration-mediated stress.
