Project description:Retinoic acid (RA) is a central developmental signal whose perturbation results in teratogenic outcomes. RA is tightly regulated during early embryonic development to achieve fine-tuning of the instructive and permissive signals regulating multiple developmental processes including head formation. We aim to identify the key mechanisms underlying the homeostatic regulation and tuning of the retinoic acid signaling levels, the disruption of which in the head lead to wide ranging developmental malformations including microcephaly and other craniofacial defects. We performed transient physiological RA signaling disturbances during Xenopus laevis embryogenesis followed by kinetic gene expression analysis of the recovery at multiple time points.

Project description:Chelonus formosanus is an important parasitic natural enemy of Lepidopteran insects and has great value in the biological control of insect pests. To screen the reference genes in the real-time fluorescence quantitative PCR (qRT-PCR) assay of Chelonus formosanus, six candidate reference genes were selected based on transcriptome data, including GAPDH, ACT, TU, TF, HSP, and RP. The expression stability of these six internal reference genes was determined by qRT-PCR in different tissues (head, chest, abdomen) and at different temperatures (11℃and 41℃), as well as with two insecticides (dinotefuran and chlorantraniliprole).

Dicer and Hsp104 function in a negative feedback loop to confer epigenetic robustness [RNA_correlations]

Project description:Epigenetic mechanisms can be influenced by environmental cues and thus evoke phenotypic variation. This plasticity can be advantageous for adaption, but also detrimental if not under tight control. Although having attracted considerable interest, it remains largely unknown if and how environmental cues such as temperature trigger epigenetic alterations. Using fission yeast, we demonstrate that environmentally induced discontinuous phenotypic variation is buffered by a negative feedback loop that involves the RNase Dicer and the protein disaggregase Hsp104. In the absence of Hsp104, Dicer accumulates in cytoplasmic inclusions and heterochromatin becomes unstable at elevated temperatures, an epigenetic state that is inherited for many generations after the heat stress. Dicer instead averts the toxic aggregation of a prionogenic protein. Our results highlight the importance of feedback regulation in building epigenetic memory and uncover Hsp104 and Dicer as homeostatic controllers that buffer environmentally induced stochastic epigenetic variation and toxic aggregation of prionogenic proteins. Technical comparison of qRT-PCR experiments with the GLYCOv2 custom array

Project description:Gene expression patterns were surveyed using RNA from 4 different murine sources. Four classes of RNA were analyzed: undifferentiated embryonic stem cells, differentiated ES cells, kidney and liver tissues. Classes were prepared in triplicate for a total of 12 samples. All samples were hybridized to the custom designed CFG GLYCOv2 glycogene array. Analysis results were used for comparison with parallel experiments using qRT-PCR.
