Project description:Background: Different diets result in significantly different phenotypes through metabolic and genomic reprogramming. Epigenetic marks, identified in humans and mouse models upon caloric restriction, high fat diet or the intake of specific bioactives, suggest that genomic reprogramming drives this reprogramming and mediates the effect of nutrition on health. Histone modifications encode the epigenetic signal that adapts genome functions to environmental conditions, including diets, by tuning the structure and properties of chromatin. So far, the effect of different diets on the genome-wide distribution of critical histone marks has not been determined Methods: We investigated by chromatin immunoprecipitation sequencing the distribution of the trimethylation of lysine 4 of histone H3 in the liver of mice fed for one year with five different diets including: chow containing corn powder as extra source of plant bioactives or specifically enriched with the cyanidin-3-O-glucoside, high fat enriched obesogenic and caloric restricted pro-longevity diets Conclusions: The comparison of the resulting histone mark profiles revealed functional food containing cyanidin determines broad effect.

Project description:Given that different diets could alter cow milk yield and composition, the effects of different feed formula on milk extracellular vesicle (EV) miRNAs were detected. Cow milk EVs contained various small RNAs, including miRNAs, snRNAs, tiRNAs, Cis-regulatory elements, and piRNAs. Two hundred and seventy-six known bos taurus miRNAs were identified by sequencing in bovine milk EVs. There were 13 immune-related miRNAs in the top 20 miRNAs in milk EVs. Nine differently expressed known miRNAs were detected in responding to different feed formulations. Cow milk EVs are abundant of small RNAs, especially miRNAs, which might be closely related to the development of maternal mammary gland and neonatal immune maturity.

Project description:Ballan wrasse (Labrus bergylta) is used as a cleaner fish in Norwegian aquaculture to control sea lice. Thus, knowledge on the digestive physiology and nutrient requirements, as well as the ontogeny of the immune system is important. In this study, two different diets were tested; diet 1 was used as control diet consisting of artemia and rotifers cultivated and enriched in the in-house facility at IMR, Austevoll, Bergen (Norway). Diet 2 consists in plankton which has successfully been used before in cod larvae generating more robust individuals. Sampling was done according to the standard length of the larvae rather than age (dph). Sampling point 0 (BW0) (4 mm), sampling point 2 (BW2) (4,5 mm), sampling point 3 (BW3) (5,7 to 6 mm), sampling 4 (BW4) (7 to 7,5 mm), sampling point 5 (BW5) (10 to 10,5 mm), sampling point 6 (BW6) (16 to 16,5 mm), and sampling point 7 (BW7) (25 to 30 mm). Ontogeny of lymphoid organs and mucosal associated lymphoid tissues in ballan wrasse and the effect of different diets were investigated.
