Project description:Glycosylation is the most common post-translational modification of proteins, yet genes relevant to the synthesis of glycan structure and function are incompletely represented and poorly annotated on the commercially available arrays. To fill the need for expression analysis of such genes we employed the Affymetrix technology to develop a focused and highly annotated glycogene chip representing human and murine glycogenes, including glycosyltranferases, nucleotide sugar transporters, glycosidases, proteoglycans and glycan-binding proteins. In this report the array has been used to generate glycogene expression profiles of nine murine tissues. Global analysis with a hierarchical clustering algorithm, reveals that expression profiles in immune tissues (thymus, spleen, lymph node and bone marrow) are more closely related, relative to those of non-immune tissues (kidney, liver, brain and testes). Of the biosynthetic enzymes, those responsible for synthesis of the core regions of N-and O-linked oligosaccharides are ubiquitously expressed, while glycosyltransferase that elaborate terminal structures are expressed in a highly tissue-specific manner, accounting for tissue and ultimately cell type-specific glycosylation. Comparison of gene expression profiles with MALDI-TOF profiling of N-linked oligosaccharides suggested that the alpha-1-3 fucosyltransferase IX, Fut9, is the enzyme responsible for terminal fucosylation in kidney and brain, a finding validated by analysis of Fut9 knockout mice. Two families of glycan-binding proteins, C-type lectins and siglecs, are predominately expressed in the immune tissues, consistent with their emerging functions in both innate and acquired immunity. Project description:Background: Alterations in IgG glycosylation have been observed in many inflammatory activities and diseases progression, uncovering IgG glycome contributes to deciphering the molecular mechanism of human diseases. Gastric cancer (GC) associated IgG glycosylation was rarely investigated.Methods: We comprehensively characterized IgG N-glycome profile in GC by high-throughput orthogonal mass spectrometry assay, and analyzed the difference of glycosylation between controls and GC patients through multiple statistical tools. Bioinformatics analysis was performed to study the candidate glycogens and their biological functions involved in GC. Results: GC associates with decrease in galactosylation and sialylation of IgG, and increase in bisection and agalactosylation of IgG.Bioinformatics analysis verified the differential expression of significantly changed IgG glycan associated glycogens, indicating the crucial role of IgG glycosylation in the pathogenesis of GC.Conclusions: This study demonstrates the potential diagnostic merits of specific IgG glycans for early detection of GC, as well as revealing molecular mechanism in the development of this cancer.

Project description:The protein glycome of individual cell types in the brain is unexplored, despite the critical function of these modifications in development and disease. In aggregate, the most abundant asparagine (N-) linked glycans in the adult brain are high mannose structures, and specifically Man5GlcNAc2 (Man-5), which normally exits the ER for further processing in the Golgi. Mannose structures are uncommon in other organs and often overlooked or excluded in most studies. To understand cell-specific contributions to the unique brain N-glycome and its abundance of Man-5, we performed RNA-seq and MALDI-MS TOF protein N-glycomics at several timepoints during differentiation of multiple cell types. To this end, homogeneous cultures of glutamatergic neurons, GABAergic neurons, and brain-specific endothelial cells were generated from monoclonal human inducible pluripotent stem cells (hiPSCs) through cellular reprogramming. Small molecule induction of stably integrated synthetic transcription units driving morphogen expression generated differentiated cells with distinct patterns mirroring intact tissue. Comparing uninduced hiPSCs for each cell type revealed identical transcriptomic and glycomic profiles before differentiation, with low quantities of Man-5. In differentiated glutamatergic and GABAergic neurons, the most abundant N-glycans became Man-5 and its immediate precursor Man-6, despite the presence of transcripts encoding enzymes for their subsequent modification. Differentiation to brain-specific endothelial cells showed an opposite effect, with the N-glycome displaying an abundance of complex N-glycans and terminal modifications of the late secretory pathway. These results confirm that the restricted N-glycome profile of brain is programmed into neuronal differentiation, with regulation independent of the transcriptome and under tight evolutionary constraint.

Project description:Ovarian cancer (OC) is the one of the most common gynecological tumors with high morbidity and mortality. Alterations in serum N-glycosylation have been observed in many diseases, and specific N-glycans of serum could be used for disease diagnosis or prognosis. However, the association between serum N-glycome profiles and OC progression remains unclear. Herein, high-throughput mass spectrometry was applied to characterize serum N-glycome profile in individuals with healthy controls, benign neoplasm and different stages of OC patients. It was observed that high-mannosylation, neutral bisection and agalactosylation of serum glycoproteins were uniformly increased in the initiation and development of OC, but sialylation was substantially changed on an opposite trend. Additionally, agalactosylation, high-mannosylation and sialylation gained at least moderately accurate merit for diagnosis of both OC and benign neoplasm. Notably, serum N-glycome profile could accurately discriminate OC patients from benign cohorts, with a comparable or even higher diagnostic score compared with CA125 and HE4. Furthermore, bioinformatics verified the differential expression of glycogens in OC progression. Significantly, strong correlations were found between CA 125 and high-mannosylation, galactosylation and sialylation. These findings demonstrated the great potential of serum N-glycome for OC diagnosis and precancerous lesion prediction, paving a new way for OC screening and monitoring.
