Project description:We developed transgenic mice which overexpress the human LH-R transcript in the female reproductive tract, under the control of the oviduct-specific glycoprotein mouse promoter (mogp-1). The uteri of young TG mice showed signs of increased proliferation and trans-differentiation in the endometrial layer. A transcriptomic analysis,further confirmed by immunohistochemistry, showed that up regulated genes are mainly involved in cell cycle control, while down regulated genes are related to the immune system and the metabolism of xenobiotics. Comparison of mammary glands of 16 wk old wild type and LH-overexpressing mice

Project description:LH-overexpressing mice display elevated levels of mammogenic hormones and consequently develop spontaneous mammary tumors with a median latency of 41 weeks. Tumors occur in the absence of p53 mutations and in the presence of a seemingly functional p53 signaling pathway. At 16 weeks of age, no histological evidence of tumors is observed in these transgenic mammary glands; however, mammary cells do demonstrate centrosome amplification at this age suggesting that early tumorigenic processes may have occured. Thus comparison of the transcriptome of the mammary glands from 16 week old LH-overexpressing mice to that of wild type mice will provide insight into early events occuring in p53-independent hormone-induced tumorigenesis. Effect of LH replacement on luteal transcriptome following CET-induced luteolysis

Project description:A large body of evidence suggests that the development and maintenance of corpus luteum (CL) in primates requires the action of LH. Earlier, using CET-induced luteolysis model, we demonstrated changes in luteal transcriptome suggesting nuclear actions of LH in the primate CL. To further demonstrate the role of LH in maintenance of primate CL, replacement studies were carried out and it was observed that administration of a single injection of rhLH was sufficient to restore the progesterone to pre-CET treatment levels and prevent the CET-induced luteolysis. To elucidate the molecular mechanisms underlying the rescue of CL function, we used LH-replacement model to study immediate early changes in gene expression at a global level (Affymetrix oligonucleotide microarray) following LH replacement in CET-treated monkeys and to evaluate if the changes in gene expression mediated by LH-withdrawal can be reversed by LH replacement. Results demonstrated up-and down-regulation of various genes following LH replacement and suggested that LH-withdrawal induced changes in gene expression are reversible at least for some genes. Transcriptional profiling of the luteinizing hormone surge in bovine granulosa cells (GC) during the pre-ovulation period comparing 2 h pre-LH GC, 6 h post-LH GC and 22 h post-LH GC

Project description:The LH surge induces panoply of events that are essential for ovulation and corpus luteum formation. The transcriptional responses to the LH surge of pre-ovulatory granulosa cells are complex and still poorly understood. In the present study, a genome wide bovine oligo array was used to determine how the gene expression profiles of granulosa cells are modulated by the LH surge. Granulosa cells from three different statuses were used (1) 2 h before the induction of the LH surge, (2) 6 h and (3) 22 h after the LH surge to assess the short and long term effects of this hormone on follicle differentiation. The results obtained were a list of differentially expressed transcripts for each granulosa cell group. To provide a comprehensive understanding of the processes at play, biological annotations were used to reveal the different functions of transcripts, confirming that the LH surge acts in a temporal manner. The pre-LH group is involved in typical tasks such as cell division, development and proliferation, while the short response of the LH surge included features such as response to stimulus, vascularisation and lipid synthesis, which are indicative of cells preparing for ovulation. The late response of granulosa cells revealed terms associated with protein localization and intra-cellular transport corresponding to the future secretion task that will be required for the transformation of granulosa cells into corpus luteum. Overall, results described in this study provide new insights into the different transcriptional steps that granulosa cells go through during ovulation and before luteinization. Three biological granulosa cells samples: 2 h pre-LH vs. 6 h post-LH vs. 22 h post-LH. Biological replicates: 3 with a technical dye-swap replicates (Dy 547 and Dy 647) for each biological replicate. Hybridizations were performed in a loop design for a total a 9 hybridizations.
