Project description:Here we have compared adult wildtype (N2) C. elegans gene expression when grown on different bacterial environments/fod sources in an effort to model naturally occuring nematode-bacteria interactions at the Konza Prairie. We hypothesize that human-induced changes to natural environments, such as the addition of nitrogen fertalizer, have effects on the bacterial community in soils and this drives downstream changes in the structure on soil bacterial-feeding nematode community structure. Here we have used transcriptional profiling to identify candidate genes involved in the interaction of nematodes and bacteria in nature.

Project description:<p>Tobacco fermentation is a crucial process for improving tobacco quality by reducing undesirable substances. In this study, an enzyme–microbe combined fermentation strategy was established and evaluated. Cytobacillus oceanisediminis C4 combined with amylase pretreatment was applied in the fermentation of tobacco powder (TP) to improve the quality of tobacco, yielding an enhanced sensory score of 84.50, compared with 82.00 in the untreated TP. Compared with the control, the reducing sugar content increased significantly, whereas starch and protein contents decreased markedly after combined fermentation. Furthermore, total aromatic components of TP increased by 34.77%, along with noticeable changes in TP surface structure. Bacterial community structure analysis revealed a noteworthy shift at the genus level, with the relative abundance of Bacillus and Pseudomonas escalating from 0.30% and 1.15% to 74.67% and 4.40%, respectively. Additionally, fungal community structure analysis revealed that the relative abundance of Monascus surged from 1.69% to 87.59%. Metabolomic analysis demonstrated that fermentation reprogrammed the metabolic profile toward flavor-active and aroma-precursor compounds, characterized by increased levels of amino acids and phenolic acids and decreased levels of lipids and flavonoids. These coordinated changes contributed to enhanced aroma, reduced irritation, and overall improvement in tobacco quality. Overall, this study established an enzyme–microbe combined fermentation process, demonstrated the effectiveness of the process in improving Heat-not-Burn tobacco products, and provided multi-omics insights into the enzyme–microbe combined fermentation process.</p>

Project description:Tobacco (Nicotiana tabacum L.) is an important cash crop, and the size of its leaves significantly influences both yield and quality. However, the upper part of tobacco leaves, due to its dense tissue structure, often faces issues such as narrow and thick leaves during the production of roasted cigarettes. These problems have a severe impact on the yield and quality of the upper leaf. Although the mechanism of leaf size regulation in Arabidopsis thaliana has been extensively studied, it remains unclear for tobacco. Therefore, this research aimed to investigate the role of the NtAN3 gene in regulating tobacco leaf size by utilizing the NC82 variety. The researchers created both an overexpression mutant (G27) and a silencing mutant (M21) of the NtAN3 gene and examined their impact on leaf size using cell morphology observation and transcriptome analysis. These research findings offer valuable insights for molecular breeding aimed at improving tobacco yield and enhancing the availability of upper leaves.

Project description:<p>In this study, a strain isolated from the surface of flue-cured tobacco leaves, identified as <em>Bacillus velezensis</em> HJ-16, was applied in the solid-state fermentation of tobacco leaves. This strain, known for producing thermally stable enzymes, including amylase, cellulase, and protease, significantly improved the sensory qualities of tobacco, enhancing aromatic intensity, density, and softness, while reducing irritation. Whole-genome sequencing and functional annotation revealed that <em>B. velezensis</em> HJ-16 possesses a single circular chromosome containing genes associated with enzyme production and metabolic activities, particularly in carbohydrate metabolism and amino acid metabolism. Untargeted metabolomics analysis identified significant changes in non-volatile metabolites induced by fermentation. These metabolites were enriched in pathways related to flavonoid biosynthesis, alkaloid biosynthesis, aromatic amino acid metabolism, lipid metabolism, and carbon metabolism. Metagenomic analysis showed that Bacillus became the dominant genus on the tobacco leaf surface following inoculation with <em>B. velezensis</em> HJ-16, altering the microbial community composition, reducing diversity and evenness, and enhancing microbial metabolic activity. These findings underscore the potential of <em>B. velezensis</em> HJ-16 as a biotechnological tool to improve tobacco leaf quality.</p>

Project description:We used two hybrid combinations and their 4 parents as materials. One week after topping the tobacco plant, samples were taken, including three tissues: root, stalk, and leaf. The RNA of these samples was extracted, and transcriptome sequencing analysis was carried out to explore the transcriptional profile differences between hybrids and their parents, and to analyze the gene differential expression between different tissues.
