Oleic Acid Promotes Lipid Accumulation in Bovine Adipocytes: The Role of PPARα Activity

Project description:Background: Excessive lipid mobilization due to negative energy balance (NEB) in periparturient dairy cows increases their risk for diseases and decreases their productivity. Improving energy intake during the immediate postpartum limits NEB. One way to enhance energy density of the diet is through fatty acid (FA) supplementation. In periparturient cows, oleic acid (OA) limits lipolysis and improves adipose tissue (AT) insulin sensitivity. However, the mechanisms behind OA effect are still unknown. Our objective was to characterize the effect of OA, alone and in combination with palmitic acid (PA), on lipid accumulation, and identify a mechanism of action in bovine adipocytes. Results: Compared with CON, FA treatments had no effect on preadipocyte differentiation (P = 0.65). The treatments 300PA, 200OA, 300OA, 60-40, 50-50, and 40-60 increased lipid accumulation compared with CON (P < 0.0001). At d 7, 300PA and 300OA increased PPARα protein expression compared with CON (P = 0.04). PPARα inhibition decreased triglyceride levels of OA and 60-40 (P<0.05) but not for PA (P=0.26).

Project description:Compared to ordinary rapeseed, high-oleic acid rapeseed has higher levels of monounsaturated fatty acids and lower levels of saturated fatty acid and polyunsaturated fatty acids, and thus is of high nutritional and health value. In addition, high-oleic acid rapeseed oil imparts cardiovascular protective effects. Based on these properties, high-oleic acid oil crops have been extensively investigated and cultivated. In this study, we employed a microarray analysis with high oleic acid line and low oleic acid line from the developing seeds (27 days after flowering) of Brassica napus.

Transcriptome analysis of Bovine granulosa cells of preovulatory follicles at 30, 60, 90 and 120 days postpartum

Project description:Cows in Negative Energy Balance (NEB) may preferentially divert nutrients away from reproduction, thereby experiencing a period of anovulatory anestrus, delayed ovulation of large follicles and a condition of impaired fertility. To better understand the changes occurring in these large follicles as a function of time post-partum granulosa cells of preovulatory follicles have been collected at different times: 30, 60, 90 and 120 days after calving . An analysis of the transcriptome was performed using a global bovine oligo-array microarray to map the differences in genes expression and cellular functions that occur in the follicular microenvironment during the progressive recovery from NEB condition in dairy cow Four time points experiment: 30, 60, 90 and 120 days. Granulosa cells from the 30, 60 and 90 days compare to the 120 days (reference). Biological replicates: 3 from each time point. One replicate per array.

Dietary oleic acid drives obesogenic adipogenesis via modulation of LXRasignaling

Project description:Dietary fat composition has changed substantially as the obesity epidemic has spread. Adipocyte hyperplasia is a major mechanism of fat mass expansion in obesity, but how dietary fats contribute to obesogenic adipocyte hyperplasia is unknown. Here we employed various dietary strategies, including an unbiased dietary screen, to determine the effect of dietary fat composition on adipocyte hyperplasia in mice. We identified oleic acid (OA) as the only dietary fatty acid that induces adipogenesis and obesogenic hyperplasia at physiologic levels. OA induced differentiation via AKT2 signaling, a hallmark of obesogenic hyperplasia. We also demonstrate that reduction of LXR signaling in APCs is necessary for OA-stimulated adipogenesis. Furthermore, blocking LXRaphosphorylation at S196 results in APC hyperproliferation upon HFD feeding, suggesting that LXRaphosphorylation inhibits HFD-stimulated proliferation. As OA is associated with numerous health effects, our findings that OA is a unique physiologic metabolic regulator underscores the importance of understanding the mechanisms by which OA effects metabolic health.

Protective role of oleic acid in angiotensin II-induced cardiac remodeling

Project description:To gain mechanistic insights into the protective effects of oleic acid (OA) on angiotensin II(AngII)-induced cardiac remodeling, a whole transcriptomic analysis of heart were performed using RNA-seq.Comparing with heart of normal mice, a total of 1468 genes (987 upregulated and 481 down regulated) were differentially expressed (FDR<0.05) in heart of AngII-induced mice. Comparing with heart of AngII-induced mice, a total of 785 genes (162 upregulated and 623 down regulated) were differentially expressed (FDR<0.05) in heart of OA treated mice. Importantly, among the 987 upregulated genes in heart of AngII-induced mice,388 genes were significantly reversed in heart of OA treated mice.
