Project description:Mice lacking Caveolin-1 (Cav1), a major protein of the lipid raft of plasma membrane, show deregulated cellular proliferation of mammary gland and an abnormal fetoplacental communication during pregnancy. This study leverages multi-omics approach to test the hypothesis that the absence of Cav1 elicits a coordinated crosstalk of genes among the mammary gland, placenta and fetal brain in pregnant mice. Integrative analysis of metabolomics and transcriptomics data of mammary glands showed that the loss of Cav1 significantly impacted specific metabolites and metabolic pathways in the pregnant mice. Next, gene expression changes of the deregulated metabolic pathways of the mammary gland were compared with the gene expression changes of the placenta and fetus. The analysis showed that genes associated with specific metabolic and signaling pathways changed in a coordinated manner in the placenta, mammary gland and fetal brain in Cav1-null mice. The cytokine signaling pathway emerged as a key player of the molecular crosstalk among these tissues. By interrogating the single-nuclei gene expression data of placenta and fetal brain previously generated from Cav1-null mice, the study further revealed that these metabolic and signaling genes were differentially regulated in specific cell types of the placenta and fetal brain. Though a causal effect of mammary gland on the placenta and/or fetal brain can’t be inferred from this study, the findings show that the mammary gland, placenta and fetal brain show a coordinated molecular crosstalk in response to the absence of Cav1 in mice.

Project description:Glands of the uterus are essential for the establishment of pregnancy in mice, sheep and likely humans. Forkhead box a2 (FOXA2) is expressed specifically in the glands of the uterus and a critical regulator of glandular epithelium (GE) differentiation, development and function. Mice with a conditional deletion of FOXA2 in the adult uterus, created using the lactotransferrin iCre (Ltf-iCre) model, have a morphologically normal uterus with glands, but lack a considerable number of FOXA2-dependent GE-expressed genes. Adult FOXA2 conditional knockout (cKO; Ltf-iCre:Foxa2 flox) mice are infertile due to defective embryo implantation arising from a lack of leukemia inhibitory factor (LIF), a critical implantation factor of uterine gland origin. Intraperitoneal injections of LIF can initiate embryo implantation in the uterus of adult FOXA2 cKO mice with pregnancies maintained to term. Here, we tested the hypothesis that FOXA2-regulated genes in the uterine glands impact development of the decidua, placenta and fetus. On gestational day (GD) 8.5, the anti-mesometrial and mesometrial decidua transcriptome was noticeably altered in FOXA2 cKO mice. Viable fetuses were reduced in FOXA2 cKO mice on GDs 12.5 and 17.5. Sex-dependent differences in fetal weight, placenta histoarchitecture, and the placenta and metrial gland transcriptome were observed between control and FOXA2 cKO mice. The transcriptome of the placenta with a female fetus was considerably more altered than the placenta with a male fetus in FOXA2 cKO dams. These studies reveal previously unrecognized sexually dimorphic effects of FOXA2 and uterine glands on fetoplacental development with potential impacts on offspring health into adulthood.

Project description:Placenta is an epitome of cell signalling, including the signalling between two semi-allogenic individuals, mother and fetus. We have studied the putative crosstalk by analyzing the expression of corresponding ligands and receptors on maternal fetal interface. This transcriptome represents one of the important cellular sources, namely the multinucleated fetal layer that is directly bathed in the maternal blood. It was collected by microdissection, as it is too large to be included in the single cell preparation. In combination with other compartments, whose transcriptomes are provided along with this one, we think this study provides a rich resource of gene expression profiles of term placenta.

Project description:Preeclampsia (PE), a hypertensive disorder in pregnancy, is linked to placental vascular remodelling, increasing risks of fetal growth restriction and long-term offspring health problems. The role of fetoplacental endothelial cell-derived extracellular vesicles (EVs) in PE remains underexplored. This study investigates whether EV composition in PE is altered, potentially contributing to impaired fetal development. Small EVs (sEVs) were isolated from primary fetoplacental endothelial cells (fpECs) of term (T), preterm (PT), and early-onset PE (EO-PE) pregnancies. sEVs were characterized using transmission electron microscopy, nanoparticle tracking analysis, and Western blotting, confirming spherical morphology, size (<200nm), and expression of canonical EV and endothelial markers. Proteomic profiling via nano-LC MS/MS and gene set enrichment analysis revealed consistent proteomes in fpEC-derived sEVs, but EO-PE-derived sEVs showed heterogeneity and functional alterations compared to T- and PT-derived sEVs. Notably, EO-PE sEVs were enriched in proteins linked to epithelial-to-mesenchymal transition and myogenesis, processes tied to tissue remodelling and vascular homeostasis, all hallmarks in PE. This signature may transmit signals of endothelial dysfunction to the fetus. In contrast, T-sEVs were enriched in cell cycle and DNA repair pathways. These findings underscore the role of fetoplacental-derived EVs in placental-fetal communication under pathophysiological conditions.

Project description:Gestational diabetes mellitus (GDM) leads to poor pregnancy outcomes and fetoplacental endothelial dysfunction; however, the underlying mechanisms are still unknown. This study aimed to investigate the effect of placenta-derived exosomal miRNAs on fetoplacental endothelial dysfunction in GDM, and to further explore the role of chemerin to this end. Placenta-derived exosomal miR-140-3p and miR-574-3p expression (next-generation sequencing, quantitative real-time PCR), its interactions with cell function (Cell Counting Kit-8, Transwell, tube formation assay), chemerin interactions (Western blotting), and placental inflammation (immunofluorescence staining, enzyme-linked immunosorbent assay) were investigated. Placenta-derived exosomal miR-140-3p and miR-574-3p were downregulated in GDM. Additionally, miR-140-3p and miR-574-3p inhibited the proliferation, migration, and tube formation ability of umbilical vein endothelial cells by targeting vascular endothelial growth factor. Interestingly, miR-140-3p and miR-574-3p expression levels were negatively correlated with chemerin, which induced placental inflammation through the recruitment of macrophage cells and release of IL-18 and IL-1β. These findings indicate that chemerin reduces placenta-derived exosomal miR-140-3p and miR-574-3p levels by inducing placental inflammation, thereby promoting the proliferation, migration, and tube formation of umbilical vein endothelial cells in GDM, providing a novel perspective on the underlying pathogenesis and therapeutic targets for GDM and its offspring complications.

Project description:Tissues throughout the human body exert finely-tuned specific functions even when there is considerable overlap in the cellular constituents. Even when the major cellular component of tissues is the same – for example in the case of smooth muscle-rich tissues - discrete phenotypes are evident. Yet, the molecular expression profiles underpinning such specialised functionality is often unclear. This presents a challenge for introducing new medicinal therapies for pathophysiologies: can one target what makes tissue functionality unique without producing unwanted effects on the features that are similar. Such a scenario can be considered for the human uteroplacental unit. Ideally, one would like to target complications of pregnancy like pre-eclampsia (hypertension and proteinuria) or spontaneous preterm birth (early activation of parturition) by targeting particular types of smooth muscle tissues (e.g. arteries versus myometrium) without adversely affecting others. This situation is further complicated by the need to consider maternal and fetoplacental circumstances. Three smooth muscle-rich tissues of importance here are myometrium, maternal uterine (myometrial) arteries and placental arteries. Indeed, each display subtly distinct phenotypes suggesting that distinct molecular signatures may underlie these differences. Hence, we aimed to compare expression profiles of these three tissues to identify the underpinning molecular signatures using multi-omics approaches.
