Project description:Spodoptera frugiperda is the world’s major agricultural pests, and has the distinctive features of high fecundity, strong migratory capacity and high resistance to most insecticides. At present, the control of S. frugiperda in China relies mainly on the spraying of chemical insecticides. MicroRNAs (miRNAs) are a class of small, single-stranded, non-coding RNAs, and play crucial regulatory roles in various physiological processes, including the insecticide resistance in insects. However, little is known about the regulatory roles of miRNAs on the resistance of S. frugiperda to insecticides. In the present research, the miRNAs that were differentially expressed after cyantraniliprole, spinetoram, emamectin benzoate and tetraniliprole treatment were analyzed by RNA-Seq. A total of 504 miRNAs were systematically identified from S. frugiperda, and 24, 22, 31 and 30 miRNAs were differentially expressed after treatments of cyantraniliprole, spinetoram, emamectin benzoate and tetraniliprole. GO and KEGG enrichment analyses were used to predict the function of differentially expressed miRNAs’ target genes. Importantly, ten miRNAs were significantly differentially expressed among the treatments of three insecticides. MiR-278-5p, miR-13b-3p, miR-10485-5p and miR-10483-5p were significantly down-regulated among the treatments of three insecticides by RT-qPCR. Furthermore, overexpression of miR-278-5p, miR-13b-3p, miR-10485-5p and miR-10483-5p significantly increased the mortality of S. frugiperda to cyantraniliprole and emamectin benzoate. The mortality was significantly increased with spinetoram treatment after overexpression of miR-13b-3p, miR-10485-5p and miR-10483-5p. These results suggest that miRNAs, which are differentially expressed in response to insecticides, may play a key regulatory role in the insecticide resistance in S. frugiperda.

Project description:miRNAs play important roles in various biological processes through post-transcriptional regulation of gene expression. We previously identified 203 mature miRNAs in Diamondback moth, Plutella xylostella. This species has developed extremely high levels of resistance to chlorantraniliprole and other class of insecticides in the field. In this study, we examined the miRNA profile of P. xylostella in response to chlorantraniliprole exposure. The smRNA-seq data analyses showed that insecticide treatment caused significant changes in the abundance of some miRNAs. Increasing exposure time (6h to 24h) and insecticide concentration (0.01 to 0.1 ppm) induced more dysregulated miRNAs in DBM larvae.

Project description:In the field, plants usually have to face combined effects of abiotic and biotic stresses. In our study, two spring wheat cultivars Septima and Quintus were subjected to three water regimes (70, 50, 40 % soil water capacity SWC), aphid (Metopolophium dirhodum) infestation or the combination of both stresses, i.e., drought (50, 40% SWC) and aphids. The study has a 2x3x2 factorial design with three biological replicates. Our previous study (Saska et al. 2022) investigated plant morphophysiological characteristics and aphid life table. In the present study, the results of proteomic analysis using 2D-DIGE followed by MALDI-TOF/TOF protein identification are presented. Drought, but also aphid infestation led to alterations in 113 protein spots including proteins assigned to a variety of biological processes ranging from signalling via energy metabolism, redox regulation, stress and defense responses to secondary metabolism indicating a long-term adaptation to adverse conditions. The absence of specific proteins involved in plant response to herbivorous insects indicates a loss of resistance to aphids in modern wheat cultivars during the breeding process. Septima revealed enhanced tolerance with respect to Quintus as indicated by higher values of morphophysiological characteristics (above-ground biomass, leaf length, osmotic potential per full water saturation) and rel. abundance of proteins involved in mitochondrial respiration and ATP biosynthesis.

Project description:Modifications of metabolic pathways are important in insecticide resistance evolution. Mutations leading to changes in expression levels or substrate specificities of cytochrome P450 (P450), glutathione-S-transferase (GST) and esterase genes have been linked to many cases of resistance with the responsible enzyme being shown to utilize the insecticide as a substrate. Many studies show that the substrates of enzymes are capable of inducing the expression of those enzymes. We investigated if this was the case for insecticides and the enzymes responsible for their metabolism. The induction responses for P450s, GSTs and esterases to six different insecticides were investigated using a custom designed microarray in Drosophila melanogaster. Even though these gene families can all contribute to insecticide resistance, their induction responses by insecticides is minimal. The insecticides spinosad, diazinon, nitenpyram, lufenuron and dicyclanil did not induce any P450, GST or esterase gene expression. DDT was the only insecticide tested capable of eliciting an induction response, but only low levels for one GST and one P450. These results are in contrast to the induction responses observed for the natural plant compound caffeine and the barbituate drug phenobarbital, both of which induced a number of P450 and GST genes to high. Our results show that although insects evolve metabolic resistance to insecticides, induction does not usually have a role in survival after insecticide application, and induction studies cannot be used to predict which genes are capable of metabolizing insecticides. This has implications for managing the evolution of metabolic insecticide resistance in natural insect populations. Keywords: induction response after treatment by various compounds

Project description:Emerging and neglected pathogens pose challenges as their biology is frequently poorly understood, and genetic tools often do not exist to manipulate them. Organism agnostic sequencing technologies offer a promising approach to understand the molecular processes underlying these diseases. Here we apply dual RNA-seq to Orientia tsutsugamushi (Ot), the obligate intracellular causative agent of the vector-borne human disease scrub typhus. Half the Ot genome is composed of repetitive DNA, and there is minimal collinearity in gene order between strains. Integrating RNA-seq, comparative genomics, proteomics, and machine learning, we investigated the transcriptional architecture of Ot, including operon structure and non-coding RNAs, and found evidence for wide-spread post-transcriptional antisense regulation. We compared the host response to two clinical isolates and identified distinct immune response networks that are up-regulated in response to each strain, leading to predictions of relative virulence which were confirmed in a mouse infection model. Thus, dual RNA-seq can reveal the biology and host-pathogen interactions of a poorly characterized and genetically intractable organism such as Ot.
