Project description:Purpose: To establish the steady state heterogeneity of murine Dendritic Epidermal T Cells (DETC) and to investigate the function of their constitutive interaction with neighbouring keratinocytes via Skint1 Methods: CD3+ cells were FACS purified from ear epidermal suspensions prepared from FVB mice administered isotype control or anti-Skint1 antibody 7d prior, then loaded into the 10x Genomics Chromium Platform and sequenced using Illumina HiSeq 4000. Conclusions: This analysis identifies four states of DETC in unperturbed epidermis, and reveals that DETC TCR sensing of keratinocytes via Skint1 potentially enables natural immune surveillance and maintains their thymic maturation signature in the periphery

Gene expression profiles in dendritic cells from Peyer's patches and epidermal of skin

Project description:Many studies have shown that the mucous membranes and skin are at the interface with different external environments and face the disparity of pathogenic effects, such as biological agents, chemical or physical environment. This difference may demand distinct immune responses. However, the mechanism to induce the distinct immune responses in mucous and skin is largely unknown. Dendritic cells of mucosa and skin are crucial in the initiation of immune responses, maintenance of self-tolerance and antigens presentation T cells. The different functions between mucosal and epidermal dendritic cells may play an important role in different immune responses. To compare the different gene expression of the mucosal DC and Langerhans cells (LC), we utilized microarrays to investigate different gene expression profiles in mucosal DC isolated from PPs (PDC) and epidermal LC from skin (ELC). 3548 genes were shown to be differentially expressed between PDC and ELC. According to genes annotations, 105 genes may be involved in immunity process. The genes involved in immune process were categorized to five groups related to DC function, including antigen presentation, antigen uptake, cytokines chemokines, and receptors, cell surface molecules and signal transduction. 11 of the highest expressed genes were selected as the candidate genes and reformed by real-time PCR. These 11 selected genes might be suitable candidates to further study the difference of gene expression between mucosal DC and epidermal LC and would be used for design for new vaccine. Beads and FACS sort were used to isolate dendritic cells from Peyer's patches and epidermal of skin from 80 four-week-old female BALB/c mice. Dendritic cells from Peyer's patches were pooled as one sample, and dendritic cells from epidermal of skin were pooled as another sample. RNA isolation, amplification, cDNA labelling, microarray hybridization and analyses were performed according to the Affymetrix manual book.

BCL2 inhibition reveals a dendritic cell-specific immune checkpoint that controls tumor immunosurveillance

Project description:We developed a screening platform in which immortal dendritic cell (DC) precursors can be genetically manipulated and then de-immortalized and differentiated into DCs for functional in vitro assays, as well for adoptive transfer into syngeneic tumor-bearing mice to explore their capacity to instate immunosurveillance. A genome-wide CRISPR screen led to the identification of BCL2 as an endogenous inhibitor of DC function. Knockout of BCL2 in DCs enhanced antigen presentation and the expression of activation markers in vitro, as well as the capacity of DCs to control tumor growth in vivo and to synergize with PD-1 blockade. These effects where phenocopied by the pharmacological BCL2 inhibitors venetoclax and navitoclax, both of which required DCs of the cDC1 subtype to efficiently reduce the growth of orthotopic lung cancers and fibrosarcomas. Thus, solid tumors failed to respond to venetoclax and navitoclax in mice constitutively devoid of cDC1 cells due to the knockout of Batf3, and this defect was reversed by the infusion of DCs. Moreover, depletion of cDC1 cells by systemic injection of cytochrome c reduced the therapeutic efficacy of BCL2 inhibitors alone or in combination with PD-1 blockade. In vivo treatment with venetoclax caused cDC1 cells to express several activation markers, both in mice and in patients. In conclusion, genetic and pharmacological BCL2 inhibition unveils a DC-specific immune checkpoint that restrains tumor immunosurveillance

Tumor-Pulsed Dendritic Cells Used as a Tumor Vaccine

Project description:This study is being conducted to determine the efficacy, side effects, and toxicity of an investigational vaccine that consists of tumor-pulsed dendritic cells administered with an immune stimulating drug called interleukin-2 (IL-2). Dendritic cells are immune cells that are obtained from a subject’s blood and are important in the body’s immune response to foreign substances. This study will examine the response of a subject’s immune system after receiving several vaccinations containing their own dendritic cells which have been exposed to dead fragments of their cancer cells in the laboratory. This may result in sensitizing a subject’s dendritic cells to their cancer cells so that their dendritic cells will react with other cells of the immune system and attack the cancer. It has been shown in the laboratory that dendritic cells exposed to cancer cell fragments can provide lymphocytes (a type of white blood cell) with signals they require in order to become fully activated and acquire the ability to kill cancer cells.
