Project description:Mosaic loss of chromosome Y (mLOY) is the most frequent chromosome alternation in the blood cells of aging adult male and is closely associated with AML, so we designed experiments to explore the mechanism. Bulk RNA sequencing was performed with AML1-ETO;sgSsty2/sgScramble;Trp53-/- Bone Marrow in vivo and sgSsty2/sgSsty1/sgScramble;Trp53-/- c-kit+ HSPCs in vitro. And our study demonstrates that mLOY is a functional driver for clonal hematopoiesis and leukemogenesis. KDM5D, a gene mediated H3K4 demethylation and located on both human and mouse chromosome Y, we wondered whether KDM5D could be a critical gene responsible for the pathology of mLOY. So bulk RNA sequencing was performed with sgKdm5d/sgScramble;Trp53-/- Bone marrow in vivo and Trp53-/- c-kit+ HSPCs in vitro to explore the mechanism of Kdm5d in mLOY.

Project description:Mosaic loss of chromosome Y (mLOY) in blood cells is one of the most frequent chromosome alterations in adult males. It is strongly associated with clonal hematopoiesis, hematopoietic malignancies, and other hematopoietic and nonhematopoietic diseases. However, whether there is a causal relationship between mLOY and human diseases is unknown. Here, we generated mLOY in murine hematopoietic stem and progenitor cells (HSPCs) with CRISPR/Cas9 genome editing. We found that mLOY led to dramatically increased DNA damage in HSPCs. Interestingly, HSPCs with mLOY displayed significantly enhanced reconstitution capacity and gave rise to clonal hematopoiesis in vivo. mLOY, which is associated with AML1-ETO translocation and p53 defects in patients with acute myeloid leukemia (AML), promoted AML in mice. Mechanistically, loss of KDM5D, a chromosome Y-specific histone 3 lysine 4 demethylase in both humans and mice, partially recapitulated mLOY in DNA damage and leukemogenesis. Thus, our study validates mLOY as a functional driver for clonal hematopoiesis and leukemogenesis.

Here, we developed a technique that combines mosaic mutagenesis with color labeling of HSPCs to study how acquired mutations affect clonal fitness in a native environment. Mutations in CH-associated genes, like asxl1, promoted clonal dominance. Single-cell transcriptional analysis revealed that mutations stimulated expression of proinflammatory genes in mature myeloid cells and anti-inflammatory genes in progenitor cells of the mutant clone. Biallelic loss of one such immunomodulator, nr4a1, abrogated the ability of asxl1-mutant clones to establish clonal dominance. These results support a model where clonal fitness of mutant clones is driven by enhanced resistance to inflammatory signals from their mutant mature cell progeny.

Project description:Clonal hematopoiesis plays a critical role in the initiation and development of hematologic malignant diseases. FOXM1, a well-known transcription factor, is often downregulated in CD34+ cells from patients with del(5q) Myelodysplastic Syndrome (MDS). Here we show that Foxm1 haploinsufficiency disturbs normal hematopoiesis and confers a competitive repopulation advantage to hematopoietic stem cells (HSCs) for a short period, but disrupts the long-term self-renewal capacity of HSCs, recapitulating the phenotypes of abnormal HSCs in MDS patients. Moreover, heterozygous inactivation of Foxm1 leads to an increase in DNA damage in hematopoietic stem/progenitor cells (HSPCs). Foxm1 haploinsufficiency induces an MDS-like disease in a mouse model with LPS-induced chronic inflammation, and accelerates AML-ETO9a-mediated leukemogenesis. We also identified PARP1, an important enzyme responding to various kinds of DNA lesions, as a previously unrecognized target of Foxm1. Foxm1 haploinsufficiency inhibits DNA damage response (DDR) in HSCs by downregulating PARP1 expression in HSPCs. Given that PARP1 inhibitors increase the risk of MDS and AML in clinical therapy for solid tumors, our results suggest that downregulation of the Foxm1-PARP1 molecular axis plays a pivotal role in the pathogenesis of MDS by promoting clonal hematopoiesis and reducing genome stability.

Project description:Clonal hematopoiesis of indeterminate potential is prevalent in elderly individuals and associated with increased risks of all-cause mortality and cardiovascular disease. However, mouse models to study the dynamics of clonal hematopoiesis and its consequences on the cardiovascular system under homeostatic conditions are lacking. We used a model of clonal hematopoiesis using adoptive transfer of unfractionated ten-eleven translocation 2-mutant (Tet2-mutant) bone marrow cells into nonirradiated mice. Consistent with age-related clonal hematopoiesis observed in humans, these mice displayed a progressive expansion of Tet2-deficient cells in multiple hematopoietic stem and progenitor cell fractions and blood cell lineages. The expansion of the Tet2-mutant fraction was also observed in bone marrow-derived CCR+ myeloid cell populations within the heart, but there was a negligible impact on the yolk sac-derived CCR2- cardiac resident macrophage population. Transcriptome profiling revealed an enhanced inflammatory signature in the donor-derived macrophages isolated from the heart. Mice receiving Tet2-deficient bone marrow cells spontaneously developed age-related cardiac dysfunction characterized by greater hypertrophy and fibrosis. Altogether, we show that Tet2- mediated hematopoiesis contributes to cardiac dysfunction in a nonconditioned setting that faithfully models the human clonal hematopoiesis in unperturbed bone marrow. Our data support clinical findings that clonal hematopoiesis per se may contribute to diminished health span.

Project description:Coronavirus disease 2019 (COVID-19) is associated with significant morbidity and mortality, albeit with considerable heterogeneity among affected individuals. Emerging evidence points towards an important role of preexisting host factors, such as a deregulated inflammatory response at the time of infection. Here, we demonstrate the negative impact of clonal hematopoiesis, a prevalent clonal disorder of ageing individuals, on COVID-19-related cytokine release severity and mortality. In this study we perform use the Illumina MethylationEPIC array to quantify methylation levels in PBMCs from COVID19 patients and patients with clonal hematopoiesis.
