Project description:Light provides critical information for the behavior and development of basically all organisms. Filamentous fungi sense blue light through a unique transcription factor complex that activates its targets in a light-dependent manner. In Trichoderma atroviride, the Blr-1 and Blr-2 proteins constitute this complex, which triggers the light-dependent formation of asexual reproduction structures (conidia). Using transcriptomics, we identified the genes regulated by the photoreceptors Blr-1, Blr-2, Cry-1, Cry-DASH, and Envoy. The evaluation of the transcriptional response to blue light of the Δblr-1 and Δblr-2 mutants showed that the BLR proteins have common targets, which indicates that these proteins act as a complex in gene regulation. Nevertheless, the severe alteration in the transcriptional profile of the Δblr-2 strain indicates that Blr-2 provides target specificity to the protein complex, while Blr-1 central role is as a transcriptional activator. The Δenv-1 mutant phenotypic characterization showed that Envoy functions as a growth and conidiation checkpoint, preventing excessive light responses. Similarly, we observed that Cry-1 and Cry-DASH contribute to the typical light-induced conidiation response. In the env-1 mutant, we observed, at the transcriptomic level, a general induction of DNA metabolic processes and strong repression of central metabolism. An analysis of the expression level of DNA repair genes showed that they increase their expression in the absence of Envoy. Photoreactivation experiments showed that Δenv-1 had increased DNA repair capacity, which is consistent with the transcriptomic data. Our results indicate that light perception in T. atroviride is far more complex than originally thought.

We generated an ENVOY photoreceptor mutant and performed RNA-seq analyses in the mutants of this gene and in those of the BLR-1, CRY-1 and CRY-DASH photoreceptors in response to a pulse of low intensity blue light. Like in other filamentous fungi BLR-1 appears to play a central role in the regulation of blue-light responses. Phenotypic characterization of the Δenv-1 mutant showed that ENVOY functions as a growth and conidiation checkpoint, preventing exacerbated light responses. Similarly, we observed that CRY-1 and CRY-DASH contribute to the typical light-induced conidiation response. In the Δenv-1 mutant, we observed, at the transcriptomic level, a general induction of DNA metabolic processes and strong repression of central metabolism. An analysis of the expression level of DNA repair genes showed that they increase their expression in the absence of env-1. Consistently, photoreactivation experiments showed that Δenv-1 had increased DNA repair capacity. Project description:Fungi as animals and plants conserves the ability to respond to stressful environmental conditions as biotic an abiotic stress. A correct injury response in Eukaryotes including fungi is carried out thought the activation of signaling pathways Ca2+ related dependent of DAMPs/MAPMPs recognizing for the activation of the innate immune system. During herbivory, plants, in addition to activating pathways related to injury, activates specific responses to combat its predator. Using a transcriptional approach, here, we reported the capacity of the filamentous fungus T. atroviride to activate or not a specific response to injury and predation to different predators also, we provide evidence of how fungivory by Drosophila melanogaster larvae inhibits the reported injury response for T. atroviride, although this predation negatively affects the hyphal regeneration thought the larval salivary compounds. Also, the activation of a putative alternate injury reproduction pathway is proposed.

Secretome analysis of Arabidopsis thaliana and Trichoderma atroviride reveals a diverse and dynamic repertoire of extracellular enzymes that may play a role during symbiotic interaction

Project description:The establishment of plant-mutualistic fungi interaction requires a bidirectional molecular crosstalk. The study of their secretomes help to understand a beneficial relationship. Here, a gel-free shotgun proteomic approach was used to identify secreted proteins from semi hydroponic culture systems with Arabidopsis (Arabidopsis thaliana) and T. atroviride alone or in co-culture over a time course (24, 48 and 96h). A total of 126 proteins of Arabidopsis and 1027 proteins of T. atroviride were identified, 118 and 780 of which were differentially modulated in response to their interaction, respectively. Bioinformatic analysis revealed that the secretomes of both organisms were enriched in proteins with a predicted enzymatic function. Among these, increased abundance of putative glycosidases, aspartic endopeptidases and dehydrogenases, together with decreased abundance of amidases, protein-serine/threonine kinases and hydro-lyases was observed in T. atroviride secretome in response to the plant, whereas increased abundances of peroxidases, cysteine endopeptidases and enzymes related to catabolism of secondary metabolites, together with decreased abundance of pathogenesis-related proteins and protease inhibitors was observed in the plant secretome in response to the fungus. Finally, the plant alanine-glyoxylate transaminase GGAT1, increased in abundance, is required for the systemic disease resistance against the phytopathogen Botrytis cinerea but not for plant growth stimulation by T. atroviride
