Transcriptome Profile Analysis Reveals the Regulation Mechanism of Stamen Abortion in Handeliodendron bodinieri（Levl.）Rehd

Project description:To elucidate the molecular mechanism of stamen abortion underlying sex differentiation from transcriptomic aspects in Handeliodendron bodinieri, we use abnormal stamens before and after abortion in female flowers and normal stamen in male flowers at the same stage as control material. Key genes regulating stamen abortion was identified by using RNA-seq technique. The results show that it was abortion science stage 6 during anther development, at the same time, HbGPAT, HbAMS, HbLAP5, HbLAP3 and HbTES were down-regulated , and HbML5 was up-regulated. Therefore, these information will provide theoretical foundation for the conservation, breeding, scientific research and application of Handeliodendron bodinieri.

Transcriptome Profile Analysis Reveals the Regulation Mechanism of Pistil Abortion in Handeliodendron bodinieri

Project description:To elucidate the molecular mechanism of pistils abortion underlying sex differentiation from transcriptomic aspects in Handeliodendron bodinieri, we use abnormal pistils before and after abortion in male flowers and normal pistils in female flowers at the same stage as control material. Key genes regulating pistil abortion was identified by using RNA-seq technique. The results show that it was abortion from the two-nucleate embryo sac to four-nucleate embryo sac at stage one (the diameters of the flower buds were 1.3–1.6 mm), at the same time, 12 genes related to ovule development, such as INO, BEL1, and CRC, 24 genes related to plant hormones, such as AHP6, ARF, ERF, and GID1, and 12 genes related the ROS, such as POD, SOD, CAT and RBOH might be involved in the regulation of pistil abortion. Therefore, this study will provide a theoretical foundation for the conservation, breeding, and application of H. bodinieri in the future.

SAM-synthase specific effects on metabolism and mitochondria underlie resistance to heat stress in C. elegans

Project description:S-adenosylmethionine (SAM), produced by SAM synthases, is critical for various cellular regulatory pathways and the synthesis of diverse metabolites. Studies have often equated the effects of knocking down one synthase with broader SAM-dependent outcomes such as histone methylation or phosphatidylcholine (PC) production. However, many organisms express multiple synthase genes including humans and Caenorhabditis elegans. Evidence in C. elegans, which possesses an expanded family of SAM synthases, suggest that the enzymatic source of SAM impacts its function. For instance, loss of sams-1 leads to enhanced heat shock survival and increased lifespan, whereas reducing sams-4 adversely affects heat stress survival. Here, we reveal that loss of sams-1 exerts age-dependent effects on nuclear-encoded mitochondrial gene expression, mitochondrial metabolites, and mitophagy. Notably, we find that that SAMS-1 exerts synthase-specific effects on PC production. We propose a mechanistic framework wherein the reduced SAM from SAMS-1 acts through PC to impact mitochondrial fission and mitophagy, thereby enhancing survival during heat stress. This study highlights multifaceted roles of SAM across metabolic pathways and synthase-specific SAM functions.

A distinct immature low-density neutrophil population characterizes acute generalized pustular psoriasis [PBMC]

Project description:Generalized pustular psoriasis (GPP) is a severe disease featured by neutrophilic pustules and enhanced IL-36 inflammatory pathway in skin. Recently, a transcriptomic analysis of PBMCs and neutrophils in MPO-deficient GPP patients has been performed in stable disease state. However, transcriptomic profiling of PBMCs during acute flare of GPP is unclear. Here, we reported a predominant neutrophil signature in GPP PBMC and a marked increase in the CD66+CD16+ low-density neutrophils (LDNs) within the PBMC fraction of acute GPP patients. Transcriptomic and functional analysis of LDNs revealed a hypoinflammatory phenotype yet enhanced release of neutrophil granule proteases, implicating that LDNs might contribute to the IL-36-mediated inflammation in GPP patients.
